The patterns of adaptive radiations in ancient lakes provide valuable clues to mechanisms of speciation and adaptation. In contrast to vertebrate radiations, for instance in fishes or finches, invertebrate species flocks have been largely neglected. While the increase in molecular data narrows this gap, the anatomical basis for interpreting these data against the background of evolutionary hypotheses is still widely lacking. Here we evaluate anatomical findings in the live‐bearing pachychilid freshwater gastropod Tylomelania, which has radiated extensively in ancient lakes in the Indonesian island, Sulawesi; we have aimed at reconciling these data with recently obtained molecular phylogenetic evidence. Discovered more than a century ago, the speciose and phenotypically diverse species flock with 34 currently described taxa was only occasionally cited as an example of adaptive radiation in ancient lakes, while anatomical data were entirely lacking. Our study of anatomical characters reveals very low qualitative variation at the species level. Thus, contrary to earlier views we suggest the existence of a single monophyletic lineage endemic to this island. The most conspicuous feature of Tylomelania is its uterine brooding strategy, i.e. retaining eggs and embryos in the pallial oviduct. This is unique among South‐East Asian pachychilids. Within the uterine brood pouch the offspring is surrounded by considerable amounts of nutritive material produced by a very large albumin gland, and the embryos are produced continuously. The shelled juveniles of some species are the largest known so far in viviparous gastropods, measuring almost 2 cm in length when hatching. This combination of reproductive features in Tylomelania, characterized by a high amount of maternal investment, is considered to be ovoviviparous, rendering its brooding strategy unique also among other gastropods. In addition, our data reject a previously assumed close relationship to other South‐East Asian pachychilids and instead suggest the North Australian Pseudopotamis as sister group to Tylomelania. These findings have significant consequences for the phylogenetic interpretation of morphological characters of Tylomelania in an evolutionary and biogeographical context, leading to the hypothesis that the common ancestor of both genera originated somewhere on the northern Australian continental margin. © 2005 The Linnean Society of London, Biological Journal of the Linnean Society, 2005, 85 , 513–542.  相似文献

Whole cell DNA from Leishmania tropica has 2 peaks when banded by CsCl equilibrium density centrifugation. The main band has a buoyant density of 1.721 and the satellite band a buoyant density of 1.705, with Clostridium perfringens DNA (ρ= 1.6915) used as a reference. The satellite band has been identified as the kinetoplast DNA by purifying DNA from isolated kinetoplasts. L. tropica has the highest G + C content of both nuclear and kinetoplastic DNA thus far reported for trypanosomatids. The effects of ethidium bromide, acriflavin, proflavin, and 5-aminoacridine on the kinetoplast of L. tropica have been compared. Ethidium bromide and acriflavin, but not proflavin or 5-aminoacridine, induce dyskinetoplasty. L. tropica is one of the most sensitive trypanosomatids to ethidium bromide and acriflavin. Examination of the DNA from drug-treated cells in CsCl gradients revealed a loss of the satellite band after ethidium bromide or acriflavin treatment, but not after proflavin or 5-aminoacridine treatment. Leishmania tropica promastigotes do not utilize glucose provided in the medium until late log phase. Rapid depletion of glucose from the medium, however, occurs during late log and stationary phases. At about the same time, the cells show maximal rates of glucose uptake as well as peak levels of phosphofructokinase and pyruvate kinase activities. The glucose analog, 2-deoxy-D-glucose inhibits glucose transport. Incorporation of this analog in the growth medium results in inhibition of growth. The hexokinase of L. tropica phosphorylates 2-deoxy-D-glucose. Pyruvate kinase is activated by fructose-1, 6-diphosphate and adenosine monophosphate.  相似文献

Transport of glucose by Leishmania tropica promastigotes was measured by the uptake of the nonutilizable glucose analog, 2-deoxy-D-glucose (2-DOG), using the rapid filtration method. Both D-glucose and 2-DOG show identical rates of initial uptake. Intracellular 2-DOG readily exchanges with extracellular D-glucose and 2-DOG uptake is competitively inhibited by D-glucose. These observations suggest that both sugars are taken up by the same system. Neither the glucose analog α-methyl-D-glucoside (α-MG) nor 3-0-methyl glucose (3-0-MG) is taken up to any appreciable extent. Transport of 2-DOG shows saturation kinetics with a Vmax of 3.2 nmoles/mg cells/min and a Km of 0.16 mM. There is thus a stereospecific, carrier-mediated transport system for glucose uptake in L. tropica. About 2/3 of the intracellular pool following transport consists of 2-deoxy-D-glucose phosphate (2-DOG-P) and the remainder is free, unaltered 2-DOG.  相似文献

A new species of the genus Haminaca has been found in samplesfrom the northern Adriatic Sea. The new species is very similarto H. orteai, but may be clearly distinguished by body size,the non-pigmented area around the eyes, number of leaves ofHancock's organ, number of ridges on the gizzard plates andby the morphology of male genital tract.
