Amorphophallus paeoniifolius is an herbaceous perennial tuber crop distributed widely in many Asian countries. We isolated 19 polymorphic loci from A. paeoniifolius using a dual‐suppression‐PCR technique. These loci provide microsatellite markers with high polymorphism ranging from three to 24 alleles per locus. The observed and expected heterozygosities ranged from 0.521 to 0.854 and from 0.766 to 0.930, respectively. This high allelic diversity indicates that our markers are suitable for a population study in A. paeoniifolius.  相似文献

Acid phosphatase–2, as characterized by gel electrophoresis under non-denaturing conditions, is a convenient marker for prestalk cells in Dictyostelium discoideum . We have purified this prestalk-specific enzyme and have examined its regulation during development. Under denaturing conditions, the enzyme has a molecular weight of 50,000 and an isoelectric point of 4.0. On the other hand, acid phosphatase-I have a Mr-55,000 polypeptide (AP1–55) and a minor Mr-50,000 polypeptide (AP1–50) and both have diffuse isoelectric point from 3.4 to 4.1. Using monoclonal antibodies directed against acid phosphatase-2 as probes, we showed that some acid phosphatase-2 are newly synthesized at slug stage and some are converted from AP1–50 which was synthesized during ealy development.  相似文献

Exon II of glucokinase (Gk) was deleted to produce a systemic heterozygous Gk knockout (Gk+/−) mouse. The relative expression levels of Gk in the heart, lung, liver, stomach, and pancreas in Gk+/− mice ranged from 0.41–0.68 versus that in wild (Gk+/+) mice. On the other hand, its expression levels in the brain, adipose tissue, and muscle ranged from 0.95–1.03, and its expression levels in the spleen and kidney were nearly zero. Gk knockout caused no remarkable off-target effect on the expression of 7 diabetes causing genes (Shp, Hnf1a, Hnf1b, Irs1, Irs2, Kir6.2, and Pdx1) in 10 organs. The glucose tolerance test was conducted to determine the blood glucose concentrations just after fasting for 24 h (FBG) and at 2 h after high-glucose application (GTT2h). The FBG-GTT2h plots obtained with the wild strain fed the control diet (CD), Gk+/− strain fed the CD, and Gk+/− strain fed the HFD were distributed in separate areas in the FBG-GTT2h diagram. The respective areas could be defined as the normal state, prediabetes state, and diabetes state, respectively. Based on the results, the criteria for prediabetes could be defined for the Gk+/− strain developed in this study.  相似文献

Limpet home depressions in Cretaceous ammonites   总被引:2,自引:1,他引:1  
Remarkable large pits were found on the shell surface of many ammonites from the Turonian-Maastrichtian sediments of Hokkaido, northern Japan, and Sakhalin, Russia. These pits are (1) round to elliptical in shape; (2) up to 20 mm (usually around 10 mm) in diameter; (3) shallow depressions to deep holes that almost penetrate the shell: (4) occasionally healed by a thin shell blister from inside the shell; (5) often overlapping one another; (6) several to more than 170 in number on one flank of the ammonites; (7) found on both flanks and predominantly on the body chamber and the final volution of the phragmocone; and (8) found only in the two families Pachydiscidae and Puzosiidae, predominantly of more than 300 mm in shell diameter. They can best be interpreted as the home depressions of patellogastropod limpets. The presence of pits on both flanks of the ammonites and those healed from inside the shell strongly suggest that the limpets were dwelling on mature swimming ammonites. Host specifity, their very small shell size compared with the host ammonites, and sparse Occurrence in sediments favor a mode of life as obligate pseudoplankton. We suggest that this remarkable limpet-ammonite association was well established in northwestern Pacific bioprovinces during the late Cretaceous. Taking this live association and the depth limit of algal growth as food for the limpets into consideration, the mature ammonites dwelled or periodically visited the upper layer of the euphotic zone, probably less than around 20 m in depth. *** Ammonite, limpet, home depression, life mode, pseudoplankton, Cretaceous.  相似文献

CYCLIC SURFACE CHANGES IN THE NON-NUCLEATE EGG FRAGMENT OF XENOPUS LAEVIS   总被引:6,自引:6,他引:0  
Fertilized uncleaved eggs of Xenopus laevis were divided into nucleate and non-nucleate egg fragments. Both fragments, together with the whole egg of the same batch, were observed by time-lapse cinematography.
Two kinds of cyclic surface changes, (1) rounding-up and relaxing movements and (2) surface contraction waves, accompanying each cleavage in the whole eggs and the nucleate fragments, were also observed even in the non-nucleate fragments although they do not cleave.
Cleavage intervals of the whole egg and the nucleate fragment were nearly equal, but the rounding-up intervals of the non-nucleate fragment were slightly but definitely longer than the cleavage intervals of the nucleate fragment and the whole egg.  相似文献
