A strain of apple mosaic virus was isolated from hop plants in Japan. The virus was purified from young hop plants and back-inoculated to virus-free hop plants obtained by meristem tip culture. Inoculated plants developed chlorotic spots, ringspots and a band pattern accompanied by necrosis in the inoculated and systemically infected leaves. Shoot tips of infected plants sometimes became necrotic and these symptoms resembled those of a ring- and band-pattern mosaiclike disease prevalent in hop gardens in Japan. Since apple mosaic virus was recovered from infected plants, it is likely that the virus was the causal agent of this disease. Agar gel double diffusion tests and ELISA showed the hop virus to be serologically closely related to apple mosaic virus (ApMV), and distantly related to prunus necrotic ringspot virus (PNRSV). The virus had a narrow host range, and infected only cucumber of 18 species of Cucurbitaceae, Chenopodiaceae, Leguminosae or Solanaceae inoculated. It produced chlorotic spots on the inoculated cotyledons of cucumber, but no systemic infection. By contrast, ApMV from apple and PNRSV from peach had wide host ranges and infected cucumber plants systemically.  相似文献

A histochemical analysis was made of the differentiation ofcontact cells and isolation cells in the xylem ray parenchymaof Populus maximowiczii. The contact cells formed secondarywalls at approximately the same time as adjoining vessel elements.The lignification of the cell walls of contact cells and vesselelements began earlier than that of wood fibres and isolationcells. Thus, the formation of the secondary wall, includinglignification, of the contact cells might occur at the sametime as that of the vessel elements to which they are directlyconnected. By contrast, the isolation cells began to form secondarywalls later than the vessel elements and wood fibres in thevicinity of the isolation cells. After the deposition of thesecondary wall, a protective layer was formed in contact cellsbut no isotropic layer was observed in isolation cells. Eggs of Pseudocentrotus depressus were activated artificially by Loeb's "double treatment method". 50 min after activation, a number of asters were produced in the eggs. It was confirmed by electron microscopy that centrioles with a typical fine structure were present in artificially induced asters.
An unfertilized egg of Hemicentrotus pulcherrimus was divided into 2 halves, nucleated and non-nucleated, by centrifugation on a sucrose bed. Each half was activated by the same method as mentioned above. Several asters were produced in both halves after a certain period of incubation. The presence of bodies considered to be centrioles were demonstrated in the asters in both nucleated and non-nucleated halves.
The results add probability to the view that the centrioles are produced de novo in artificially activated eggs and fragments.  相似文献

Two kinds of peroxidase were isolated chromatographically frometiolated Alaska pea seedlings. One had absorption maxima at421 and 530 mµ, and the other, at 405, 500 and 640 mµ.From their affinities for cyanide and migrating patterns onelectrophoresis, the latter was concluded to be a modified formof the former. Epithelial-mesenchymal interaction in the early development of the anterior pituitary gland was examined by chronological observations on fetal pituitary epithelium grafted in vivo with and without its own mesenchyme. At 8.5 days of gestation, the RATHKE'S pouch began to evaginate toward the diencephalon. The mesenchymal tissue around the pouch was at first very sparsely scattered, but then condensed, on day 10 becoming visible under a dissecting microscope. When RATHKE'S pouch epithelia from 10- and 12-day fetuses were transplanted alone under the kidney capsule, they proliferated slightly to form cysts, the cells of which differentiated into ACTH-producing cells, but not into prolactin-producing cells. Pituitary morphogenesis did not occur. When these epithelia were recombined with homotypic mesenchyme and transplanted, the epithelia proliferated remarkably on one side of the wall of the pouch, resulting in formation of a pars distalis that contained both ACTH-producing cells and prolactin-producing cells. Heterotypic mesenchyme, such as lung, dermis and mammary gland mesenchyme, could induce 12-day epithelium, but not 10-day epithelium to develop into pars distalis. Thus, fetal pituitary epithelium has the capacity of autodifferentiation into ACTH-producing cells, not into prolactin-producing cells, and requires mesenchymal support for development of the pars distalis.  相似文献

Spermatozoa can enter the separated blastomeres of 8- and 16-cell stage embryos, the cells of blastulae and even somatic cells of the oesophagus wall of an adult sea urchin, under certain conditions. In the presence of egg jelly solution, the rate of entrance of spermatozoa is remarkably increased. In the case of the blastomere of 8-cell stage embryos, characteristic cytoplasmic protrusions are formed at the sites of sperm entry, in succession to the formation of the cytoplasmic bulge. These protrusions elongate until 4 min after insemination, and then they retract gradually. The nucleus of penetrated sperm swells and decondenses to form a pronucleus. In most cases, the pronucleus seems to fuse with the preexisting diploid nucleus of the blastomere. When the dissociated oesophagus cells were inseminated, a certain type of the cells was found to fuse with spermatozoa, although the percentage of fused cells was very low.  相似文献

When spermatozoa are treated with egg-water and undergo the acrosome reaction, their fertilizing capacity is lost within 5 min. However, if insemination is carried out within 4 min after the egg-water treatment, there is no difference in fertilizing capacity between spermatozoa treated with egg-water and non-treated ones. With such spermatozoa, eggs can be fertilized even in the virtual absence of calcium, whereas with spermatozoa treated with Ca-free egg-water, no fertilization occurs under the same conditions. It is postulated that in normal fertilization the acrosome reaction has occurred before the attachment of the gametes. The failure of fertilization with normal spermatozoa in Ca-free sea water may be due to the failure of occurrence of the acrosome reaction.  相似文献

An anastomosing network of extracellular fibrils on the inner surface of the ectoderm layer of amphibian gastrulae has been shown to provide an adequate substratum for attachment and migration by the mesodermal cells. These fibrils contain fibronectin as shown by immunostaining at the light and electron microscope levels. Now we report the presence of laminin, another cell adhesion glycoprotein, as a fibrillar network on the inner surface of the ectoderm layer in gastrulae of the Japanese newt ( Cynops pyrrhogaster ), but its absence on the blastula ectoderm layer, by the immunofluorescent staining using an antiserum specific for mouse laminin. The same antiserum was shown to stain basement membranes of adult newt organs as expected.  相似文献
