To elucidate the regulatory mechanism for tyrosinase gene expression in vivo, we microinjected a mouse tyrosinase minigene, mg-Qrs-J, into the fertilized eggs of BALB/c albino mice. As a result, we obtained six pigmented founder mice that exhibited non-standard coat color variations as well as the wild-type phenotype. These founder mice were subsequently crossed with BALB/c albino mice to establish the transgenic lines. As a consequence, two primary lines and five sublines have been obtained from four of the six founder mice. We found that not only uniformly pigmented phenotypes but also patterned phenotypes were inherited by their descendants. The possible underlying mechanism of the patterned phenotypes is discussed. 相似文献

玉米大斑病是严重危害玉米生产的一个世界性真菌病害。由于玉米大斑病菌(Exserohilum turcicum)在无性生长过程中迅速产生黑色素,致使原生质体难以分离。测试了包括Fungase、Funcelase、Novozyme、Glucanex、Driselase、Uskizyme、Kitalase在内的7种细胞壁降解酶及其组合、病原菌菌株和培养基对原生质体分离效果的影响。结果表明菌株的培养形态和菌丝的生长状态显著影响原生质体的分离效率;酶组合Kitalase Glucanex Driselase,Kitalase Glucanex和Kitalase Uskizyme能够有效地分离玉米大斑病菌的原生质体。初步的转化试验表明,质粒pAN71可以用于该病原菌的转化。这些结果将为E.turcicum和Exserohium属其它真菌的基因克隆提供一些有用的信息。 相似文献

Summary In order to complete growth and development, the endoparasitoid wasp, Cotesia (=Apanteles) kariyai, inhibits pupation of its armyworm host, Pseudaletia (=Leucania) separata. In host larvae retardation of testis and spermatocyst development caused by the parasitoid was also observed. The agents causing the retardation were found in the ovaries and venom of the female adult parasitoid. When an unparasitized male host larva was artificially injected with calyx fluid obtained from ovaries together with venom, it showed the same degree of developmental retardation of testes and spermatocysts as in natural parasitization. Testes implanted in isolated abdomens of healthy larvae did not increase in size by ecdysteroid stimulation after exposure to calyx fluid plus venom. It is suggested that both symbiotic polydnavirus existing in calyx fluid and venom in the parasitoid, C. kariyai, are responsible for the parasitic retardation of the male reproductive organs in the host, P. separata. 相似文献

The genetic and hormonal control of body colouration is investigated using two recessive genetic mutant strains, the reddish–brown (RB) mutant and an albino mutant, as well as a normal (pigmented) strain of the desert locust Schistocerca gregaria. The colour patterns of the RB nymphs are similar to those of a normal strain, although the intensity of the melanization is weaker in the former. Reciprocal crosses between the RB and albino mutants produce only normal phenotypes in the F1 generation. In the F2 generation, the normal, RB and albino phenotypes appear in a ratio of 9 : 3 : 4, indicating that two Mendelian units might determine the appearance of dark body colour and the intensity of melanization, respectively. In other words, at least two steps of regulation might be involved in the expression of body colour. Injections of [His7]‐corazonin, a neuropeptide inducing dark colour in this locust, fail to induce dark colour in albino nymphs but show a dose‐dependent darkening in RB nymphs in the range, 10 pmol to 1 nmol. Some RB nymphs become indistinguishable from normal individuals after injection of the peptide. Implantation of corpora cardiaca (CC) taken from RB mutants into other RB individuals induces darkening in the latter and CC from RB, albino and normal strains have similar dark colour‐inducing activity when implanted into albino Locusta migratoria. These results suggest the possibility that the RB mutant gene regulates the intensity of melanization, possibly through controlling the pathway of pigment biosynthesis associated with [His7]‐corazonin. 相似文献

Coat colors of four chimeric pigs produced by the microinjection of dissociated blastomeres of (Landrace × Large White) blastocysts to the blastocyst cavity of (Duroc × Duroc) blastocysts (Kashiwazaki et al., 1992) exhibited characteristic horizontal stripe-patterns. We carried out quantitative analysis of those patterns in order to derive information concerning the genetic regulatory mechanisms of the dominant black-eyed white phenotypes in the pig. In the four chimeras, the theoretical mean widths of the single-clone stripe calculated from the estimated widths of minimal recognizable stripe (MRS) (Tachi, 1988) were 2.1 ± 0.1, 2.23 ± 0.15,1.89 ± 0.06, and 1.93 ± 0.28 cm respectively. The estimated number of single-clone stripes in the thoracico-lumbar region of those animals were 42.3, 40.7, 46.3, 44.2, and about twice the mean number of vertebrae in the same region (Duroc, 20 or 21; Large White, 21 or 22). Furthermore, the mean length of thoracico-lumbar vertebrate in two of the chimeric pigs, as measured on X-ray radiographs, was approximately twice the mean single-clone stripe width. It was concluded that the stripe-patterns of the chimeric pigs probably represented the dermatome patterns of epidermis; and in the pig, a single somite was likely to be derived from the clones of two primordial cells, as originally proposed by Gearhart & Mintz (1972) in the mouse. It was suggested, furthermore, that in the Large White→Duroc chimeric pigs, melanocytes that migrated into the region of skin formed by a Large White dermatome could not survive, thus creating a clearly demarcated white stripe. The effects of five environmental factors, background color, density, humidity, photoperiod and temperature, on the body color of a pyrgomorphid green/brown polymorphic grasshopper, Atractomorpha lata, were examined in rearing experiments, in which the differences in color morph frequencies between males and females were also examined. Field censuses were conducted to determine whether the sex difference in body‐color polymorphism occurs under field conditions. Among the environmental factors examined, temperature was the only factor that significantly affected body color: the frequency of brown morphs tended to be higher at higher temperatures. The frequency of brown morphs in the female was higher than in the male, a consistent finding in both the rearing experiments and the field censuses. The threshold temperature at which body color changes may be lower in the female than in the male. 相似文献
