Clonal culture of muscle cells from thigh muscles of the term fetus of the mouse was undertaken. Myoblasts were spherical or spindle-shaped, and proliferated exponentially until day 4 in culture. The generation time of the muscle cells from 2 to 4 days' culture was 9.1 to 13.4 hr. The fusion of myoblasts began on day 4 in culture; many myotubes had been formed by day 6 and spontaneous contraction was observed on day 7. Clonal efficiency was 30%, and the proportion of muscle colonies in all the colonies was 72%.  相似文献

1. Morphological characteristics, especially coloration, are related to thermoregulation and camouflage, both of which are crucial for species survival and fitness. In cool environments such as the understorey of closed rainforests, darker organisms have thermal advantages and may be able to absorb heat more efficiently. However, such habitats are also suitable for darker organisms with respect to camouflage, making it difficult to elucidate whether the association of dark‐coloured organisms with shady environments is a consequence of thermal stress or predation pressure, or both. 2. In this study, butterfly communities were surveyed and artificial butterflies (mealworms attached to plastic sheeting to mimic adult butterflies) used to test whether differences in wing luminance are related to predation rates within open and closed habitats in monsoonal tropical forests of southwestern China. 3. Using artificial butterflies, significantly lower predation rates were found for dark‐coloured artificial butterflies within closed habitats, whereas such relationships were not found within open habitats. It was found that actual butterfly communities were also significantly darker in closed than in open habitats. 4. These results demonstrate that darker colours may have the effect of reducing predation rates in shady environments and that different habitat types can have contrasting effects on luminance and therefore predation risk.  相似文献

Round spermatids (steps 1–8) were isolated from rat testes and the effect of pyruvate on their intracellular ATP level was examined. Results showed that although the spermatids consumed a considerable amounts of pyruvate, this substrate alone did not maintain their ATP level. However, their ATP level was maintained in the presence of both pyruvate and α-ketovalerate or α-ketobutyrate. Maintenance of the ATP level by these substrates was associated with electron trasnport and oxidative phosphorylation. α-Ketoacid inhibited pyruvate reduction to lactate in the lactate dehydrogenase (LDH) reaction, but increased pyruvate oxidation to CO2. The NADH level in spermatids was too low to be detectable, but the NAD level remained unchanged in the presence of pyruvate and α-ketovalerate. These results suggest that pyruvate by itself is not an adequate energy-yielding substrate for spermatids and that a high NADH/NAD ratio may be essential for maintenance of their ATP level.  相似文献

The coconut hispine beetle Brontispa longissima has been causing serious damage to the coconut palm Cocos nucifera in the Pacific, and Southeast and East Asia. This beetle also attacks Satakentia liukiuensis, an endemic palm on Ishigaki and Iriomote Islands, Japan. To assess the potential impact of B. longissima on S. liukiuensis, we examined the development period, survival rate, egg production and body size of B. longissima on S. liukiuensis at 22–31°C and consumption of S. liukiuensis leaf at 28°C. We also examined these life history traits on C. nucifera at 28°C to compare with those on S. liukiuensis. Brontispa longissima completed their development and reproduced on S. liukiuensis at 22–30°C. Linear regression analysis was used to estimate the effective cumulative temperature (k) as 891.2 degree‐days, and the lower developmental threshold as 13.3°C. On the basis of these values and monthly average temperatures on Ishigaki Island, it was estimated that B. longissima has four generations per year on the island. Satakentia liukiuensis was less suitable for B. longissima than C. nucifera in terms of immature survival, development time, resultant adult size and reproduction. The low quality of S. liukiuensis as a host‐plant may prevent the outbreak of B. longissima, which has not occurred yet on Ishigaki and Iriomote Islands.  相似文献

Vertical and seasonal distributions of flying beetles were investigated in asuburban temperate deciduous forest in Kanazawa, Japan using water pan traps to determine the abundance and composition among vertical strata, change in the abundance and composition through seasons and determinant factors in generating the distributions. Traps were placed at three levels (0.5 m, 10 m, and 20 m above ground) on a tower. Samplings were carried out seasonally from May to November in 1999 and 2000. Variations in the abundance of flying beetles were observed from different layers. The results showed that the abundance and composition of flying beetles varied among strata and seasons. In both 1999 and 2000,Elateridae was consistently most abundant in the bottom layer, while Attelabidae and Cantharidae were most abundant in the upper layer. In 1999, Eucnemidae and overall scavengers were most abundance in the bottom layer, but results were not consistent with those in 2000. In general, the abundance of herbivores reaches a peak in the early season(May/June) and decreases in the following months. Peaks of abundance in predators vary vertically. In the bottom layer a peak was observed in the early season (May/June), while in the upper layer this was observed in July. Scavengers had two peaks, in May/June and September. These patterns indicated that vertical distributions in the abundance of differentfeeding guilds varied through seasons.  相似文献

Understanding of leaf stomatal responses to the atmospheric CO2 concentration, [CO2], is essential for accurate prediction of plant water use under future climates. However, limited information is available for the diurnal and seasonal changes in stomatal conductance (gs) under elevated [CO2]. We examined the factors responsible for variations in gs under elevated [CO2] with three rice cultivars grown in an open‐field environment under flooded conditions during two growing seasons (a total of 2140 individual measurements). Conductance of all cultivars was generally higher in the morning and around noon than in the afternoon, and elevated [CO2] decreased gs by up to 64% over the 2 years (significantly on 26 out of 38 measurement days), with a mean gs decrease of 23%. We plotted the gs variations against three parameters from the Ball‐Berry model and two revised versions of the model, and all parameters explained the gs variations well at each [CO2] in the morning and around noon (R2 > 0.68), but could not explain these variations in the afternoon (R2 < 0.33). The present results provide an important basis for modelling future water use in rice production.  相似文献

The following results were obtained using etiolated Alaska pea epicotyls. Gibberellic acid (GA) had the remarkable effect on the elongation in part I (elongating region) of epicotyls, whereas it had little effect on that in part II (mature region) of epicotyls. In cortex of part I and II of epicotyls, the cell number in longitudinal direction was hardly affected by GA. On the increase in width of epicotyls, GA was hardly effective in any parts of epicotyls. In both part I and II GA enhanced the incorporation of 32P into all nucleic acid fractions prepared by methylated albumin kieselguhr (MAK) columns, i.e. sRNA, DNA and rRNA + mRNA. In part I the net increase in DNA and RNA content during the incubation period was slightly promoted by GA, whereas in part II the net decrease in both nucleic acids content was slightly promoted by GA. The relationship between GA-induced growth and nucleic acid synthesis is discussed.  相似文献
