The intermediate-sized filaments present in epidermal keratinocytes derived from mouse skin and in an established cell line (HEL) derived from spontaneous transformation of murine keratinocytes grown in vitro, have been examined by immunofluorescence microscopy, using antibodies directed against subunit proteins of different classes of intermediate-sized filaments, as well as by electron microscopy and gel electrophoresis of cytoskeletal preparations highly enriched in intermediate-sized filaments. The keratinocytes derived from neonatal skin, which are capable of only limited replication in vitro, show only a single type of intermediate-sized filaments, i.e., the tonofibril-like arrays of filaments containing prekeratin. HEL cells, which proliferate indefinitely in vitro, retain the tonofilament-like structures typical of differentiated epidermal cells but in addition display intermediate-sized filaments of the vimentin type, i.e., the filament system typically found in mesenchymal and mesenchyme-derived cells. We discuss the possibility that (i) the advent of vimentin-type filaments in epidermal cells in culture is related either to the transformed state or the in vitro growth conditions as such and (ii) other differentiated epithelial cells proliferating in vitro may have more than one system of intermediate-sized filaments.  相似文献

The flowering pattern of plant species, including orchid species, may fluctuate irregularly. Several explanations are given in the literature to explain that pattern, including: costs associated with reproduction, herbivory effects, intrinsically triggered unpredictable variation of the system, and external conditions (i.e. weather). The influence of age is discussed, but is difficult to determine because relevant long-term field observations are generally absent in the literature. The influence of age, size, reproductive effort and climatic conditions on flowering variability of Himantoglossum hircinum are examined using data collected in a long-term project (1976–2001) in Germany. PCA and multiple regression analysis were used to analyse variability in flowering pattern over the years as a function of size and weather variability. We studied future size after flowering to quantify costs of reproduction. Flowering probability was strongly determined by plant size, while there was no significant influence of age class on flowering probability of the population. Costs associated with reproduction resulted in a decrease in plant size, causing reduced flowering probability of the plants in the following year. The weather explained about 50% of the yearly variation in the proportion of large plants and thus had an indirect, strong influence on the flowering percentage. We conclude that variability in flowering is caused mainly by the variability of weather conditions in the previous and current year, whereby reproductive effort causes further variability in flowering at the individual and, consequently, the population levels.  © 2006 The Linnean Society of London, Botanical Journal of the Linnean Society , 2006, 151 , 511–526.  相似文献

THERE is little or no humoral cross reactivity between the native and denatured forms of a protein antigen. Antibodies to native bovine ribonuclease do not bind ribonuclease oxidized with performic acid1–3, indicating that antibodies can distinguish between two molecular conformations. There is no humoral cross reactivity between rabbit IgG and the product of reduction of all the disulphide bridges4–6. Abstract The short‐lived adult wheat midge Sitodiplosis mosellana deposits eggs on the seed head of various grasses close to the developing seeds on which larvae feed. The time taken to make egg‐laying decisions is investigated using three types of wheat Triticum aestivum seed heads. Young Roblin, Old Roblin and Young Key differ in their effects on ovipositing females (72%, 22% and 6% of eggs in choice tests, respectively) and effects on feeding larvae (75%, 25% and 5% larval survival, respectively). Within seconds of arriving, the female is able to distinguish Young Roblin from the two lower‐ranked types. However, the lower‐ranked types are not rejected at this time. Instead, all head types are examined before the female eventually flies away. On Young Roblin, probing with the ovipositor is the first behaviour that occurs. Thereafter probing and insertion of the ovipositor occupy most of the female’s time and behavioural transitions tend to be ‘progressive’, signalling a shift from low to high intensity examining. Differences between females visiting Old Roblin and Young Key are significant but take longer to emerge. On both, sitting is the first behaviour but, over the next 5–10 min, the female on Young Key exhibits more sitting, walking and ‘regressive’ transitions than the female on Old Roblin. It is suggested that, when the ovipositing female is short‐lived and incapable of controlled flight in all but essentially windless conditions, her behaviour is designed to thoroughly, rather than rapidly, examine a suboptimal host before abandoning it for the uncertain future of finding a better host.  相似文献

Eight pathotypes of Plasmopara halstedii were screened to investigate the occurrence of virions and the potential viral influence on the pathogenicity of the sunflower downy mildew pathogen. In 23 of 26 P. halstedii isolates derived from eight countries in Europe, North America and South America, virions were detected by transmission electron microscopy. By contrast, there were no ultrastructural indications of virus‐like particles in eight other related Oomycetes. The virions of representative P. halstedii isolates were morphologically and biochemically characterized and compared among each other. Regardless of their host's pathotypes, the geographical origin of the isolate and the sensitivity towards the fungicide metalaxyl, the viral characters obtained were uniform. The virions were isometric and measured approximately 37 nm in diameter. One polypeptide of c. 36 kDa and two segments of single‐stranded RNA (3.0 and 1.6 kb) were detected. Both viral RNA segments were detected by capillary electrophoresis in the three remaining P. halstedii isolates where virions were undetectable by transmission electron microscopy. Virus‐specific primers for the 1.6 kb‐segment were synthesized and used to determine and compare a partial sequence of the viral coat protein among virions of different P. halstedii pathotypes. In all tested isolates, fragments of 0.7 kb were amplified which were directly sequenced. Sequence variation was insignificant. As both less aggressive and more aggressive P. halstedii isolates contained virions, the presence or absence of virions could not explain the diverse aggressiveness of the downy mildew pathogen towards sunflower. Moreover, the results indicated that pathogenicity of P. halstedii was not related to variation in morphological or biochemical characters of the virions.  相似文献

The taxonomic distribution and evolution of viviparity in Diptera is critically reviewed. The phenomenon ranges from ovoviviparity (eggs deposited at an advanced stage of embryonic development; larva emerges immediately after deposition), through viviparity (larva hatches inside female before deposition) to pupiparity (offspring deposited as pupa). Some Diptera are known to be facultatively viviparous, which is hypothesized to be a step towards the evolution of obligate viviparity. Obligate viviparity is found to comprise unilarviparity (single large larva in maternal uterus) which evolved many times independently, the rare oligolarviparity (more than one but not more than 12 larvae) and multilarviparity (large numbers of developing eggs or larvae in uterus) which is typical for the two largest clades of viviparous Diptera. Unilarviparity is either lecithotrophic (developing larva nourished by yolk of egg) or pseudo-placental (larva nourished by glandular secretions of mother). Viviparity has clearly evolved on many separate occasions in Diptera. It is recorded in 22 families, and this review identifies at least 61 independent origins of viviparity. Six families appear to have viviparity in their ground-plan. Some families have a single evolution of viviparity, others multiple evolutions. Guimaraes' model for the evolution of viviparity in Diptera is tested against phylogenetic information and the adaptive significance of viviparity is reviewed in detail. Possible correlations with life-history parameters (coprophily, parasitism, breeding in ephemeral plant parts, malacophagy and adult feeding habits – especially haematophagy) are analysed critically, as are potential advantages (shorter larval life, less investment in yolk by mother, protection of vulnerable stages, better access to breeding substrates, predation on competitors). Morphological constraints, adaptations and exaptations are reviewed, including the provision of an incubation space for the egg(s), the positioning of the egg(s) in the uterus, and maternal glands. The main morphological adaptations include greater egg size, reduction of egg respiratory filaments, thinning of chorion, modified larval respiratory system and mouthparts, and instar skipping. Female morphology and behaviour is particularly strongly modified for viviparity. The terminalia are shortened, the vagina is more muscular and tracheated, and the ovaries of unilarviparous species have a reduced number of ovarioles with alternate ovulation. Many of the final conclusions are tentative, and a plea is made for more detailed morphological and experimental study of many of the viviparous species. Viviparity in Diptera provides a fascinating example of multiple parallel evolution, and a fertile field for future research.  相似文献

The reproductive tract of B. truncatus was investigated histologically in order to study possible effects of castration upon the accessory sex glands. In the female part of the reproductive tract—subdivided into albumen gland, oviduct, muciparous gland, oothecal gland, uterus, vagina and bursa copulatrix—13 histochemically different secretory cell types were distinguished. The majority produce different types of (acid) mucopolysaccharides. The roles of the various parts of the female tract in the production of an egg mass were elucidated by comparing the histochemistry of the egg mass to that of the female tract; the abundance and location of the cell types were also taken into account for this purpose.

The male part appeared to contain 12 histochemically different secretory cell types. These produce mainly (phospho lipoproteins together with some polysaccharides and neutral lipids.

Castration causes an acceleration of the growth of the snails. The volumes of female (albumen gland) and male (prostate gland) accessory sex glands were measured on histological sections. It appeared that growth of the albumen gland is not arrested by castration. This was not established beyond doubt for the prostate gland. The results suggest that the stimulating effects of the dorsal body hormone on the growth and synthetic capacity of the female accessory sex glands—such effects have been established for Lymnaea stagnalis—are not exerted via the ovotestis in B. truncatus.  相似文献
