A diversity of evolutionary processes can be responsible for generating and maintaining biodiversity. Molecular markers were used to investigate the influence of Plio-Pleistocene climatic oscillations on the evolutionary history of taxa restricted to the freshwaters of a classical glacial refugium. Population genetic, phylogenetic and phylogeographical methods allowed the inference of temporal dynamics of cladogenesis and processes shaping present-day genetic constitution of Barbus sclateri , a polytypic taxon found in several independent river drainages in southern Iberian Peninsula. Results from different analyses consistently indicate several range expansions, high levels of allopatric fragmentation, and admixture following secondary contacts throughout its evolutionary history. Using a Bayesian demographical coalescent model on mitochondrial DNA sequences calibrated with fossil evidence, all cladogenetic events within B. sclateri are inferred to have occurred during the Pleistocene and were probably driven by environmental factors. Our results suggest that glaciation cycles did not inhibit cladogenesis and probably interacted with regional geomorphology to promote diversification. We conclude that this polytypic taxon is a species complex that recently diversified in allopatry, and that Pleistocene glaciation–deglaciation cycles probably contributed to the generation of biological diversity in a classical glacial refugium with high endemicity.  相似文献

1. In contrast to extensive studies of zooplankton in lakes, the role of microcrustaceans in wetlands is not well studied. In this study, spatial and temporal patterns of microcrustacean assemblage structure and secondary production were quantified over a 2-year period in a southeastern U.S.A. wetland.
2. Thirty-two species, including 19 cladocerans, 10 copepods and three ostracods, generated different temporal patterns of density and production between vegetated ( Nymphaea ) and non-vegetated (open-water) zones reflecting species-specific differences in life histories.
3. Summer assemblages were dominated by small, planktonic filter-feeders, typified by high annual production/biomass ( P / B ) and daily production. In contrast, winter assemblages were dominated by larger, epibenthic detritivores with low P / B and high biomass. Seasonal shifts in the relative importance of planktonic species in the warmer months to benthic and epiphytic species in the cooler months suggest that energy flow pathways through microcrustaceans may vary seasonally.
4. Total annual production was higher during both years in the Nymphaea zone (13.0 g and 13.6 g DM m−2 year−1) than the open-water (8.2 and 6.3 g DM m−2 year−1), and was similar between years for the entire wetland pond (12.3 and 12.2 g DM m−2 year−1).
5. Although wetland ecosystems have been the subject of considerable ecological research in the past 20 years, our study is one of the few to demonstrate a highly diverse and relatively productive microcrustacean assemblage. Such comprehensive production studies can be used to quantify the ecological importance of microcrustaceans in freshwater wetland ecosystems.  相似文献

  • 1 Bottlenose dolphins Tursiops truncatus are amongst the best‐known cetaceans. In the Mediterranean Sea, however, modern field studies of cetaceans did not start until the late 1980s. Bottlenose dolphins have been studied only in relatively small portions of the basin, and wide areas remain largely unexplored.
  • 2 This paper reviews the ecology, behaviour, interactions with fisheries and conservation status of Mediterranean bottlenose dolphins, and identifies threats likely to have affected them in historical and recent times.
  • 3 Whilst intentional killing was probably the most important cause of mortality until the 1960s, important ongoing threats include incidental mortality in fishing gear and the reduced availability of key prey caused by overfishing and environmental degradation throughout the region. Additional potential or likely threats include the toxic effects of xenobiotic chemicals, epizootic outbreaks, direct disturbance from boating and shipping, noise, and the consequences of climate change.
  • 4 The flexible social organization and opportunistic diet and behaviour of bottlenose dolphins may allow them to withstand at least some of the effects of overfishing and habitat degradation. However, dolphin abundance is thought to have declined considerably in the region and management measures are needed to prevent further decline.
  • 5 Management strategies that could benefit bottlenose dolphins, such as sustainable fishing, curbing marine pollution and protecting biodiversity, are already embedded in legislation and treaties. Compliance with those existing commitments and obligations should be given high priority.

ABSTRACT. We report the characterization of cell-associated and extracellular peptidases of Bodo sp., a free-living flagellate of the Bodonidae family, order Kinetoplastida, which is considered ancestral to the trypanosomatids. This bodonid isolate is phylogenetically related to Bodo caudatus and Bodo curvifilus . The proteolytic activity profiles of Bodo sp. were determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis containing co-polymerized gelatin, casein, hemoglobin, or bovine serum albumin as substrates. The enzymatic complex degraded gelatin better in acidic pH, and under these conditions four proteolytic bands (120, 100, 90, and 75 kDa) were detected in the cellular or extracellular extracts. Two peptidases (250 and 200 kDa) were exclusively detected with the substrate casein. All these enzymes belong to the serine peptidase class, based on inhibition by aprotinin and phenylmethylsulfonyl fluoride. This is the first biochemical characterization of peptidases in a free-living Bodo sp., potentially providing insight into the physiology of these protozoa and the evolutionary importance of peptidases to the order Kinetoplastida as some of these enzymes are important virulence factors in pathogenic trypanosomatids.  相似文献

Tobacco (Nicotiana tabacum) Bright Yellow‐2 (TBY‐2) cells undergo different fates when exposed for 10 minutes to heat stresses of different severity. A 35 °C treatment causes a homeostatic response (HRE) allowing cells to cope with the stress; 55 °C triggers processes leading to programmed cell death (PCD), which is complete after 72 h. We have used a proteomic approach to gain insight into the molecular mechanisms defining the fate of TBY‐2 cells induced by these two heat stresses. Tandem mass spectrometry (MS/MS) and two‐dimensional electrophoresis (2‐DE) analysis revealed little overlap of differentially‐accumulated proteins: the different severities of heat treatment induced the modulation of specific proteins, some of which are responsible for different cell fates. When the imposed heat shock is beyond a certain threshold, the overall reduced metabolism may be the result of a series of events involving gene expression and oxidative damage that would lead to PCD. Our data suggest that the down‐accumulation of several proteins involved in cellular redox homeostasis could provide, until now, an unappreciated contribution to understanding how many partners are involved in promoting the redox impairment leading to PCD. Moreover post‐translational modifications seem to play important regulatory roles in the adaptation of TBY‐2 cells to different intensities of heat stress.  相似文献

Empirical evidence based on grazing exclusion at the scale of years to decades shows that grazing modifies carbon (C) and nitrogen (N) cycling. However, long‐term effects at the scale of centuries are less known, yet highly relevant to understand local and global impacts of grazing. Additionally, most studies have focused on the isolated response of C and N, with little understanding of their interactions. Using CENTURY, a process‐based biogeochemical model, we analyzed the impacts of 370 years of livestock grazing (i.e. long term, from early European colonization to present) in 11 sites across the Río de la Plata grasslands and compared them with those resulting from two decades of grazing (i.e. mid‐term, typical exclosure experiment). In the long term, livestock grazing primarily altered the N cycle through faster N returns to the soil via urine and dung, which were offset by uninterrupted N outputs by volatilization and leaching. As a result, soil organic N decreased by ?880 kg ha?1 or ?19%. Higher N outputs (mainly as NH3) opened the N cycle, potentially decreasing N2O and NOx emissions and increasing N depositions over the region. These greater outputs of N constrained C accumulation in soils, reducing soil organic C by ?21 200 kg ha?1 (?22%, a reduction of ?1.5 Pg of C for the whole region) and net primary production by ?2192 kg ha?1 yr?1 (?24%). Mid‐term simulations showed that the effects of livestock introduction in a decadal time scale were substantially different both in magnitude and direction from long‐term responses. Long‐term results were not substantially affected when atmospheric CO2 content, species composition and fire regime were changed within plausible ranges, but highlighted fire‐grazing interactions as a major constraint of long‐term C and N dynamics in these grasslands.  相似文献

The hybrid origin of the western Mediterranean orchid Dactylorhiza insularis was demonstrated by genetic markers. Allozyme data showed that throughout its range D. insularis has an allotriploid constitution and reproduces apomictically. The parental species of D. insularis were identified as D. romana andD. sambucina; they contributed 2 alleles and 1 allele, respectively, at the allozyme loci studied. The maternal species of D. insularis was D. romana , as inferred from cpDNA ( trn L(UAA) intron). High genetic similarities were found when comparing present populations of D. romana and D. sambucina with their respective genomes 'frozen' in D. insularis. Dactylorhiza insularis showed fixed (or nearly fixed) heterozygosity at 11 out of the 19 loci studied, and poor genetic variation: eight multilocus genotypes were detected at allozyme level. No multilocus genotype differs from the most similar one by more than one allele substitution. All D. insularis individuals showed the same cpDNA haplotype (I) , regardless of their geographic origin and multilocus genotype. The I haplotype is similar, but not identical to that found in D. romana (R). No recurrent formation of D. insularis was observed in hybrid zones between D. romana and D. sambucina , where diploid sexual hybrids (F1; Fn, backcrosses) were detected. Available data agree with a single origin for D. insularis , which possibly occurred in the present postglacial, when D. romana and D. sambucina , expanding from their glacial refugia, came into contact. The genetic homogeneity found between D. romana and D. markusii , both from their locus classicus , indicates that the latter is a junior synonym of D. romana; on the other hand, D. romana and D. sambucina are well differentiated species ( DNei = 0.59).  相似文献

Life reconstructions are a useful means of providing a package of information about morphology, functionality, behaviour, biology and ecological characteristics of an extinct organism. These reconstructions are of interest not only to researchers but also to a wider public. Reconstructions of sabretoothed cats in particular are widely published and exhibited, despite the absence of a general consensus on either how their canines were used or the prey sought. Cat-like restorations of the American Pleistocene sabretooth genus Smilodon prepared by Charles Knight under the direction of J.C. Merriam were accepted as valid for over three decades until GJ. Miller criticized them, claiming that Smilodon should have looked very different from modern felids. In particular, he argued for a longer mouth opening and lip line to provide a wider gape, a retracted nose and ears set relatively lower on a head with a straighter dorsal profile. These arguments were accepted by many authors employing reconstructions, and have lead to depictions of bizarre appearance and interpretations of rather specialized feeding behaviour. We believe that phylogenetic, anatomical and functional considerations point to substantial flaws in the basis for such depictions, and argue for a return to more felid-like morphology and to interpretations of broadly cat-like eating patterns.  相似文献
