Crithidia deanei from the reduviid hemipteron, Zelus leucogrammus, unlike most lower trypanosomatids cultivated in defined medium, required only 2 amino acids, methionine and tyrosine; only 4 vitamins, folic acid, thiamine, biotin, and nicotinamide; and neither hemin nor a purine source. Electron microscopy reveals an endosymbiont, probably bacterial, which presumably provides the other basic trypanosomatid essential nutrients.  相似文献

Age was determined in three anuran species inhabiting a temperate oceanic climate ( Rana temporaria, Bufo calamita and Bufo bufo ) by means of the histological structure of the phalanges. Narrow, haematoxylinophilic lines were visible in these, and evidence from the examination of subadults and frog ( R. temporaria ) recaptures showed the lines to be annual. The validity of using the lines as age indicators was tested most extensively on natural frog populations and the effects of endosteal resorption were not found to be significant. The evidence for this conclusion was provided directly by recapture data and indirectly by an analysis of histological measurements made on the phalanges.  相似文献

Pigtailed macaques (Macaca nemestrina) provide an important model for biomedical research on human disease and for studying the evolution of primate behavior. The genetic structure of captive populations of pigtailed macaques is not as well described as that of captive rhesus (M. mulatta) or cynomolgus (M. fascicularis) macaques. The Washington National Primate Research Center houses the largest captive colony of pigtailed macaques located in several different housing facilities. Based on genotypes of 18 microsatellite (short tandem repeat [STR]) loci, these pigtailed macaques are more genetically diverse than captive rhesus macaques and exhibit relatively low levels of inbreeding. Colony genetic management facilitates the maintenance of genetic variability without compromising production goals of a breeding facility. The periodic introduction of new founders from specific sources to separate housing facilities at different times influenced the colony's genetic structure over time and space markedly but did not alter its genetic diversity significantly. Changes in genetic structure over time were predominantly due to the inclusion of animals from the Yerkes National Primate Research Center in the original colony and after 2005. Strategies to equalize founder representation in the colony have maximized the representation of the founders’ genomes in the extant population. Were exchange of animals among the facilities increased, further differentiation could be avoided. The use of highly differentiated animals may confound interpretations of phenotypic differences due to the inflation of the genetic contribution to phenotypic variance of heritable traits. Am. J. In two heavily infested rural villages of Santiago del Estero, Argentina, where no indoor-spraying with residual insecticides had ever been carried out by official control services, we studied the influence of roof and wall structure, domestic use of insecticide, family size and the number of domestic dogs, on the domiciliary density of Triatoma infestans (Klug). Bug density was significantly associated with (1) the interaction between insecticide use and type of roof, (2) the structure of indoor walls, (3) the number of dogs sharing sleeping areas of people (room-mate dogs), and (4) the number of people plus room-mate dogs, but not with just the number of people resident in the house. The interaction between insecticide use and a roof made of 'simbol', a locally available grass (Pennisetum sp.), also reflected a younger age structure of domestic bug populations. In infested houses, the density of bugs infected with Trypanosoma cruzi Chagas was significantly correlated with overall bug density. Our data suggest that the application of environmental management measures by the affected people, such as plastering of walls and modification of roofs, coupled with keeping dogs away from bedrooms and application of insecticides, should limit the domestic population density of T. infestans and thus reduce the transmission of T. cruzi to people.  相似文献

Transaminase, amylase and protease activities were comparedin seedlings of normal and Opaque-2 (o2) maize. Transaminaseactivity, greater in normal maize, was highest in the scutellumfrom which it decreased rapidly in activity from day 2 afterimbibition; only low activity was observed in endosperm andaxis tissue. Amylolytic activity, optimal around pH 5, was greater in normalmaize at all stages of endosperm degradation. Activity whichwas low on day 2, rose to a peak on day 6 and declined afterwards.The level of free sugars in the endosperm of normal was higherthan in o2 maize, and in both varieties was highly correlatedwith amylolysis. Protease activity, optimal at pH 3.6, was also greater in normalendosperms and increased up to day 6 and activity was maintainedat this level until around day 14. Although the activity ofall three enzyme systems examined was greater in normal maizethere were no apparent differences in the overall growth ofnormal and o2 seedlings during this period. SYNOPSIS. An electron microscope study of Colpoda maupasi Enriques, isolated from the intestine of the blue-tongued skink Tiliqua nigrolutea, showed that the fine structure of this ciliate is similar in all respects to that of free-living ciliates. The correspondence applies particularly to the structure, distribution and number of mitochondria. This organelle has a rich intramitochondrial structure in the form of microvilli; it is found close to the periphery, near the nuclear apparatus and in other parts of the cytoplasm. It was concluded that the association between Colpoda maupasi and Tiliqua nigrolutea was probably accidental and limited to the cyst stage. Thus electron microscopy confirmed a conclusion arrived at by light microscopy. The presence of numerous food vacuoles made it possible to study stages of digestion within this organelle. Four major types of food vacuole were distinguished. Type 1 food vacuoles are characterized by their large size, the presence of intact bacteria and abundance of water. In type 2 the food vacuole is deprived of water, the bacteria are pressed together and the nuclei have lost their structure. Type 3 food vacuoles contain only bacterial ghosts, cytoplasmic and nuclear material having been digested. Food vacuoles of this type are found only occasionally, suggesting their short duration. It is of interest that during this transient stage the bulk of digestion takes place. In type 4 nothing reminiscent of bacteria is found; there are only myelin figures and vesicles of different sizes. Evaginations and invagnations of the vacuolar membrane and vesicles of different size and structure inside and outside the food vacuoles of types 1, 3 and 4 suggest that extensive communication exists between the cytoplasm and the food vacuole. It seems likely that enzymes are delivered to the food vacuole and digested materials are released from the food vacuole to the cytoplasm.  相似文献

The aim of the present study was to investigate possible changes of inositol 1,4,5-trisphosphate (IP3) mass in Torpedo cholinergic synaptosomes in conditions promoting stimulated acetylcholine (ACh) release. For this purpose, we used a radioreceptor IP3 mass assay and a chemiluminescent method for ACh detection. Torpedo cholinergic synaptosomes have consistent IP3 mass levels under resting conditions. The IP3 mass was neither modified by changes in external Ca2+ nor by a Ca2+-free medium containing EGTA. IP3 mass and ACh release, measured in the same conditions and in parallel, were increased by depolarization with high K+ and by the ionophores A-23187 and gramicidin-D in a manner dependent on external Ca2+ emphasizing that Ca2+ entry, independently of the influx mechanism involved, leads to an IP3 increase. The phospholipase Cβ inhibitors U-73122 and U-73343 reduced K+-stimulated IP3 levels while K+-evoked ACh release was almost completely blocked suggesting an additional effect of these drugs on depolarization-neurotransmitter secretion coupling. The effect reported showing an increase of IP3 by agents that stimulate ACh release may suggest a possible link between IP3 metabolism and the neurotransmitter release mechanism.
