Wheat plants (Triticum aestivum L., cv. Warigal) were subjectedto 20 d of water deficit during the period of endosperm celldivision. Drought accentuated the differences in final grainweight between spikelets and between grains within spikelets.The distal grains of top spikelets were most affected by drought.The maximum number of endosperm cells was, respectively, 30and 40 per cent lower in basal grains and distal grains of draughtedplants. In basal grains of middle spikelets, the number of largestarch granules per cell was unaffected but the number of smallstarch granules per cell was 45 per cent lower in grains ofdraughted plants. The initiation of small starch granules wasmore affected than cell division because severe water deficitoccurred earlier during the former process than the latter.Final dry weight appeared to correlate well with the maximumnumber of endosperm cells, but depended also on the number ofstarch granules per cell. Consequently, the amount of dry matterper cell was not constant in both treatments. The concentration of sucrose per endosperm cell was lower onlyin the droughted distal grains of top spikelets. The Swahili of Mombasa are Muslims, part of an ethnic group whoseforebears founded East Africas pre-colonial cities. Their cosmopolitanculture, in common with the cultures of some other, mainly Muslim,groups elsewhere includes a system of beliefs concerning the bodysfunctioning and illness as the result of relations among the bodysfour humours. The Swahili version of this system and its use in curingis described and briefly compared to several others. Despite theavailability of a variety of other systems, including the biomedicalemployed in the much patronized cost free treatment at governmenthospitals, the humoural system of beliefs is almost universallyheld. An hypothesis seeking to explain this suggests that becausethe Swahili schema is conceptualized by what Lakoff calls ametaphorical model emphasizingbalance which is also used to conceptualize the values applying tokey social relationships, the two schemata support one another,presumably by making the knowledge and emotions applying to oneavailable in the other.  相似文献

THE immune response in dissociated lymphoid cell cultures offers an opportunity to investigate the interaction of antigen with the surface receptors of immunocompetent cells. Using polymerized flagellin of Salmonella adelaide (POL), evidence was obtained that in vitro processes as different as immunity and tolerance both depend on the direct interaction between antigen and antigen-sensitive cells1–4. The use of chemically defined determinants in place of natural antigens could simplify the study of the molecular mechanisms underlying immunity and tolerance. Systems used in the past to induce immunity to defined determinants in vitro involved either a particulate antigen5 or spleen fragment cultures6 and were therefore unsuitable for the detailed study of the interactions occurring on the surface of lymphoid cells. A new system had to be devised. Here I describe the induction of a primary immune response to a hapten–protein conjugate in dissociated spleen cell cultures and the immune tolerance to a chemically defined determinant in vitro.  相似文献

THE induction of blast transformation by incubating lymphocytes with anti-immunoglobulin1 and anti-allotype2 sera has suggested that these cells have immunoglobulin on their surface. This hypothesis was directly verified by the demonstration of immunoglobulin on living mouse lymphoid cells by Raff et al.3. There is much evidence to indicate that immunocompetent cells have surface receptors for antigen. This idea is based on the finding that lymphocytes can bind radioactively labelled antigen to their surface4,5 and that specific immune unresponsiveness occurs if lymphoid cells are exposed to either highly radioactive antigen6 or haptens capable of forming covalent bonds with proteins7,8. The immunoglobulin nature of these antigen receptors is suggested by recent work showing that the binding of radioactively labelled antigen can be blocked by anti-immunoglobulin sera5,9. Reports that the adoptive immune response of mouse spleen cells can be inhibited by anti-mouse immunoglobulin sera (AMS)9,10 suggest that the interaction of antigen with the immunoglobulin receptor sites is a crucial step in the induction of the antibody response. We report here that the inhibitory action of AMS on the immune response is potentially reversible and that the induction of immune tolerance to polymerized flagellin (POL) in vitro may be blocked in the presence of AMS.  相似文献

Although many bone adaptation theories have been formulated to address both trabecular and cortical adaptation, most applications have focused on trabecular adaptation. Thus far, no thorough investigations of the influence of different types of loading on predicted patterns of long bone cross-sectional adaptation have been reported. In the current study, we present a new model for long bone cross-sectional adaptation that incorporates axial, bending and torsional loading components. We found that bending moments have a strong potential to modulate cross-sectional geometry, but can produce unforseen (and unrealistic) geometric instabilities. Torsional moments have the ability to suppress these instabilities, suggesting that torsion may play a more significant role in guiding long bone development than previously recognized. Our results also call into question the concept of strict "remodeling equilibrium," suggesting that long bones do not necessarily approach a state of uniform mechanical stimulation. This modeling approach provides an additional perspective on experimental studies, and may lead to a greater understanding of the interaction between mechanics and biology in long bone adaptation.  相似文献

We developed an alternate method for density-based load estimation and applied it to estimate hip joint load distributions for two femora. Two-dimensional finite element models were constructed from single energy quantitative computed tomography (QCT) data. Load estimation was performed using five loading regions on the femoral head. Within each loading region, individual nodal loads, normal to the local surface, were supplied as input to the load estimation. An optimization procedure independently adjusted individual nodal load magnitudes in each region, and the magnitudes of muscle forces on the greater trochanter, such that the applied tissue stimulus approached the reference stimulus throughout the model. Dominant estimated load resultant directions were generally consistent with published experimental data for loads during gait. The estimated loads also suggested that loads near the extremes of the articulating surface may be important (even required) for development and maintenance of normal bone architecture. Estimated load distributions within nearly all regions predicted bicentric loading patterns, which are consistent with observations of hip joint incongruity. Remodeling simulations with the estimated loads predicted density distributions with features qualitatively similar to the QCT data sets. This study illustrates how applications of density-based bone load estimation can improve understanding of dominant loading patterns in other bones and joints. The prediction of bicentric loading suggests a very fine level of local adaptation to details of joint loading.  相似文献

Climate increases regional tree-growth variability in Iberian pine forests   总被引:3,自引:0,他引:3  
Tree populations located at the geographical distribution limit of the species may provide valuable information about tree‐growth response to changes on climatic conditions. We established nine Pinus nigra, 12 P. sylvestris and 17 P. uncinata tree‐ring width chronologies along the eastern and northern Iberian Peninsula, where these species are found at the edge of their natural range. Tree‐growth variability was analyzed using principal component analysis (PCA) for the period 1885–1992. Despite the diversity of species, habitats and climatic regimes, a common macroclimatic signal expressed by the first principal component (PC1) was found. Moreover, considering the PC1 scores as a regional chronology, significant relations were established with Spanish meteorological data. The shared variance held by the tree chronologies, the frequency of narrow rings and the interannual growth variability (sensitivity) increased markedly during the studied period. This shows an enhancement of growth synchrony among forests indicating that climate might have become more limiting to growth. Noticeably, an upward abrupt shift in common variability at the end of the first half of the 20th century was detected. On the other hand, moving‐interval response functions showed a change in the growth–climate relationships during the same period. The relationship between growth and late summer/autumn temperatures of the year before growth (August–September, negative correlation, and November, positive correlation) became stronger. Hence, water stress increase during late summer previous to tree growth could be linked to the larger growth synchrony among sites, suggesting that climate was driving the growth pattern changes. This agrees with the upward trend in temperature observed in these months. Moreover, the higher occurrence of extreme years and the sensitivity increase in the second half of the 20th century were in agreement with an increment in precipitation variability during the growing period. Precipitation variability was positively related to tree‐growth variability, but negatively to radial growth. In conclusion, a change in tree‐growth pattern and in the climatic response of the studied forests was detected since the mid‐20th century and linked to an increase in water stress. These temporal trends were in agreement with the observed increase in warmer conditions and in precipitation variability.  相似文献

1. Generalist predatory insects that exploit unpredictable and ephemeral prey should allocate metabolic resources differently to soma and gonads than specialist species. As the former have more opportunities to encounter a wide array of prey than specialists, the expectation is that they will more rapidly resorb oocytes when food is scarce. By doing so, they reallocate resources to the soma to support the search for oviposition sites of a better quality. Similarly, they are expected to resume oogenesis faster than the specialist when good conditions return. 2. This hypothesis was tested by comparing a generalist and a specialist ladybird species belonging to the same genus. 3. A resorption index was calculated for females of both species subjected to several starvation regimes. This index indicated that over a period of fasting of 3 days, the intensity of resorption was greater in the generalist than the specialist. When food was again supplied, oogenesis resumed and within 1 day was faster in the generalist than in the specialist. 4. As predicted, the resorption of oocytes and replenishment occurred faster in the generalist than in the specialist species. This is the first time, to our knowledge, that the speed and intensity of the ovarian dynamics of a predatory insect have been linked to its way of life.  相似文献
