Wild type Crithidia fasciculata and three drug-resistant mutant strains that have shown “flagellar adherence” were studied as to their ability to agglutinate with lectins specific for receptor molecules containing N-acetyl glucosamine, N-acetyl galactosamine, galactose, mannose-like residues, fucose, and sialic acid. Escherichia coli with mannose-sensitive fimbriae was also used as an agglutination probe. The presence of D-GalNAc, D-Gal, and mannose-like residues was detected in the wild strain. Generally, in the mutants, residues of these sugar units were present in greater concentrations when compared to the wild type strain. β-Galactosidase treatment showed that β-D-Galp units are exposed on the cell membrane. All types of cell agglutination including flagellum-flagellum (F-F), flagellum-soma (F-S), and soma-soma (S-S) were observed when lectins were used; however, with E. coli only the F-F type of cell agglutination was observed with the wild type strain and the TFRR1 mutant. ABSTRACT. The energy metabolism was compared among four different representatives of the genus Phytomonas isolated from different plants and localities: the sieve tubes of the hartrot-infected coconut palm in French Guyana, the latex fluid of Euphorbia hyssopifolia in French Guyana and the fruits of tomato and cherimoya in Spain. All four isolates produced acetate, ethanol, glycerol and glycine as metabolic end-products. In addition, small amount of succinate and pyruvate were excreted. Only minor quantitative differences were observed in the four isolates. Glycosomes, harboring the glycolytic enzymes, were present in all isolates. No evidence was found for an active involvement of the mitochondrion in metabolism. Respiration was insensitive to the classical inhibitors of the respiratory chain, such as antimycin and potassium cyanide, but inhibited by salicylhydroxamic acid. No evidence was found for the functioning of a citric-acid cycle. It is concluded that representative of this genus share the same highly active carbohydrate metabolism combined with a complete suppression of mitochondrial activity. 相似文献

Blue‐tailed skinks (genus Plestiodon) are a common component of the terrestrial herpetofauna throughout their range in eastern Eurasia and North and Middle America. Plestiodon species are also frequent subjects of ecological and evolutionary research, yet a comprehensive, well‐supported phylogenetic framework does not yet exist for this genus. We construct a comprehensive molecular phylogeny of Plestiodon using Bayesian phylogenetic analyses of a nine‐locus data set comprising 8308 base pairs of DNA, sampled from 38 of the 43 species in the genus. We evaluate potential gene tree/species tree discordance by conducting phylogenetic analyses of the concatenated and individual locus data sets, as well as employing coalescent‐based methods. Specifically, we address the placement of Plestiodon within the evolutionary tree of Scincidae, as well as the phylogenetic relationships between Plestiodon species, and their taxonomy. Given our sampling of major Scincidae lineages, we also re‐evaluate ‘deep’ relationships within the family, with the goal of resolving relationships that have been ambiguous in recent molecular phylogenetic analyses. We infer strong support for several scincid relationships, including a major clade of ‘scincines’ and the inter‐relationships of major Mediterranean and southern African genera. Although we could not estimate the precise phylogenetic affinities of Plestiodon with statistically significant support, we nonetheless infer significant support for its inclusion in a large ‘scincine’ clade exclusive of Acontinae, Lygosominae, Brachymeles, and Ophiomorus. Plestiodon comprises three major geographically cohesive clades. One of these clades is composed of mostly large‐bodied species inhabiting northern Indochina, south‐eastern China (including Taiwan), and the southern Ryukyu Islands of Japan. The second clade comprises species inhabiting central China (including Taiwan) and the entire Japanese archipelago. The third clade exclusively inhabits North and Middle America and the island of Bermuda. A vast majority of interspecific relationships are strongly supported in the concatenated data analysis, but there is nonetheless significant conflict amongst the individual gene trees. Coalescent‐based gene tree/species tree analyses indicate that incongruence amongst the nuclear loci may severely obscure the phylogenetic inter‐relationships of the primarily small‐bodied Plestiodon species that inhabit the central Mexican highlands. These same analyses do support the sister relationship between Plestiodon marginatus Hallowell, 1861 and Plestiodon stimpsonii (Thompson, 1912), and differ with the mitochondrial DNA analysis that supports Plestiodon elegans (Boulenger, 1887) + P. stimpsonii. In a previous study, important acclimation to water stress was observed in the Ramellet tomato cultivar (TR) from the Balearic Islands, related to an increase in the water‐use efficiency through modifications in both stomatal (gs) and mesophyll conductances (gm). In the present work, the comparison of physiological and morphological traits between TR accessions grown with and without water stress confirmed that variability in the photosynthetic capacity was mostly explained by differences in the diffusion of CO2 through stomata and leaf mesophyll. Maximization of gm under both treatments was mainly achieved through adjustments in the mesophyll thickness and porosity and the surface area of chloroplasts exposed to intercellular airspace (Sc). In addition, the lower gm/Sc ratio for a given porosity in drought‐acclimated plants suggests that the decrease in gm was due to an increased cell wall thickness. Stomatal conductance was also affected by drought‐associated changes in the morphological properties of stomata, in an accession and treatment‐dependent manner. The results confirm the presence of advantageous physiological traits in the response to drought stress in Mediterranean accessions of tomato, and relate them to particular changes in the leaf anatomical properties, suggesting specific adaptive processes operating at the leaf anatomical level. 相似文献

Whole‐genome duplication, or polyploidy, is common in many plant species and often leads to better adaptation to adverse environmental condition. However, little is known about the physiological and molecular determinants underlying adaptation. We examined the drought tolerance in diploid (2x) and autotetraploid (4x) clones of Rangpur lime (Citrus limonia) rootstocks grafted with 2x Valencia Delta sweet orange (Citrus sinensis) scions, named V/2xRL and V/4xRL, respectively. Physiological experiments to study root–shoot communication associated with gene expression studies in roots and leaves were performed. V/4xRL was much more tolerant to water deficit than V/2xRL. Gene expression analysis in leaves and roots showed that more genes related to the response to water stress were differentially expressed in V/2xRL than in V/4xRL. Prior to the stress, when comparing V/4xRL to V/2xRL, V/4xRL leaves had lower stomatal conductance and greater abscisic acid (ABA) content. In roots, ABA content was higher in V/4xRL and was associated to a greater expression of drought responsive genes, including CsNCED1, a pivotal regulatory gene of ABA biosynthesis. We conclude that tetraploidy modifies the expression of genes in Rangpur lime citrus roots to regulate long‐distance ABA signalling and adaptation to stress. 相似文献

Climate warming is known to have effects on population dynamics through variations in survival, fecundity and density. However, the impacts of climate change on population composition are still poorly documented. Morphotypes are powerful markers to track changes in population composition. In the common lizard, Lacerta vivipara , individuals display two types of dorsal patterns: reticulated (R individuals) and linear (L individuals). We examined how local warming affected intrapopulation frequencies of these morphotypes across 11 years. We observed changes in morph frequency of dorsal patterns across years, paralleling the rise of spring temperatures. The proportion of R individuals increased with June temperatures in juveniles, yearlings, and adult males and females. Three mechanisms could explain these changes: phenotypic plasticity, microevolution and/or dispersal between populations. We investigated the ontogenetic determinism, fitness and recruitment rates associated with dorsal morphotypes. Dorsal pattern ontogeny showed temperature dependence but this relationship was not associated with the warming trend during this study. We found variation by morphotype in survival and clutch size, but these factors did not explain R frequency increases. Among all the parameters considered in this study, only a decrease of immigration, which was more pronounced in the L morphotype, could explain the change in population composition. To our knowledge, this provides the first evidence of the impact of climate warming on population composition due to its effects on immigration. 相似文献

Parasite pressure and nutrition are two of the most important factors affecting the trade-off between nestling growth and immune development. During development, energy and nutrients are often limited, and nestlings should only dedicate differentially more valuable resources to their immune system when the associated benefits are high (i.e. in situations of an increased risk of parasitism). In this study, we manipulated nutritional condition of Eurasian Magpie Pica pica nestlings by providing a food supplement. Additionally, the study area was subdivided into two categories of habitat (irrigated and arid) based on the presence of irrigation canals. Nestling diet composition was more varied in the irrigated compared with in the arid habitat. In addition, nestlings of the irrigated habitat showed a significantly higher infestation of both ectoparasites and blood parasites and a significantly higher cell-mediated immunity, but lower tarsus length compared with nestlings of the arid habitat. Food supplementation to nestlings did not affect tarsus length, but increased nestling cell-mediated immunity in the arid habitat only. Based on the recent demonstrated trade-off between growth and immunocompetence in nestlings, we suggest that differences between habitats in nestling diet and parasite prevalence may have caused different priority rules in the allocation of resources between both fitness traits. 相似文献

A pooled DNA method was used to produce fully informative EST (expressed sequence tag)‐derived markers for the Picea genus. Nine markers were produced from 10 cDNA identified as candidates for cold tolerance or embryogenesis. Indels and SNPs (single nucleotide polymorphisms) were characterized from sequences obtained from pools of 10 individuals for each of the three species: Picea glauca (white spruce), Picea mariana (black spruce) and Picea abies (Norway spruce). Indels were present in 28% of the sequences and SNPs with a frequency greater than 10% were present on average in 1.2% of the positions. 相似文献
