A human melanoma-associated antigen immunogenic in patients was recently identified by screening an expression cDNA library constructed from cultured human melanoma cell line with sera from patients with melanoma. The nucleic acid sequence of the cloned D-1 cDNA has no significant homology with previously reported mammalian genes. The cDNA D-1 encodes a peptide of about 37 kDa, which showed fivefold higher reactivity with sera from patients with melanoma than with sera from normal donors. In order to detect D-1 gene expression in vivo, in-situ hybridization and immunostaining with cRNA probe and murine anti-D-1 sera were carried out on surgically removed tissues. Digoxigenin-labeled cRNA D-1 was exclusively hybridized with mRNA in the cytoplasm of melanoma cells but not with keratinocytes and fibroblasts adjacent to melanoma nests. Polyclonal anti-D-1 antibodies were obtained by immunization of Balb/c mice with recombinant D-1 peptide and clearly reacted with melanoma cells but not with keratinocytes and fibroblasts, similar to the results of in-situ hybridization. The above information will help to assess the suitability of recombinant D-1 peptide to implement active specific immunotherapy in patients with melanoma.  相似文献

Outbreaks of paralytic shellfish poisoning caused by the toxic dinoflagellate Alexandrium minutum (Dinophyceae) are a worldwide concern from both the economic and human health points of view. For population genetic studies of A. minutum distribution and dispersal, highly polymorphic genetic markers are of great value. We isolated 12 polymorphic microsatellites from this cosmopolitan, toxic dinoflagellate species. These loci provide one class of highly variable genetic markers, as the number of alleles ranged from four to 12, and the estimate of gene diversity was from 0.560 to 0.862 across the 12 microsatellites; these loci have the potential to reveal genetic structure and gene flow among A. minutum populations.  相似文献

Flowering phenology and allozyme variation were studied to test the existence of positive assortative mating for flowering time in a natural population of Primula sieboldii E. Morren, a heterostylous perennial herb, consisting of approximately 180 genets in a deciduous forest. There was significant variation in flowering date among genets, but not between heterostylous morphs. The temporal order of the flowering time of genets was fairly constant for the two years of the study. The spatial heterogeneity of light availability at the study site was small during the flowering season of the species. In order to analyze the extent of genetic differentiation between early- and late-flowering genet groups, allozyme diversities were analyzed with 10 loci. The GST between the early- and late-flowering groups was not significantly different from zero. Evidence of positive assortative mating for flowering time was not detected. Prolongation of flowering duration due to pollen limitation may be one important factor preventing the genetic differentiation of early- and late-flowering groups by enhancing the overlap of flowering time among genets.  相似文献

  1. Effect of light on ion absorption and resting potential of theinternodal cell of Nitella flexilis was investigated under variousconditions.
  2. On illumination, the resting potential increasedby about 30mVin 10–4 M KCl and by about 60 mV in 10–4M NaClsolution. A similar photoelectric response was also observedin 10–3 M KCl, 10–2 M CaCl2 and 5 x 10–2 MCaCl2 solutions, but not at all in 10–2 M KCl solution.
  3. Absorption of ions by the cell took place in parallel withthelight-induced change in resting potential.
  4. Red and bluelights were very effective in increasing the restingpotential,while green light was almost ineffective. These differenteffectsof color lights were in good agreement with their effectsinincreasing the osmotic value of the cell.
  5. The photoelectricresponse was not affected by phenylurethane,which, on the otherhand, strongly inhibited the light-inducedion absorption.
  6. Theuptake of ions by the cell from the external medium intothevacuole is assumed to proceed in two different steps: thefirstis the process involving the ion movements across theoutermostplasmalemma, and the second is that involved in thetransportof ions through the cytoplasmic layer and tonoplast.The formerprocess is considered to be influenced by the increasein restingpotential probably caused by the light absorbed bychlorophyll.The process was, however, suggested to be independentof photosynthesis.On the other hand, the latter process issupposed to be relatedto photosynthesis. We isolated 15 polymorphic microsatellites from one of the most noxious red tide‐causing dinoflagellate species, Heterocapsa circularisquama. These loci provide one class of highly variable genetic markers, as the number of alleles ranged from two to six, and the estimate of gene diversity from 0.205 to 0.684 across the 15 microsatellites. These loci have the potential to reveal genetic structure and gene flow among H. circularisquama populations.  相似文献
