A 450-kDa human epidermal autoantigen was originally identified as a protein that reacted with the serum from an individual with a subepidermal blistering disease. Molecular cloning of this protein has now shown that it contains 5065 amino acids and has a molecular mass of 552 kDa. As reported previously this protein, which we call epiplakin, belongs to the plakin family, but it has some very unusual features. Epiplakin has 13 domains that are homologous to the B domain in the COOH-terminal region of desmoplakin. The last five of these B domains, together with their associated linker regions, are particularly strongly conserved. However, epiplakin lacks a coiled-coil rod domain and an amino-terminal domain, both of which are found in all other known members of the plakin family. Furthermore, no dimerization motif was found in the sequence. Thus, it is likely that epiplakin exists in vivo as a single-chain structure. Epitope mapping experiments showed that the original patient's serum recognized a sequence unique to epiplakin, which was not found in plectin. Immunofluorescence staining revealed the presence of epiplakin in whole sheets of epidermis and esophagus, in glandular cells of eccrine sweat and parotid glands and in mucous epithelial cells in the stomach and colon.  相似文献

Grazing by the planktonic phytoflagellate, Pyramimonas gelidicola McFadden (Chlorophyta: Prasinophyta), and heterotrophic nanoflagellates (HNAN) in meromictic saline Ace Lake in the Vestfold Hills, eastern Antarctica was investigated in the austral summers of 1997 and 1999. Up to 47% of the P. gelidicola population ingested fluorescently labeled prey (FLP). Ingestion rates varied with depth. In January 1997 and November 1999, maximum P. gelidicola ingestion rates of 6.95 and 0.79 FLP·cell?1·h?1, respectively, were measured at the chemocline (6–8 m) where a deep chl maximum composed of phototrophic nanoflagellates (PNAN DCM), predominantly P. gelidicola, persisted all year. During the summers of 1997 and 1999, the grazing P. gelidicola community removed between 0.4% and approximately 16% of in situ bacterial biomass, equivalent to between 4% and>100% of in situ bacterial production. Because of their higher abundance, the community clearance rates of HNAN in Ace Lake generally exceeded those of P. gelidicola, but HNAN removed approximately only 3%–4% of bacterial biomass, equivalent to between 28% and 32% of bacterial production. Pyramimonas gelidicola growth rates were highest at the PNAN DCM concomitant with the highest ingestion rates. It is estimated that during the summer P. gelidicola can derive up to 30% of their daily carbon requirements from bacterivory at the PNAN DCM. This study confirms mixotrophy as an important strategy by which planktonic organisms can survive in extreme, polar, lacustrine ecosystems.  相似文献

Prototype inhibitors of protein arginine methyltransferases (PRMTs) have been constructed by attaching guanidine functionality via a variable linker to non-reactive amine analogues of the cellular co-factor (S)-adenosyl methionine (AdoMet). Potent inhibition of PRMT1 (IC50 of ~3–6 μM) combined with weak inhibition of the lysine methyltransferase SET7 (~50% of activity at 100 μM) was observed for two such compounds.  相似文献

Twelve species of microalgae, isolated from north Australian marine, freshwater and hypersaline environments, were grown under controlled conditions of temperature, pH, photon flux density and salinity, and analysed for ash, total protein, water soluble carbohydrates, chlorophylla, total lipids, total fatty acids and fatty acid composition. Highest levels of the polyunsaturated fatty acid eicosapentaenoic acid [20:5(n-3)] were found in the marine diatoms.Nitzschia (frustulum) andN. closterium (23.1% and 15.2% of total fatty acids, respectively). None of the species studied had levels of docosahexaenoic acid [22:6(n-3)] greater than 1.1 % of total fatty acids. None of the chlorophyte species contained significant levels of either 20:5(n-3) or 22:6(n-3). The highest total fatty acid concentration of all species in the study was found in the freshwater chlorophyte speciesScenedesmus dimorphus (105 mg g–1 dry wt). The hypersaline speciesDunaliella salina had the highest total lipid content (28.1% dry wt), followed byN. closterium, N. (frustulum) andNavicula sp. (24.2–27.8% dry wt).Chlamydomonas sp. had the highest protein content (66.9% dry wt).N. The "circle analysis" method of Williams (1969) and a new improved method employing hypothetical grains described in the previous paper have been used to analyze the distribution of autoradiographic grains over erythroid bone marrow cells labeled with radioactive iron, 55Fe. The resolution obtainable with this isotope was determined by measuring the distribution of grains about a thin line source. This distribution was also used in calculation of the circle size for the Williams's analysis and the distances of hypothetical grains for the new method. The new method provides estimates for the amount of activity in the regions of condensed and extended nuclear chromatin and for the concentration of isotope at the junction between these two areas. The possible significance of activity in this junctional region is discussed.  相似文献

The total protein, carbohydrate, lipid and ash compositions, and fatty acid contents of two species of marine microalgae, the eustigmatophyte Nannochloropsis oculata (formerly ‘Chlorella sp., Japan’) and the chrysophyte Isochrysis sp. (Tahitian) used in tropical Australian mariculture, were studied. The microalgae were grown under a range of culture conditions (41 and 601 laboratory culture, 3001 bag culture, and 80001 outdoor culture) and four light regimes (100 to 107 μ E m−2 s−1, 240 to 390 μ E m−2 s−1, 340 to 620 μ E m−2 s−1, and 1100 to 1200 μE m−2 s−1 respectively) to determine the effect of light intensity on the chemical composition of large scale outdoor cultures. Laboratory and bag cultures were axenic and cultured in Walne medium while outdoor cultures were grown in a commercial medium designed for optimum nutrition in tropical outdoor aquaculture operations. Change in growth medium and photon flux density produced only small changes in the proximate biochemical composition of both algae. N. oculata and Isochrysis sp. both showed a trend towards slightly lower carbohydrate and higher chlorophyll a in shaded outdoor culture. Isochrysis sp. showed significant concentrations of the essential polyunsaturated fatty acid 22:6(n−3) (docosahexaenoic acid) from 5.3 to 10.3% of total fatty acid, and 20:5(n−3) (eicosapentaenoic acid) ranged from 0.6 to 4.1%. In contrast, N. oculata had high concentrations of 20:5(n−3) (17.8 to 39.9%) and only traces of 22:6(n−3). The fatty acid composition of Isochrysis sp. grown at high photon flux density (1100–1200 μE m−2 s−1) under outdoor culture showed a decrease in the percentage of several highly unsaturated fatty acids, including 20:5(n−3), and an increase in 22:6(n−3). N. oculata showed a similar decrease in the percentage of 20:5(n−3). High light intensity caused a decrease in the ratio of total C16 unsaturated fatty acids to saturated 16:0 in N. oculata, and a decrease in the ratio of total C18 unsaturated fatty acids to saturated 18:0 together with a decrease in the ratio of total unsaturated fatty acids to total saturated fatty acids in both microalgae.  相似文献

Snake venom serine proteinases, which belong to the subfamily of trypsin-like serine proteinases, exhibit a high degree of sequence identity (60-66%). Their stringent macromolecular substrate specificity contrasts with that of the less specific enzyme trypsin. One of them, the plasminogen activator from Trimeresurus stejnegeri venom (TSV-PA), which shares 63% sequence identity with batroxobin, a fibrinogen clotting enzyme from Bothrops atrox venom, specifically activates plasminogen to plasmin like tissue-type plasminogen activator (t-PA), even though it exhibits only 23% sequence identity with t-PA. This study shows that TSV-PA, t-PA, and batroxobin are quite different in their specificity toward small chromogenic substrates, TSV-PA being less selective than t-PA, and batroxobin not being efficient at all. The specificity of TSV-PA, with respect to t-PA and batroxobin, was investigated further by site-directed mutagenesis in the 189-195 segment, which forms the basement of the S(1) pocket of TSV-PA and presents a His at position 192 and a unique Phe at position 193. This study demonstrates that Phe(193) plays a more significant role than His(192) in determining substrate specificity and inhibition resistance. Interestingly, the TSV-PA variant F193G possesses a 8-9-fold increased activity for plasminogen and becomes sensitive to bovine pancreatic trypsin inhibitor.  相似文献
