Globally, over 7.4 million accessions of crop seeds are stored in gene banks, and conservation of genotypic variation is pivotal for breeding. We combined genetic and biochemical approaches to obtain a broad overview of factors that influence seed storability and ageing in barley (Hordeum vulgare). Seeds from a germplasm collection of 175 genotypes from four continents grown in field plots with different nutrient supply were subjected to two artificial ageing regimes. Genome‐wide association mapping revealed 107 marker trait associations, and hence, genotypic effects on seed ageing. Abiotic and biotic stresses were found to affect seed longevity. To address aspects of abiotic, including oxidative, stress, two major antioxidant groups were analysed. No correlation was found between seed deterioration and the lipid‐soluble tocochromanols, nor with oil, starch and protein contents. Conversely, the water‐soluble glutathione and related thiols were converted to disulphides, indicating a strong shift towards more oxidizing intracellular conditions, in seeds subjected to long‐term dry storage at two temperatures or to two artificial ageing treatments. The data suggest that intracellular pH and (bio)chemical processes leading to seed deterioration were influenced by the type of ageing or storage. Moreover, seed response to ageing or storage treatment appears to be significantly influenced by both maternal environment and genetic background.  相似文献

Aedes aegypti (L.) (Diptera: Culicidae), the main vector of yellow fever and dengue viruses, was eradicated from Argentina between 1955 and 1963, but reinvaded the country in 1986. In Uruguay, the species was reintroduced in 1997. In this study we used highly polymorphic inter‐simple sequence repeats (ISSR) markers to analyse the genetic structure of Ae. aegypti populations from Uruguay and northeastern Argentina to identify possible colonization patterns of the vector. Overall genetic differentiation among populations was high (FST = 0.106) and showed no correlation with geographic distance, which is consistent with the short time since the reintroduction of the species in the area. Differentiation between pairs of Argentine populations (FST 0.072 to 0.221) was on average higher than between Uruguayan populations (FST?0.044 to 0.116). Bayesian estimation of population structure defined four genetic clusters and most populations were admixtures of two of them: Mercedes and Treinta y Tres (Uruguay) were mixtures of clusters 1 and 3; Salto (Uruguay) and Paraná (Argentina) of clusters 1 and 4; Fray Bentos (Uruguay) of clusters 2 and 3, and Gualeguaychú (Argentina) of clusters 2 and 3. Posadas and Buenos Aires in Argentina were fairly genetically homogeneous. Our results suggest that Ae. aegypti recolonized Uruguay from bordering cities in Argentina via bridges over the Uruguay River and also from Brazil.  相似文献

ABSTRACT. Euglena gracilis is a freshwater free‐living organism able to grow with ethanol as carbon source; to facilitate this metabolism several alcohol dehydrogenase (ADH) activities have been detected. We report the gene cloning, over‐expression, and biochemical characterization of a medium‐chain NAD+‐dependent ADH from E. gracilis (EgADH). The enzyme's amino acid sequence displayed the highest percentages of similarity and identity with ADHs of bacteria and fungi. In the predicted three‐dimensional model, all the residues involved in Zn2+, cofactor, and substrate binding were conserved. A conventional signal peptide for import into mitochondria could not be clearly identified. The protein of 37 kDa was over‐expressed, purified to homogeneity, and kinetically characterized. The enzyme's optimal pH was 7.0 for ethanol oxidation displaying a Vm of 11.7±3.6 U/mg protein and a Km of 3.2±0.7 mM for this substrate. Isopropanol and isopentanol were also utilized, although with less efficiency. It showed specificity for NAD+ with a Km value of 0.39±0.1 mM and Mg2+ or Zn2+ were essential for activity. The recombinant EgADH reported here may help to elucidate the roles that different ADHs have on the metabolism of short‐ and long‐chain alcohols in this microorganism.  相似文献

Twenty‐three isolates of the widely distributed marine bacteria Alteromonas macleodii have been analysed by multilocus sequence analysis combined with phylogenetic and multivariate statistical analyses. The strains originated from the Pacific Ocean, Mediterranean Sea, English Channel, Black Sea and Thailand. Using the nucleotide sequences of nine loci for each of the 23 isolates, a robust identification was achieved of different clades within the single species. Strains generally clustered with the depth in the water column from which the isolate originated. Strains also showed more recombination with isolates from the same vicinity, suggesting that genetic exchange plays a role in diversification of planktonic marine prokaryotes. This study thus shows for the first time for a large set of isolates of a species of planktonic marine prokaryotes that multilocus sequence analysis overcomes the problems associated with the analysis of individual marker genes or presence of extensive recombination events. It can thus achieve intraspecific identification to the level of genotypes and, by comparison with relevant environmental data, ecotypes.  相似文献

Abstract Mediterranean representatives of the galling aphid tribe Fordini (Hemiptera: Aphididae: Eriosomatinae) are usually grouped under the subtribe term Fordina. Aphids within Fordina display two‐year life cycles, alternating between Pistacia shrubs, where they induce conspicuous galls, and roots of Poaceae species. The high number of morphs present in a given species, the lack of knowledge of the complete cycle in some species, and the similarity between homologous morphs observed in different species pose many taxonomic problems in this group. We present results of a survey to elucidate the phylogenetic relationships among Fordini species present in the Iberian Peninsula and the Canary Islands. Sequences from the nuclear long‐wavelength opsin (LWO) and translation elongation factor 1α (EF‐1α) genes and from a region of the mitochondrial DNA containing the genes encoding the subunits six and eight of the F‐ATPase were used to infer relationships among 10 Fordina species, namely Smynthurodes betae, Aploneura lentisci, Baizongia pistaciae, two Geoica species (G. utricularia and G. setulosa), three species of Forda and two of Paracletus. Relationships between and within representatives of the genera Forda and Paracletus, both exemplifying taxonomic and ecological problems, were investigated in greater depth through extensive sampling and morphometric analysis. A total of eight, six and six samples from F. formicaria, F. marginata and F. riccobonii, respectively, were included in the survey, along with 40 and 4 samples classified initially as P. cimiciformis and P. donisthorpei, respectively. Our results are relevant to current studies on the evolution of host selection by aphids and on the evolution of gall morphology. Our phylogeny suggests that the group can be divided into two main clades. One clade is composed of aphids inducing small, low‐capacity galls on either P. atlantica or P. terebinthus (Smynthurodes betae and genera Forda and Paracletus). The second clade is composed of species inducing larger galls on P. lentiscus and P. terebinthus (Aploneura lentisci, Baizongia pistaciae and Geoica species). Our results suggest that revision of diagnostic characters used in the taxonomy of Paracletus is needed, and suggest Forda rotunda as a new synonym of Paracletus cimiciformis syn.n.  相似文献

The estimation of the carbon balance in ecosystems, regions, and the biosphere is currently one of the main concerns in the study of the ecology of global change. Current remote sensing methodologies for estimating gross primary productivity are not satisfactory because they rely too heavily on (i) the availability of climatic data, (ii) the definition of land‐use cover, and (iii) the assumptions of the effects of these two factors on the radiation‐use efficiency of vegetation (RUE). A new methodology is urgently needed that will actually assess RUE and overcome the problems associated with the capture of fluctuations in carbon absorption in space and over time. Remote sensing techniques such as the widely used reflectance vegetation indices (e.g. NDVI, EVI) allow green plant biomass and therefore plant photosynthetic capacity to be assessed. However, there are vegetation types, such as the Mediterranean forests, with a very low seasonality of these vegetation indices and a high seasonality of carbon uptake. In these cases it is important to detect how much of this capacity is actually realized, which is a much more challenging goal. The photochemical reflectance index (PRI) derived from freely available satellite information (MODIS sensor) presented for a 5‐year analysis for a Mediterranean forest a positive relationship with the RUE. Thus, we show that it is possible to estimate RUE and GPP in real time and therefore actual carbon uptake of Mediterranean forests at ecosystem level using the PRI. This conceptual and technological advancement would avoid the need to rely on the sometimes unreliable maximum RUE.  相似文献

Stem radial growth responds to environmental conditions, and has been widely used as a proxy to study long‐term patterns of tree growth and to assess the impact of environmental changes on growth patterns. In this study, we use a tree ring dataset from the Catalan Ecological and Forest Inventory to study the temporal variability of Scots pine (Pinus sylvestris L.) stem growth during the 20th century across a relatively large region (Catalonia, NE Spain) close to the southern limit of the distribution of the species. Basal area increment (BAI) was modelled as a function of tree size and environmental variables by means of mixed effects models. Our results showed an overall increase of 84% in Scots pine BAI during the 20th century, consistent with most previous studies for temperate forests. This trend was associated with increased atmospheric CO2 concentrations and, possibly, with a general increase in nutrient availability, and we interpreted it as a fertilization effect. Over the same time period, there was also a marked increase in temperature across the study region (0.19 °C per decade on average). This warming had a negative impact on radial growth, particularly at the drier sites, but its magnitude was not enough to counteract the fertilization effect. In fact, the substantial warming observed during the 20th century in the study area did not result in a clear pattern of increased summer drought stress because of the large variability in precipitation, which did not show any clear time trend. But the situation may change in the future if temperatures continue to rise and/or precipitation becomes scarcer. Such a change could potentially reverse the temporal trend in growth, particularly at the driest sites, and is suggested in our data by the relative constancy of radial growth after ca. 1975, coinciding with the warmer period. If this situation is representative of other relatively dry, temperate forests, the implications for the regional carbon balance would be substantial.  相似文献

The present study aimed to infer evolutionary scenarios for Vipera latastei and Vipera monticola in the Iberian Peninsula and the Maghreb through the identification of spatial patterns in morphological character variation and biogeographic patterns in morphological variability distribution. Ten morphological traits from 630 vipers were analysed with geostatistic and ecological niche modelling in a geographical information system. Interpolation by Kriging was used to generate surfaces of morphological variation, which were combined with spatial principal components analysis (SPCA). Putative morphological differentiated groups generated by SPCA maps were tested with discriminant function analysis (DFA). Maximum entropy modelling and nine environmental variables were used to identify factors limiting the distribution of groups and areas for their potential occurrence. Groups supported by DFA were: Western Iberia, Eastern Iberia, Rif plus Middle Atlas, Algeria, and High Atlas. Their distribution is influenced by common environmental factors such as precipitation. Areas of probable sympatry between Iberian groups matched the morphological clines observed by geostatistics tools. Geographic variation patterns in V. latastei‐monticola are probably due to vicariant separation of Iberian and African populations during the opening of the Strait of Gibraltar, and population refugia during the Quaternary glaciations with secondary contact. The taxonomic status of northern Morocco and Algerian groups should be further investigated. This study used pollen morphology to address taxonomic controversies related to several species belonging to subgenus Hebeclada of the genus Polygala (Polygalaceae). According to the last traditionally accepted revision of this subgenus, it comprises 40 species. Nevertheless, a recent taxonomic treatment applying more comprehensive criteria reduced this number to only nine species. Our work focused on 15 traditionally accepted species that occur in Brazil, fourteen of which have been considered as only six species in this recent taxonomic study. Pollen grains from floral buds at pre‐anthesis were collected from herbarium specimens and analysed using both visible light and scanning electron microscopy. Most of the traditionally accepted species were distinguished by different combinations of several pollen‐related features, in particular colpi number (13 or 15), pollen size and shape, and endoaperture type (endocingulate or not) and arrangement (parallel or sinuous).
