The nearly complete, PCR-amplified, 16S rRNA gene sequences have been determined from the representative type strains of eight xanthomonad phena, including six validly described species of the genus Xanthomonas and Stenotrophomonas maltophilia. Pairwise sequence comparisons and phylogenetic analysis demonstrated that the xanthomonads comprise a monophyletic lineage within the γ-subclass of the Proteobacteria. Although the genus Xanthomonas was observed to comprise a cluster of very closely related species, the observed species-specific primary sequence differences were confirmed through sequencing additional strains belonging to the respective species.  相似文献

Plasmodesmata (PD) are nano-sized membrane-lined channels controlling intercellular communication in plants. Although progress has been made in identifying PD proteins, the role played by major membrane constituents, such as the lipids, in defining specialized membrane domains in PD remains unknown. Through a rigorous isolation of “native” PD membrane fractions and comparative mass spectrometry-based analysis, we demonstrate that lipids are laterally segregated along the plasma membrane (PM) at the PD cell-to-cell junction in Arabidopsis thaliana. Remarkably, our results show that PD membranes display enrichment in sterols and sphingolipids with very long chain saturated fatty acids when compared with the bulk of the PM. Intriguingly, this lipid profile is reminiscent of detergent-insoluble membrane microdomains, although our approach is valuably detergent-free. Modulation of the overall sterol composition of young dividing cells reversibly impaired the PD localization of the glycosylphosphatidylinositol-anchored proteins Plasmodesmata Callose Binding 1 and the β-1,3-glucanase PdBG2 and altered callose-mediated PD permeability. Altogether, this study not only provides a comprehensive analysis of the lipid constituents of PD but also identifies a role for sterols in modulating cell-to-cell connectivity, possibly by establishing and maintaining the positional specificity of callose-modifying glycosylphosphatidylinositol proteins at PD. Our work emphasizes the importance of lipids in defining PD membranes.  相似文献

Epithelial tubes perform functions that are essential for the survival of multicellular organisms. Understanding how their polarised features are maintained is therefore crucial. By analysing the function of the clathrin adaptor AP-1 in the C. elegans intestine, we found that AP-1 is required for epithelial polarity maintenance. Depletion of AP-1 subunits does not affect epithelial polarity establishment or the formation of the intestinal lumen. However, the loss of AP-1 affects the polarised distribution of both apical and basolateral transmembrane proteins. Moreover, it triggers de novo formation of ectopic apical lumens between intestinal cells along the lateral membranes later during embryogenesis. We also found that AP-1 is specifically required for the apical localisation of the small GTPase CDC-42 and the polarity determinant PAR-6. Our results demonstrate that AP-1 controls an apical trafficking pathway required for the maintenance of epithelial polarity in vivo in a tubular epithelium.  相似文献

Phosphorus (P) is often a limiting factor of forest growth but our knowledge of the processes governing P availability in forest soils is rather limited. In the present work, we combined a isotopic dilution method with extraction methods to evaluate the P status in Pinus pinaster plantation forests on highly P-deficient soils. Total, organic, and inorganic P, dissolved and diffusive P, i.e. ionic P species that can be transferred from the solid phase to the soil solution due a gradient of concentration, were determined to a soil depth of 120 cm in a gradient of 18 forest sites (seven humid sites, five mesic sites, and six dry sites). Our objective was to assess the potential contribution of organic and inorganic P to plant available P. Based on results and our original assumptions, we observed that the contribution of organic P fractions (mineralization of soil organic P) to P availability related to the contribution of inorganic P fractions (diffusive P for durations up to 1 year) was predominant in litter, less important in top soil horizons, and negligible at depths below 30 cm. This was partly due to a decreasing proportion of organic P and an increasing proportion of diffusive P with soil depth. Owing to a very low amount of diffusive P in the top soils in dry sites, the relative contribution of organic P was actually higher in these sites than in humid and mesic sites, despite a lower overall organic P fraction. The combination of extraction and isotopic dilution methods in our study shed new light on P status in this forest range. In particular, these methods enable assessment of both the size of the pools and their dynamic fractions.  相似文献

The postgonopodial gland of the myriapod Glomeris marginata (Villers), which produces a pheromone, is an integumentary gland comprising numerous functional secretory units. Each secretory unit consists of two proximal secretory cells, an intermediary cell lacking secretory characteristics and a canal cell surrounding the canal, which is secretory in nature. Secretory proximal cells exhibit a zone of small channels originating from invaginations of the plasma membrane and through which secreted material is released. Apposing each invagination of these cells is a corresponding invagination of the intermediary cell: the two units in the centre of the intermediary cell join another which communicates with the canal. Secretion produced by the latter passes through the canal wall and blends with secretion of the two proximal cells. The most striking feature of all these cells is the abundance of tubules and fibrils in the small canal zone in the proximal cells, which also exhibits a centriole; in the intermediary cell around cytoplasmic membrane invaginations where a diplosome is present, and in almost the entire canal cell.  相似文献

Because oxidative stress is involved in arterial hypertension, impairment of hepatic antioxidant defences could develop in the course of this disease. Metallothionein (MT), an antioxidant protein, is present in high rates in the liver. The aim of this study was to investigate the effect of a mineralocorticoid-salt treatment on blood pressure, hepatic antioxidant enzyme activities, and cardiac MT levels in transgenic MT null mice compared with control mice to further clarify the role of MT during the experimental development of arterial hypertension. Control and transgenic MT -/- mice were submitted to an 8-week mineralocorticoid-salt treatment. Hepatic glutathione peroxidase, glutathione reductase, superoxide dismutase, and catalase activities and cardiac MT and mineral levels were measured. Mineralocorticoid-salt treatment induced an increase in blood pressure in both transgenic MT -/- and control mice that was associated with an impairment of liver antioxidant status. MT deficiency was associated with modifications of hepatic antioxidant enzyme activities and with a decrease in cardiac iron levels. Adaptive processes of antioxidant systems may explain the absence of an effect of metallothionein deficiency on the development of mineralocorticoid-salt hypertension. The interactions that occur between the in vivo antioxidant systems probably produce a complex regulation of the oxidative balance and consequently prevent antioxidant deficiency.  相似文献
