Abstract: Mutations in the myelin proteolipid protein (PLP) gene, such as that found in the jimpy mouse, result in an abnormal structure of the myelin, severe dysmyelination, and a reduction in the number of mature oligodendrocytes. To examine the functions of the two alternatively spliced isoforms of proteolipid protein, transgenic mice were generated that express either PLP or DM20 cDNAs placed under control of the PLP upstream regulatory region. The transgenes were bred into jimpy mice, and the effect of the transgenes on the dysmyelinating phenotype was analyzed. Neither the PLP transgene nor the DM20 transgene alone had an effect on myelination in the jimpy mice. Combining the two transgenes substantially increased the number of myelinated axons, suggesting that the two alternatively spliced products of the PLP locus perform distinct functions in oligodendrocytes. The enhanced myelination was not sufficient, however, for completely correcting the dysmyelinating phenotype of the jimpy mice, nor was it accompanied by the restoration of normal levels of myelin gene expression. The inability to rescue the jimpy phenotype is most likely attributable to a dominant negative action of the abnormal proteolipid proteins present in jimpy mice. These results demonstrate the complexity of proteolipid protein function in myelination.  相似文献

An annotated checklist of species of Euphorbiaceae of the Guianas (Guyana, Surinam, French Guiana) and the adjacent region is presented and a key to the genera is provided. A total of 47 indigenous genera and 153 species are known from the Guianas, where species endemism is 14%, with the greatest number of endemics occurring in Guyana and the fewest in French Guiana. An additional 86 species and an additional seven genera are known from neighboring areas of the Guayana Region; many of these may be expected to occur in the Guianas. Taxa known only in cultivation in the Guianas include 22 species and an additional six genera. The following new combination is proposed,Croton vergarenae.  相似文献

Abstract: At least four morphologically distinct forms of common dolphins are found in the eastern Pacific, We compared length data for common dolphins photographed from the northern, central and southern regions as defined by Perrin et al. (1985) and found significant differences in average length for adult animals (>150 cm) and for "adult females" defined for our purposes as animals accompanied by calves. Analyses of calculated birth dates for calves demonstrated differences in timing of reproduction between the geographically adjacent forms. Length distributions from aerial photographs and samples collected from the purse seine fishery were strikingly similar. This work demonstrates a new, non-invasive method for obtaining unbiased life history and morphological data.  相似文献

A recombinant PHYB minigene (m PHYB ) consisting of the complete Arabidopsis PHYB cDNA sequence fused to 2.3 kb of upstream PHYB promoter sequence has been introduced into wild-type Arabidopsis and into a strain containing the Bo64 allele of the hy3 mutation. The Bo64 mutant has previously been shown to contain a nonsense mutation in the PHYB coding sequence. Transformation of this strain with the m PHYB gene results in complementation of all of the mutant phenotypic characteristics tested including hypocotyl length and hypocotyl cell size, response to end-of-day far-red light, leaf morphology, chlorophyll level, and flowering time. Presence of the m PHYB transgene in a wild-type genetic background causes exaggeration of this same set of phenotypic characteristics, indicating that these diverse photomorphogenic responses are sensitive to the copy number of the PHYB gene. The transgene inserts in the Bo64(m PHYB ) and WT(m PHYB ) lines are shown to be single locus and single copy and the immunologically detectable level of phytochrome B is shown to vary linearly with PHYB gene copy number. These results demonstrate a complex role for phytochrome B in Arabidopsis photomorphogenesis and suggest that the expression level of this phytochrome gene is an important determinant of the intensity of light-induced plant responses.  相似文献

Recent studies have shown an association between uncontrollable stress and abdominal fat distribution. It has been suggested that changes in cortisol secretion might represent one possible mechanism for this relationship. This study investigated whether body fat distribution, determined by waist-to-hip ratio (WHR), is related to salivary cortisol levels in response to laboratory stressors. Subjects were 41 overweight women with a Low or a High WHR. Multiple measures of cortisol and mood were obtained during a session of stressful tasks (eg., timed arithmetic) and during a time-matched, control rest session. Also, background life stress and psychological trait variables were assessed. Compared to Low WHR subjects, High WHR subjects secreted significantly more cortisol during the stressful session after 60 minutes of stress, and considering the total area under the curve of secretion. This difference was not seen on the rest day. In terms of background and psychological measures, High WHR subjects were characterized by poorer coping skills and differences in mood reactivity. Specifically, although all subjects became more angry in response to the stressful session, High WHR subjects showed smaller increases in anger. This could indicate that they are more likely to evidence a helpless reaction to uncontrollable stress. These findings support the hypothesis that cortisol secretion might represent a mechanism for the observed association between stress and abdominal fat distribution. Furthermore, differences in coping and appraisal may suggest that a particular psychological pattern might influence the reactivity of the adrenal-cortical system to stress, and subsequent fat distribution.  相似文献

Summary A new cell line was developed from 3-d-old embryonated eggs of the cabbage looper,Trichoplusia ni, and has been designated IPLB-TN-R2. It contains a variety of morphological cell types, including myoblastlike, neuroblastlike, and epithelial-like cells. Chromosome analysis revealed typical lepidopteran chromosomes. Isozyme characterization showed patterns similar to two other cabbage looper cell lines (TN-368 and IAL-TND1) in the case of five enzymes but differed from these two lines for two other enzymes. Virus infectivity tests revealed the line is highly susceptible toAutographa californica nuclear polyhedrosis virus, but no cytopathology was observed after inoculation with several other lepidopteran viruses.  相似文献

Randomly cloned fragments of DNA from Bacteroides thetaiotaomicron were used as hybridization probes for differentiation of B. thetaiotaomicron from closely related Bacteroides species. HindIII digestion fragments of DNA from B. thetaiotaomicron (type strain) were inserted into plasmid pBR322 and labeled with [alpha-32P]dCTP by nick translation. These labeled plasmids were screened for hybridization to HindIII digests of chromosomal DNA from type strains of the following human colonic Bacteroides species: B. thetaiotaomicron, Bacteroides ovatus, reference strain 3452-A (formerly part of B. distasonis), Bacteroides uniformis, Bacteroides fragilis, Bacteroides vulgatus, Bacteroides distasonis, Bacteroides eggerthii, and reference strain B5-21 (formerly B. fragilis subsp. a). Two of the five cloned fragments hybridized only to DNA from B. thetaiotaomicron. Each of these two fragments hybridized to the same DNA restriction fragment in five strains of B. thetaiotaomicron other than the strain from which the DNA was cloned. One of the cloned fragments (pBT2) was further tested for specificity by determining its ability to hybridize to DNA from 65 additional strains of colonic Bacteroides.  相似文献

The α- and β-subunits of chick embryo brain tubulin have been isolated under denaturing conditions and compared with respect to their molecular weight, amino acid composition, tryptic peptide maps, amide content and isoelectric focusing properties. An 8 M-Urea-containing polyacrylamide gel system with varying acrylamide concentrations was used for calculation of the retardation coefficients (KR) of the tubulin subunits. A molecular weight of 53,000 was estimated for each subunit by comparison to KR values for standard proteins. Amide contents of approx 41% of the carboxyl groups of α-tubulin and 48% of the carboxyl groups of β-tubulin were calculated using the average PI value, the pKintrinsic for the ionizable side chains of the amino acids and the amino acid composition of each subunit. Comparative peptide maps of trypsin digested α- and β-tubulin demonstrated 16 peptides unique to each subunit and 23 peptides which comigrate. Both subunits give rise to multiple species on electrofocusing gels. The average isoelectric points for the α- and β-subunits are 5.4 and 5.2, respectively.  相似文献

Summary The pollination biology as related to divergent post-fire reestablishment strategies was examined during flowering sequences for two years in sympatric populations of Arctostaphylos pringlei and A. glandulosa var. mollis. Arctostaphylos pringlei reestablishes post-fire populations by means of fire-stimulated germination of previously dormant seeds, while A. glandulosa var. mollis resprouts from large subsurface burls. Arctostaphylos pringlei produced more flowers per unit plant size, exhibited faster nectar production and higher nectar concentration, attracted nearly twice the number of pollinator visitations, and set more seed when self-pollinated. This substantiated the hypothesis that in these two species the amount of reproductive energy allocated to flower, nectar, and seed production reflected the relative significance of seeding compared to a resprouting post-fire establishment strategy.  相似文献
