An injury and recovery phenomenon was observed in Listeria monocytogenes inoculated into a medium containing 2·2 mmol l−1 NaCl, a concentration that was inhibitory to growth. The apparent loss then recovery of viability, as determined by plate counts, was compared with the uptake of ethidium bromide by the cells and found to be inversely related. Dry, sandy scrub habitats of the Floridapeninsula represent naturally fragmentedremnants of xeric ecosystems that werewidespread during the Pliocene and earlyPleistocene. This habitat is characterized byhigh endemism, and distribution of genetic andevolutionary diversity among scrub ``islands' isof compelling interest because Florida scrub israpidly disappearing under human development. We compare range-wide diversity inmitochondrial cytochrome b sequences forthree scrub-associated lizards with contrastinglevels of habitat specificity. All speciesshow strong geographic partitioning of geneticdiversity, supporting the hypothesis that scrubfauna is highly restricted by vicariantseparations. The mole skink (Eumecesegregius), the least habitat specific, has thelowest phylogeographic structure among thelizards (st = 0.631). The mtDNAgeneology for E. egregius is not entirelyconcordant with the five recognized subspeciesand supports a link between populations incentral Florida (E. e. lividus) and theFlorida Keys (E.e. egregius) rather thana previously proposed affiliation betweennorthern and southern populations. The Floridascrub lizard (Sceloporus woodi) is themost habitat specific of the lizards and hasthe strongest phylogeographic structure (st = 0.876). The sand skink (Neosepsreynoldsi) falls between the moleskink and scrub lizard in terms of habitatspecificity and phylogeographic structure (st = 0.667). For all three species,networks of mtDNA haplotypes coalesce on twocentral ridges that contain the oldest scrub. The geographic structure and deep evolutionarylineages observed in these species have strongimplications for conservation, includingstrategies for translocation, reserve design,and management of landscape connectivity. 相似文献

Introduction
We evaluated the associations between bone marrow lesion (BML) volume change and changes in periarticular bone mineral density (paBMD) as well as subchondral sclerosis to determine whether BML change is associated with other local bone changes.Methods
The convenience sample comprised participants in the Osteoarthritis Initiative (OAI) with weight-bearing posterior-anterior knee radiographs and magnetic resonance images (MRIs) at the 24- and 48-month visits and dual-energy x-ray absorptiometry (DXA) at the 30-/36-month and 48-month visits. The right knee was assessed unless contraindicated for MRI. We used knee DXA scans to measure medial tibia paBMD and medial/lateral paBMD ratio (M:L paBMD). Knee radiographs were scored for sclerosis (grades 0 to 3) in the medial tibia. Two raters determined BML volume on sagittal fat-suppressed MRI by using a semiautomated segmentation method. To focus on knees with only medial tibia BML changes, knees with lateral tibial BMLs were excluded. Medial tibial BML volume change was classified into three groups: BML regression (lowest quartile of medial tibial BML volume change), no-to-minimal change (middle two quartiles), and BML progression (highest quartile). We used proportional odds logistic regression models to evaluate the association between quartiles of changes in medial paBMD or M:L paBMD ratio, as outcomes, and BML volume change.Results
The sample (n = 308) included 163 (53%) female subjects, 212 (69%) knees with radiographic osteoarthritis, and participants with a mean age of 63.8 ± 9.3 years and mean body mass index of 29.8 ± 4.7 kg/m2. We found an association between greater increases in medial tibia paBMD and BML regression (OR = 1.7 (95% confidence interval (CI) = 1.1 to 2.8)) and a similar trend for BML progression (OR = 1.6 (95% CI = 1.0 to 2.6]). We also detected associations between greater increase in M:L paBMD and BML regression (OR = 1.6 (95% CI = 1.0 to 2.7]) and BML progression (OR = 1.8 (95% CI = 1.1 to 3.0)), although BML regression had borderline statistical significance. The frequency of sclerosis progression in the medial tibia (n = 14) was greater among knees with BML progression or regression compared with knees without BML change (P = 0.01 and P = 0.04, respectively).Conclusion
BML regression and BML progression are characterized by concurrent increases in paBMD and sclerosis, which are characteristic of increased radiographic osteoarthritis severity. At least during 24 months, BML regression is not representative of improvement in other periarticular bone measures. 相似文献77.

The impact of SO2 dry deposition from mining emissions on cricket abundance, diversity and composition was investigated at Mount Isa in the semiarid tropics of northern Australia. Seventy-one sites were sampled, stratified at two levels: sulphur deposition zones (high, medium, low, and background zones) and habitat (rocky ridge, rocky plain and alluvial plain). The three habitats did not support distinctly different cricket assemblages. Crickets responded to SO2 emissions, but impacts were largely restricted to the high-sulphur zone. Generalised linear modelling showed significant reductions in the probability of presence in the high-sulphur zone for the three genera Eurepella, Salmanites and Endacusta , and the subfamily Eneopterinae. In ordination space, the three high-sulphur zone by habitat combinations were clearly separated from all other combinations, and vector fitting of environmental variables showed soil SO4 to be a primary correlative factor of this separation. Crickets are sensitive to SO2 emissions and they appear to be a good indicator group in this context. 相似文献

It has been speculated that autoimmune diseases are caused by failure of central tolerance. However, this remains controversial. We have suggested that CD40 expression identifies autoaggressive T cells in the periphery of autoimmune prone mice. In this study, we report that CD40 was cloned from autoaggressive T cells and that engagement induces expression and nuclear translocation of the recombinases, recombination activating gene (RAG) 1 and RAG2 in the autoaggressive, but not in the nonautoaggressive, peripheral T cell population. Furthermore, we demonstrate that CD40 engagement induces altered TCR Valpha, but not Vbeta, expression in these cells. Therefore, CD40-regulated expression of RAG1 and RAG2 in peripheral T cells may constitute a novel pathway for the generation of autoaggressive T cells. 相似文献

Mitogen-activated protein kinases (MAPKs) are key regulators of cellular physiology and immune responses, and abnormalities in MAPKs are implicated in many diseases. MAPKs are activated by MAPK kinases through phosphorylation of the threonine and tyrosine residues in the conserved Thr-Xaa-Tyr domain, where Xaa represents amino acid residues characteristic of distinct MAPK subfamilies. Since MAPKs play a crucial role in a variety of cellular processes, a delicate regulatory network has evolved to control their activities. Over the past two decades, a group of dual specificity MAPK phosphatases (MKPs) has been identified that deactivates MAPKs. Since MAPKs can enhance MKP activities, MKPs are considered as an important feedback control mechanism that limits the MAPK cascades. This review outlines the role of MKP-1, a prototypical MKP family member, in physiology and disease. We will first discuss the basic biochemistry and regulation of MKP-1. Next, we will present the current consensus on the immunological and physiological functions of MKP-1 in infectious, inflammatory, metabolic, and nervous system diseases as revealed by studies using animal models. We will also discuss the emerging evidence implicating MKP-1 in human disorders. Finally, we will conclude with a discussion of the potential for pharmacomodulation of MKP-1 expression. 相似文献
