Stenotopic specialization to a fragmented habitat promotes the evolution of genetic structure. It is not yet clear whether small-scale population structure generally translates into large-scale intraspecific divergence. In the present survey of mitochondrial genetic structure in the Lake Tanganyika endemic Altolamprologus (Teleostei, Cichlidae), a rock-dwelling cichlid genus comprising A. compressiceps and A. calvus, habitat-induced population fragmentation contrasts with weak phylogeographic structure and recent divergence among genetic clades. Low rates of dispersal, perhaps along gastropod shell beds that connect patches of rocky habitat, and periodic secondary contact during lake level fluctuations are apparently sufficient to maintain genetic connectivity within each of the two Altolamprologus species. The picture of genetic cohesion was interrupted by a single highly divergent haplotype clade in A. compressiceps restricted to the northern part of the lake. Comparisons between mitochondrial and nuclear phylogenetic reconstructions suggested that the divergent mitochondrial clade originated from ancient interspecific introgression. Finally, ‘isolation-with-migration’ models indicated that divergence between the two Altolamprologus species was recent (67–142 KYA) and proceeded with little if any gene flow. As in other rock-dwelling cichlids, recent population expansions were inferred in both Altolamprologus species, which may be connected with drastic lake level fluctuations.  相似文献

The Nature Conservancy’s wetland restoration at the Emiquon Preserve has been a success to date, but there are warning signs of undesirable change if left unmanaged. A water control structure built in 2016 will increase management capabilities, but periodic connection to the river, which has experienced human alterations typical of rivers in eastern North America and Europe, also introduces risks. The Conservancy’s planning process has identified (1) management targets (e.g., diverse native fish populations); (2) Key Ecological Attributes (KEAs) that maintain the targets (e.g., relatively deep over-wintering habitats for fishes); (3) measurable indicators for the KEAs (e.g., depth in winter); and (4) desirable ranges for the indicators (e.g., 10% of the aquatic area has depths of 2–3 m and dissolved oxygen levels of 4–6 mg/l). Assessments and experiments completed to date have focused on documenting the restoration, evaluating effects of the record flood of 2013, and predicting outcomes of management actions. Simulation models of hydrology, hydraulics, and vegetation response developed during the planning process allayed some concerns of stakeholders, but not all outcomes are predictable from either current theory or management experience. Therefore, each action can be considered not only as an adaptive management experiment focused on sustaining targets, but also contributing to ecological theory and restoration practice on a broader scale.  相似文献

There is substantial evidence that nonsteroidal anti-inflammatory drugs (NSAIDs) affect cellular processes regulated by Ca(2+) ions, including the metabolic responses of the liver to Ca(2+)-dependent hormones. The aim of the present study was to determine whether the effects of naproxen are mediated by a direct action on cellular Ca(2+) fluxes. The effects of naproxen on 45Ca(2+) fluxes in mitochondria, microsomes and inside-out plasma membrane vesicles were examined. Naproxen strongly impaired the mitochondrial capacity to retain 45Ca(2+) and inhibited also ATP-dependent 45Ca(2+) uptake by microsomes. Naproxen did not modify 45Ca(2+) uptake by inside-out plasma membrane vesicles, but it inhibited the hexokinase/glucose-induced Ca(2+) efflux from preloaded vesicles. Additional assays performed in isolated mitochondria revealed that naproxen causes mitochondrial uncoupling and swelling in the presence of Ca(2+) ions. These effects were prevented by EGTA, ruthenium red and cyclosporin A, indicating that naproxen acts synergistically with Ca(2+) ions by promoting the mitochondrial permeability transition. The experimental results suggest that naproxen may impair the metabolic responses to Ca(2+)-dependent hormones acting by at least two mechanisms: (1) by interfering with the supply of external Ca(2+) through a direct action on the plasma membrane Ca(2+) influx, and (2) by affecting the refilling of the agonist-sensitive internal stores, including endoplasmic reticulum and mitochondria.  相似文献

Environmental conditions may affect invasibility and potentially prevent successful invasions of the freshwater bivalve Limnoperna fortunei Dunker, 1857. However, even though the larval stage may be considered an important species’ characteristic, the invasion processes of L. fortunei are mainly evaluated using only the adult stage. Therefore, the aim of this study was to identify what, and how, environmental filters might predict the likelihood of occurrence of each of the L. fortunei larval stages. Logistic regressions were applied using the larval stages of L. fortunei as a surrogate of population age structure and the environmental variables as the main filters that potentially predicted the likelihood of the occurrence of each larval stage of L. fortunei. The turbidity predicted the occurrence of the D-shaped larvae and straight-hinged veliger stages, while the umbonated-veliger and pediveliger stages were predicted by the pH and conductivity. Finally, the phytoplankton density (diatoms) predicted the occurrence of the umbonated-veliger, pediveliger and mainly the plantigrade stages. Our findings suggested that, during larvae development, from younger to older stages, the main environmental predictor of larvae occurrence shifted from abiotic to biotic variables, indicating that larval stages are indeed an important factor that helps in the evaluation of freshwater invasibility. These findings are particularly important for floodplain systems, where flood pulse dynamics may increase propagule pressure, leading to a successful spread among habitats. Then, even though environmental filters were important to predict successful invasions, we highlight that a successful invasion might depend on the population age structure of the invading species arriving in the new habitat. Thus, successful L. fortunei invasions in floodplain systems may depend on the interaction between invasiveness, invasibility and propagule pressure.  相似文献

Plasmodium simium is a parasite from New World monkeys that is most closely related to the human malaria parasite Plasmodium vivax; it also naturally infects humans. The blood-stage infection of P. vivax depends on Duffy binding protein II (PvDBPII) and its cognate receptor on erythrocytes, the Duffy antigen receptor for chemokines (hDARC), but there is no information on the P. simium erythrocytic invasion pathway. The genes encoding P. simium DBP (PsDBPII) and simian DARC (sDARC) were sequenced from Southern brown howler monkeys (Alouatta guariba clamitans) naturally infected with P. simium because P. simium may also depend on the DBPII/DARC interaction. The sequences of DBP binding domains from P. vivax and P. simium were highly similar. However, the genetic variability of PsDBPII was lower than that of PvDBPII. Phylogenetic analyses demonstrated that these genes were strictly related and clustered in the same clade of the evolutionary tree. DARC from A. clamitans was also sequenced and contained three new non-synonymous substitutions. None of these substitutions were located in the N-terminal domain of DARC, which interacts directly with DBPII. The interaction between sDARC and PvDBPII was evaluated using a cytoadherence assay of COS7 cells expressing PvDBPII on their surfaces. Inhibitory binding assays in vitro demonstrated that antibodies from monkey sera blocked the interaction between COS-7 cells expressing PvDBPII and hDARC-positive erythrocytes. Taken together, phylogenetic analyses reinforced the hypothesis that the host switch from humans to monkeys may have occurred very recently in evolution, which sheds light on the evolutionary history of new world plasmodia. Further invasion studies would confirm whether P. simium depends on DBP/DARC to trigger internalization into red blood cells.  相似文献

In this study, we evaluated how cichlid assemblages are affected by seasonal and interannual variations in water volume in the floodplain lagoons of the Cuiabá River in the Pantanal wetland. Eleven lagoons were sampled in the early dry season and the early rainy season over a three-year period. Analysis of similarity (ANOSIM) showed that the species compositions and relative abundances were distinct between the seasons, which indicates that neither persistence nor stability was maintained on a seasonal basis. However, the interannual comparisons show the opposite tendency, indicating strong stability and persistence on an annual basis. These results indicate that the cichlid assemblages in these lagoons have two equilibrium points, one in the early dry season and the other in the early rainy season, and that the assemblages alternate between these points through the hydrologic cycle. In addition to the effect of the water level variation, the abundance distribution is also affected by conductivity and pH, which also influence the total abundance of the species in the assemblage.  相似文献

Recent work with P-36 demonstrates that the replacement of the last two doses of Follicle-Stimulating Hormone (FSH) with equine chorionic gonadotropin (eCG) increases embryo yields. However, it is unclear if the positive effect of eCG is related to its FSH-like activity, LH-like activity, or both. This study aimed to verify the replacement of eCG with pLH on the last day of superstimulatory treatment. Twenty-five Nelore cows were allocated to four groups: P-36 (control), P-36/eCG, P-36/LH2, and P-36/LH4. All animals underwent four treatments in a crossover design. The control group cows were superstimulated with decreasing doses of porcine Follicle-Stimulating Hormone (pFSH, 133 mg, im). In the P-36/eCG, P-36/LH2, and P-36/LH4 groups, the last two doses of pFSH were replaced in the former group by two doses of eCG (200 IU each dose, im) and in the latter two groups by two doses of pLH (1 and 2 mg each dose, im), respectively. Donors received fixed-time artificial insemination 12 and 24 hours after pLH. Embryo flushing was performed on D16. Data were analyzed by ANOVA (Proc Mixed, SAS). There was a trend of decreasing ovulation rate when comparing groups LH2 and eCG (P = 0.06). However, there was no significant difference in the mean number of viable embryos among groups P-36 (3.3 ± 0.7), P-36/eCG (4.5 ± 0.5), P-36/LH2 (3.7 ± 0.8), and P-36/LH4 (4.2 ± 1.0). It is concluded that the replacement of eCG by pLH on the last day of superstimulatory treatment can be performed with no significant variation in the production of viable embryos.  相似文献

Strong evidence suggests that quality strategic behaviour change communication (BCC) can improve malaria prevention and treatment behaviours. As progress is made towards malaria elimination, BCC becomes an even more important tool. BCC can be used 1) to reach populations who remain at risk as transmission dynamics change (e.g. mobile populations), 2) to facilitate identification of people with asymptomatic infections and their compliance with treatment, 3) to inform communities of the optimal timing of malaria control interventions, and 4) to explain changing diagnostic concerns (e.g. increasing false negatives as parasite density and multiplicity of infections fall) and treatment guidelines. The purpose of this commentary is to highlight the benefits and value for money that BCC brings to all aspects of malaria control, and to discuss areas of operations research needed as transmission dynamics change.  相似文献
