The cyanobacterium Acaryochloris marina is unique because it mainly contains Chlorophyll d (Chl d) in the core complexes of PS I and PS II instead of the usually dominant Chl a. Furthermore, its light harvesting system has a structure also different from other cyanobacteria. It has both, a membrane-internal chlorophyll containing antenna and a membrane-external phycobiliprotein (PBP) complex. The first one binds Chl d and is structurally analogous to CP43. The latter one has a rod-like structure consisting of three phycocyanin (PC) homohexamers and one heterohexamer containing PC and allophycocyanin (APC). In this paper, we give an overview on the investigations of excitation energy transfer (EET) in this PBP-light-harvesting system and of charge separation in the photosystem II (PS II) reaction center of A. marina performed at the Technische Universität Berlin. Due to the unique structure of the PBP antenna in A. marina, this EET occurs on a much shorter overall time scale than in other cyanobacteria. We also briefly discuss the question of the pigment composition in the reaction center (RC) of PS II and the nature of the primary donor of the PS II RC.  相似文献

A rapid multiplexed fingerprinting method has been developed for bacterial artificial chromosome (BAC) contig assembly. Defined subsets of BAC DNA fragments that result from digestion by three paired restriction endonucleases are labeled with unique fluorescent F-ddATP for each subset. Lists of the labeled fragment size are generated by an ABI 377 DNA sequencer and the GeneScan analysis software and then processed by an assembly program, FPC (Fingerprinted Contigs), to produce contig maps. Data obtained from the multiplexed labeling permit detection of smaller overlaps than is observed when data from a single double-digest are analyzed. The method has been tested on 98 BACs from chromosome 22 regions where large-scale sequencing is under way and also through simulation, using randomly generated BAC clones derived from existing DNA sequence data. In each case, contig assembly results demonstrated the advantages of multiplexed fingerprinting.  相似文献

IntroductionRhoA has been shown to be beneficial in cardiac disease models when overexpressed in cardiomyocytes, whereas its role in cardiac fibroblasts (CF) is still poorly understood. During cardiac remodeling CF undergo a transition towards a myofibroblast phenotype thereby showing an increased proliferation and migration rate. Both processes involve the remodeling of the cytoskeleton. Since RhoA is known to be a major regulator of the cytoskeleton, we analyzed its role in CF and its effect on myofibroblast characteristics in 2 D and 3D models.ResultsDownregulation of RhoA was shown to strongly affect the actin cytoskeleton. It decreased the myofibroblast marker α-sm-actin, but increased certain fibrosis-associated factors like TGF-β and collagens. Also, the detailed analysis of CTGF expression demonstrated that the outcome of RhoA signaling strongly depends on the involved stimulus. Furthermore, we show that proliferation of myofibroblasts rely on RhoA and tubulin acetylation. In assays accessing three different types of migration, we demonstrate that RhoA/ROCK/Dia1 are important for 2D migration and the repression of RhoA and Dia1 signaling accelerates 3D migration. Finally, we show that a downregulation of RhoA in CF impacts the viscoelastic and contractile properties of engineered tissues.ConclusionRhoA positively and negatively influences myofibroblast characteristics by differential signaling cascades and depending on environmental conditions. These include gene expression, migration and proliferation. Reduction of RhoA leads to an increased viscoelasticity and a decrease in contractile force in engineered cardiac tissue.  相似文献

Experiments to determine the effects of extraction techniques and the influence of shipping on extraction of Heterodera glycines life stages gave variable results. Shipping did not significantly affect numbers of nematodes extracted. More second-stage juveniles (J2) were extracted with Baermann funnels than with an elutriator, probably because incubation of encysted eggs on the Baermann funnel for 1 week allowed hatching to occur. Sieving was more efficient than elutriation for extracting cysts. Adding air agitation to the water pressure during elutriation increased extraction efficiency of cysts but not J2. Sample sizes of 250 cm³ and 500 cm³ did not influence extraction efficiency of cysts; however, sample size did influence extraction of J2.  相似文献

The terpenoid composition of three fossil resins from macrofossils of Cretaceous and Tertiary conifers has been analyzed by gas chromatography–mass spectrometry (GC–MS). The mono-, sesqui- and diterpenoids which have been identified in the resin extracts are derived from precursors produced by the respective source plants and may be used as chemosystematic markers when compared with terpenoids in extant conifers. Sesquiterpenoids (cedrene, cuparene, cadinanes) and phenolic diterpenoids (ferruginol and derivatives) are the major components in Cupressospermum saxonicum Mai (Miocene). The terpenoid characteristics strongly support a relationship to the Cupressaceae s. str. The resin of Doliostrobus taxiformis (Sternberg) Kva ek (Eocene) consists of abietane and pimarane type resin acids accompanied by minor amounts of phenolic diterpenoids (ferruginol, hinokiol). According to morphological and anatomical characteristics, D. taxiformis was previously compared to both, extant Araucariaceae and Cupressaceae s.l., but the terpenoid pattern of the resin now supports a relationship to the Cupressaceae s.l. rather than to Araucariaceae. Degraded diterpenoids of the abietane type are the major compounds in the extract of Tritaenia linkii (Roemer) Mägdefrau et Rudolf (Lower Cretaceous) indicating considerable oxidative alteration of the resin. Since the terpenoids in the resin of T. linkii are highly degraded or belong to the common abietane class, the leaves cannot be assigned or compared to any modern family based on their terpenoid composition. The presence of ferruginol probably excludes pinaceous affinities. Terpenoids proved to be valuable chemosystematic markers for fossil conifers once they are adequately preserved. The analysis of resin extracts by GC–MS is a suitable tool for the investigation of soluble compounds in fossil plants.  相似文献
