Effects of mannitol and chlorpromazine pretreatment of rabbits on kidney mitochondria following in vivo ischemia and reflow 总被引:1,自引:0,他引:1
The effects of in vivo ischemia and reflow on the respiratory control ratio (RCR) of rabbit kidney mitochondria (homogenates) was studied in rabbits pretreated with mannitol or chlorpromazine (CPZ). Two hours of ischemia damages mitochondria and lowers the RCR from about 13 to 2. Reflow to ischemic kidneys does not affect the RCR (RCR = 2.1), unless the rabbits are pretreated with mannitol (RCR = 11) or CPZ (RCR = 13.9). Although mannitol or CPZ pretreatment is effective at fully restoring the RCR to normal levels, the maximal rate of ADP-stimulated respiration remains partially depressed. Three hours of ischemia followed by reflow does not allow restoration of mitochondrial RCR even with mannitol or CPZ pretreatment. The mechanism of action of mannitol and CPZ may be explained on the basis of their vascular effects resulting in increased reflow in ischemic kidneys. Although a direct effect upon the metabolism of the ischemic kidney remains a possibility. 相似文献

The release of spectrin-free vesicles from ATP-depleted human red blood cells (Lutz et al. (1977) J. Cell. Biol. 73, 548) can be considered the final step of a shape change from discocytes to echinocytes. The study of physical and chemical properties of released vesicles suggests that vesicle release is not merely a consequence of charge alterations within either monolayer of the budding membrane. Fresh membranes and released vesicles have within experimental error the same sialic acid content per surface area and the same electrophoretic mobilities. Vesicle release cannot be stimulated by doubling the charge density on the outer monolayerby means of a phospholipase D-treatment, but correlates with a breakdown of polyphosphoinositides to diacylglycerol on the inner monolayer. This breakdown does not lead to a significant change in the negative charge density on the inner monolayer, because an increased phosphatidate content compensates for this alteration. Furthermore, polyphosphoinositide breakdown and diacylglycerol production are not the rate-limiting step in vesicle release from ATP-depleting red blood cells. This is evident from the fact that 10 mM EDTA inhibits vesicle release to 75% without affecting polyphosphoinositide breakdown and diacylglycerol production. Hence, diacylglycerol formation may be sufficient for membrane budding as suggested earlier (Allan et al.(1976) Nature 261, 58), but vesicle release requires a second, as yet unidentified process. 相似文献

International Journal for Parasitology9: 153–157. Twenty-four castrated Friesian calves were divided into two equal groups on a body weight basis and grazed separate halves of an infected pasture. One group was given 1.5 mg/kg per day morantel tartrate in the feed over a period extending from 2 weeks before turning out in early May to early July. The other group of calves received an equivalent amount of unmedicated food as controls. Pasture larval counts showed a similar pattern and level of infection from February to July on both pastures. This consisted of a fall from moderate winter levels to very low levels in April/July. The control pasture then showed a typical sharp rise in August to high levels extending through September and falling to moderate levels in October. The pasture being grazed by treated calves showed only a small rise in larval counts in August. The total pasture contamination was reduced by 85% compared with controls during August/October. Faecal egg and larval counts showed similar differences between treated and control calves. Mild clinical signs of parasitic gastroenteritis and bronchitis were seen in controls but not in the treated group. The latter had a weight gain advantage of 20.5 kg the difference becoming marked in the last month of the trial. 相似文献

The carotenoids bound to reaction centers of wild, Ga and GlC strains of Rhodopseudomonas spheroides, of Rhodospirillum rubrum, strain S1 and of Rhodopseudomonas viridis, yield very similar, but unusual resonance Raman spectra. Through a comparison with resonance Raman spectra of 15,15′-cis-β-carotene, these carotenoids are shown to assume cis conformations, while the corresponding chromatophores contain all-trans forms only. These cis conformations likely are identical for all the carotenoids studied. They remain unaffected by variations of temperature from 20 to 300 K as well as by the redox state of . They are unstable, being rapidly isomerised towards the all-trans forms when extracted from the reaction centers. Summary In the present investigation experiments were carried out to determine whether the functionally obscure synaptic ribbons of mammalian pinealocytes can be affected by acute changes in environmental lighting and which chemical processes may be involved in their regulation. Experiments carried out in male guinea-pigs have shown that the amounts of synaptic ribbons are immediately affected by changes in the lighting pattern. Extension of the light period reduced the normally occurring increase, whereas extension of the dark period inhibited the normally occurring decrease in the amount of synaptic ribbons.
