The ultrastructure of some integumental glands occurring in the head, thorax and abdomen of K. flavicollis soldiers is described. The secretory units consist of two cells, the canal cell and the secretory cell (this latter filled with secretion granules). A cylindrical and distorted extracellular space, or reservoir, with an irregular outline is lined by short microvilli. The end-apparatus is made up of small overlapping cuticular laminae which in section resemble small wavy rods. The ample distribution of the units has led the authors to consider them dermal glands. Scanning electron micrographs confirm that the glands' activity consists in the secretion of material which then spreads over the surface of the integument. The dissimilar appearance of the secretion granules present in glands of different soldiers suggests that the electron-lucid granules and the granules with fibrils are two completely different secretions at different ages of the animal. The authors do not therefore rule out the hypothesis that these integumental glands may later produce or release pheromones.  相似文献

Four human hybridoma cell lines (PEB1-4) were established from a fusion of pleural effusion lymphocytes isolated from a breast cancer patient with metastatic disease, 6 years postmastectomy. The hybridomas secreted IgG-k (3 micrograms/ml/10(6) cells). These monoclonal antibodies (PEB1-4) reacted to different degrees with mouse mammary tumor virus (MMTV) and T47D particles (HuMTV). Immunological cross-reaction was also detected with antigens isolated from body fluids of breast cancer patients (BF-Ag). The binding capacity of the monoclonal antibodies (MAbs) PEB1-4 to the above-mentioned antigens was measured by RIA. The specificity of these antibodies was further demonstrated by radioimmunoprecipitation of MMTV, T47D (HuMTV) and BF-Ag. The binding of PEB1-4 to surface antigens of intact cells grown in culture was measured by RIA. Some of the MAbs were shown to bind more avidly to breast cancer cells than to nonbreast cancer cells or nonmalignant cells. The PEB1-4 human monoclonal antibodies may be found useful in analyzing the virus-breast cancer relationship.  相似文献

ExoQuick-TCTM (EQ), a chemical-based agent designed to precipitate exosomes, was calibrated for use on saliva collected from healthy individuals. The morphological and molecular features of the precipitations were compared with those obtained using the classical, physical-based method of ultracentrifugation (UC). Electron microscopy and immunoelectron microscopy with anti-CD63 showed vesicular nanoparticles surrounded by bi-layered membrane, compatible with exosomes in EQ, similar to that observed with UC. Atomic force microscopy highlighted larger, irregularly shaped/aggregated EQ nanoparticles that contrasted with the single, round-shaped UC nanoparticles. ELISA (performed on 0.5 ml of saliva) revealed a tendency for a higher expression of the specific exosomal markers (CD63, CD9, CD81) in EQ than in UC (p>0.05). ELISA for epithelial growth factor receptor, a non-exosomal-related marker, showed a significantly higher concentration in EQ than in UC (p=0.04). Western blotting of equal total-protein concentrations revealed bands of CD63, CD9 and CD81 in both types of preparations, although they were less pronounced in EQ compared with UC. This may be related to a higher fraction of non-exosomal proteins in EQ. In conclusion, EQ is suitable and efficient for precipitation of salivary exosomes from small volumes of saliva; however, EQ tends to be associated with considerably more biological impurities (non-exosomal-related proteins/microvesicles) as compared with UC.  相似文献

The effect of procaine hydrochloride and its metabolites on the diffusional water exchange through erythrocyte membranes was investigated at 37 degrees C and at concentrations ranging between 5 X 10(-5) M and 5 X 10(-1) M by using the NMR manganese doping method. Procaine hydrochloride and 2-diethylaminoethanol have a moderate stimulating effect on the water exchange, of up to 20% at concentrations ranging between 10(-3) and 10(-2) M, while an increasing inhibitory effect was found at higher concentrations. The p-aminobenzoic acid has no effect on the water exchange up to 10(-2) M and, at higher concentrations, and apparent decreasing inhibition was noticed which is thought to be an artefact due to the uptake of Mn2+ by the cells. The temperature dependence studies suggest that procaine HCl enhances the uptake of Mn2+ by the cells. An opposite effect was found for rigid erythrocytes. The p-aminobenzoic acid and 2-diethylaminoethanol appeared to be more effective than procaine hydrochloride in increasing the uptake of Mn2+.  相似文献

In developing countries, as well as in many western countries, members of the genus Campylobacter are recognized as one of the most common cause of acute bacterial enteritis. Campylobacter jejuni and Campylobacter coli isolation rates have been shown to be equal to, and sometimes higher than those of other enteric pathogens. The Microbiology Laboratory of the local Public Health Authority in Prahova County conducted a one and a half-year laboratory-based survey of Campylobacter infections in patients suffering from gastrointestinal symptoms. From a total of 3284 stool samples screened, the culture-positive ones confirmed the bacterial etiology for 551 diarrhea cases. Campylobacter was found in 345 specimens, being the most frequently isolated enteropathogen. C. jejuni outnumbered C. coli species (239 vs. 106 isolates). Salmonella isolates were the second local cause of diarrhea. The highest isolation rate of Campylobacter was found in children 5 years of age (262 strains). The prevalence of campylobacteriosis declined with age. The isolation rate of Campylobacter (10.5%), the unimodal age-specific distribution of cases, as well as the identification of polymicrobial infections among the screened population were epidemiological aspects resembling reports on campylobacteriosis in developing countries. The susceptibility of Campylobacter isolates to various antimicrobial agents, including macrolides and fluoroquinolones was also assessed. Among the screened isolates, Erythromycin retained a good activity, while an increased ciprofloxacin resistance was observed. The information gathered through this local study sustains the importance of Campylobacter in the etiology of autochthonous infectious diarrhea. A development of a national surveillance program regarding the most important foodborne pathogens would be beneficial for improving prevention and controlling measures.  相似文献

Regulation of genetic variation in natural populations is a problem of primary importance to evolutionary biology. In the reported study, the repair efficiency of double strand DNA breaks was compared in six wild barley accessions from Israeli natural populations of H. spontaneum: three from mesic populations (one from Maalot and two from Mount Meron, Upper Galilee) and three from xeric populations (one from Wadi Quilt in the Judean Desert and two from Sede Boqer, in the northern Negev Desert). Pulsed field gel electrophoresis was used to score double-strand breaks of DNA (DSBs) caused by methyl methanesulphonate (MMS) treatment. All six accessions were also tested for heat tolerance: four of these, three xeric and one mesic (from Maalot population), were scored as heat tolerant whereas both accessions from Mount Meron population displayed heat sensitivity. MMS caused a significant increase in the level of DSBs relative to the control in all accessions. The major questions were whether and how the efficiency of DNA repair after mutagenic treatment is affected by the environmental conditions and accession’s adaptation to these conditions. Differences were found among the accessions in the repair pattern. Plants of two out of the four heat tolerant accessions did not manage to repair DNA neither at 25°C nor at 37°C. The remaining two heat tolerant accessions significantly repaired the breaks at 37°C, but not at 25°C. By contrast, plants of the two heat susceptible accessions significantly lowered the level of DSBs at 25°C but not at 37°C. Therefore, the accessions that proved capable to repair the induced damages in DNA at one of the two temperatures displayed a pattern that may imply the existence of a negative feedback mechanism in regulation of genetic variation. Such a dependence of DNA integrity on environment and genotype may serve an important factor for maintaining relatively high level of mutability without increasing the genetic load.  相似文献

Flor strains of Saccharomyces cerevisiae form a biofilm on the surface of wine at the end of fermentation, when sugar is depleted and growth on ethanol becomes dependent on oxygen. Here, we report greater biofilm formation on glycerol and ethyl acetate and inconsistent formation on succinic, lactic, and acetic acids.Flor or velum formation by certain wine strains of Saccharomyces cerevisiae (flor strains) is a form of cellular aggregation observed as an air-liquid interfacial biofilm at the end of the alcoholic fermentation. Formation of the biofilm appears to be an adaptive mechanism because it ensures access to oxygen and therefore permits continued growth on nonfermentable ethanol. In general, nonbuoyant cells cease growth at the end of completed wine fermentations not for lack of carbon but for lack of oxygen. Biofilm cells have been found to have an elevated and/or altered lipid content and increased surface hydrophobicity (3, 5, 8, 9, 11). While both Hsp12, a small heat shock protein (13), and Muc1 (also known as Flo11), a hydrophobic cell wall mannoprotein (4, 6), have been shown to be required for the flor biofilm (10, 12, 14), other genetic or environmental requirements, other than an absence of glucose and the presence of ethanol and oxygen, have not been demonstrated. Here, we asked whether flor formation could be induced during growth on nonfermentable substrates other than ethanol. On the basis of dry weight of biofilm formed per mg of available carbon, the best carbon sources were found to be glycerol, ethyl acetate, and ethanol, in descending order. While subsurface growth occurred on acetic, dl-lactic, and succinic acids, an air-liquid interfacial biofilm did not always form. Microarray analysis of cells shifted from growth on glucose to growth on ethanol did not detect significant changes in expression of known biofilm formation-associated genes.  相似文献
