The nicotinic acetylcholine receptor from Torpedo marmorata was extracted and purified from the electroplax membranes by using both aqueous detergent (proteodetergent) or chloroform-methanol (proteolipid). When studied with a highly sensitive radioimmunoassay, it was found that both proteins do not cross-react immunologically against an antireceptor antiserum prepared with the proteodetergent. Treatment with organic solvents of the electroplax membranes, as well as of the proteodetergent receptor purified by affinity chromatography, impaired the radioimmunoassay. This suggests that the antigenicity has been affected by the change in solvent polarity, even though both proteins have similar binding properties for cholinergic ligands. These findings do not allow a simple immunological comparison between the cholinergic proteodetergent and the proteolipid as previously stated in the literature.  相似文献

A major challenge in network ecology is to describe the full-range of species interactions in a community to create highly-resolved food-webs. We developed a molecular approach based on DNA full barcoding and mini-barcoding to describe difficult to observe plant – leaf miner – parasitoid interactions, consisting of animals commonly regarded as agricultural pests and their natural enemies. We tested the ability of universal primers to amplify the remaining DNA inside leaf miner mines after the emergence of the insect. We compared the results of a) morphological identification of adult specimens; b) identification based on the shape of the mines; c) the COI Mini-barcode (130 bp) and d) the COI full barcode (658 bp) fragments to accurately identify the leaf-miner species. We used the molecular approach to build and analyse a tri-partite ecological network of plant – leaf miner – parasitoid interactions. We were able to detect the DNA of leaf-mining insects within their feeding mines on a range of host plants using mini-barcoding primers: 6% for the leaves collected empty and 33% success after we observed the emergence of the leaf miner. We suggest that the low amplification success of leaf mines collected empty was mainly due to the time since the adult emerged and discuss methodological improvements. Nevertheless our approach provided new species-interaction data for the ecological network. We found that the 130 bp fragment is variable enough to identify all the species included in this study. Both COI fragments reveal that some leaf miner species could be composed of cryptic species. The network built using the molecular approach was more accurate in describing tri-partite interactions compared with traditional approaches based on morphological criteria.  相似文献

Abstract Lowland temperate grasslands dominated by Themeda triandra (kangaroo grass) are an endangered ecosystem in southeastern Australia. Grass biomass must be removed frequently to maintain plant diversity, but few studies of the impacts of different biomass removal techniques have been undertaken, and no rapid monitoring schemes have been developed. Low species densities in many reserves (due to past stock grazing) make it difficult to assess the effects of management regimes on plant diversity. Management impacts could be assessed by planting indicator species in replicated enhancement plots and subjecting these plots to adaptive management trials. A protocol for selecting potential indicator species is described, based on a regional quadrat database, using clearly defined criteria. Potential indicator species need to be conspicuous, easy to identify and abundant in high quality diverse grassland remnants, to have relatively broad ecological tolerances, to occur in sites that are relatively species rich and have a comparatively low cover of dominant exotic species, to commonly persist at low densities in long‐grazed reserves, to be responsive to changes in management, and to have been studied ecologically. Only three species from western Victorian grasslands satisfied these criteria: Calocephalus citreus (lemon beauty‐heads), Chrysocephalum apiculatum (common everlasting), and Leptorhynchos squamatus (scaly buttons). All are widespread, herbaceous, hemicryptophytic daisies. Despite a number of caveats, the scheme has the potential to provide a more clearly focused framework for grassland ecosystem management than currently exists.  相似文献

Grassy woodlands dominated by Eucalyptus species such as E. albens Benth., E. melliodora A. Cunn. ex Schauer, and E. microcarpa Maiden are poorly conserved in Australia, owing to widespread agricultural development. Understorey restoration is vitally important to enhance degraded remnants but no reliable techniques are available to restore herbaceous understorey species over large areas. Reestablishment of dominant native grasses such as Kangaroo grass (Themeda triandra Forssk.) is particularly important. This study compared Themeda establishment using a variety of sowing techniques across a range of landscape positions in degraded woodlands in central New South Wales. Four localities were sampled at three landscape positions (upper, mid‐, and lower slopes). Existing vegetative cover was sprayed with herbicide and removed by mowing. A randomized block experiment was established at each site with five replicates of five seedbed treatments: all four factorial combinations of soil disturbance (disturbed, nondisturbed) and weed control (atrazine, nonatrazine), plus topsoil scalping followed by soil disturbance. After 117 days, all plots were subject to a common herbicide (atrazine) treatment to control weeds. Themeda establishment was not significantly affected by landscape position, despite significant differences in the cover of exotic species among landscape positions. The scalped/disturbed treatment resulted in significantly better establishment at 400 days (18% or 3.9 plants/m2) than the control (8% or 1.8 plants/m2). All other treatments did not differ significantly from the control suggesting that seedbed treatments are not critical to successful Themeda establishment. These results suggest that Themeda swards can be reestablished in degraded woodlands relatively simply. The similarity in outcomes across all landscape positions suggests that general recommendations for “best‐bet” establishment methods may prove robust under a wide range of environmental and seasonal conditions.  相似文献

West Nile virus (WNV) is a mosquito-borne flavivirus that primarily infects birds but occasionally infects humans and horses. Certain species of birds, including crows, house sparrows, geese, blue jays and ravens, are considered highly susceptible hosts to WNV. The nonstructural protein 1 (NS1) of WNV can elicit protective immune responses, including NS1-reactive antibodies, during infection of animals. The antigenicity of NS1 suggests that NS1-reactive antibodies could provide a basis for serological diagnostic reagents. To further define serological reagents for diagnostic use, the antigenic sites in NS1 that are targeted by host immune responses need to be identified and the potential diagnostic value of individual antigenic sites also needs to be defined. The present study describes comprehensive mapping of common immunodominant linear B-cell epitopes in the WNV NS1 using avian WNV NS1 antisera. We screened antisera from chickens, ducks and geese immunized with purified NS1 for reactivity against 35 partially overlapping peptides covering the entire WNV NS1. This study identified twelve, nine and six peptide epitopes recognized by chicken, duck and goose antibody responses, respectively. Three epitopes (NS1-3, 14 and 24) were recognized by antibodies elicited by immunization in all three avian species tested. We also found that NS1-3 and 24 were WNV-specific epitopes, whereas the NS1-14 epitope was conserved among the Japanese encephalitis virus (JEV) serocomplex viruses based on the reactivity of avian WNV NS1 antisera against polypeptides derived from the NS1 sequences of viruses of the JEV serocomplex. Further analysis showed that the three common polypeptide epitopes were not recognized by antibodies in Avian Influenza Virus (AIV), Newcastle Disease Virus (NDV), Duck Plague Virus (DPV) and Goose Parvovirus (GPV) antisera. The knowledge and reagents generated in this study have potential applications in differential diagnostic approaches and subunit vaccines development for WNV and other viruses of the JEV serocomplex.  相似文献
