Different criteria have been suggested for decisions on character state recognition for morphological characters showing continuous variation from taxon to taxon. A method proposed byMishler andDe Luna seeks to recognize as many character states as the groups of taxon means that are revealed by an analysis of variance (ANOVA) followed by multiple range tests. We apply this method as a guide in decisions of selecting taxonomic characters in species of theBletia reflexa complex. Seven species of the complex, represented by 156 individuals belonging to 54 populations, were the terminal taxa of the study. Character state codes were assigned to 20 characters. These characters together with 12 non-quantitative characters were incorporated into cladistic analyses. Results indicate that the complex is monophyletic only under one outgroup combination.  相似文献

Small specimens of formalin-fixed tissues approximatey 1 × 1 × 0.2 cm were cut from the suspect specimen. Several clean microscope slides were dipped in 1% aqueous gelatin and air-dried or dried on a slide warmer. Each tissue specimen was washed in running tap water for 2-5 min and then lightly scraped with a straight knife blade, cutting edge perpendicular to the surface of the specimen. The scrapings were allowed to build up and cling to the knife blade, which was then turned so that the broad surface contacted the slide; thus, the scrapings could be smeared onto the slide in a single motion. Sufficient pressure was applied to embed the tissue fragments in the gelatin coating. Smears, dried in air or on a slide warmer, were stained immediately by a standard direct or indirect technique to detect fluorescein-labeled antigens. This scraping method, adapted to the study of leptospirosis by fluorescent-antibody technique, could reduce the need for cryostat-cut tissues and facilitate the observation of individual leptospires.  相似文献

Phytohormones participate in many aspects of the plant life cycle, including responses to biotic and abiotic stresses. They play a key role in plant responses to the environment with direct bearing on a plant’s fitness for adaptation and reproduction. In recent years, there have been major advances in our understanding of the role of phytohormones in halophytic plants. The variability in maximal salinity level that halophytes can tolerate makes it difficult to characterize the specific traits responsible for salt tolerance. However, the most evident effect of salinity is growth disturbance, and growth is directly governed by phytohormones. Phytohormones such as abscisic acid, salicylic acid ethylene and jasmonates are traditionally related to stress responses, while the involvement of cytokinins, gibberellins and auxins has started to be analyzed. Polyamines, although they can’t be considered phytohormones because of the high concentrations required for cell responses, have been proposed as a new category of plant growth regulators involved in several plant processes and stress responses. This review integrates the advances in the knowledge about phytohormones in halophytes and their participation in salt tolerance.  相似文献

Pulmonary surfactant is a complex mixture of lipids and proteins that forms a surface-active film at the air-water interface of alveoli capable of reducing surface tension to near 0 mN/m. The role of cholesterol, the major neutral lipid component of pulmonary surfactant, remains uncertain. We studied the physiological effect of cholesterol by monitoring blood oxygenation levels of surfactant-deficient rats treated or not treated with bovine lipid extract surfactant (BLES) containing zero or physiological amounts of cholesterol. Our results indicate no significant difference between BLES and BLES containing cholesterol immediately after treatment; however, during ventilation, BLES-treated animals maintained higher PaO2 values compared to BLES+cholesterol-treated animals. We used a captive bubble tensiometer to show that physiological amounts of cholesterol do not have a detrimental effect on the surface activity of BLES at 37 degrees C. The effect of cholesterol on topography and lateral organization of BLES Langmuir-Blodgett films was also investigated using atomic force microscopy. Our data indicate that cholesterol induces the formation of domains within liquid-ordered domains (Lo). We used time-of-flight-secondary ion mass spectrometry and principal component analysis to show that cholesterol is concentrated in the Lo phase, where it induces structural changes.  相似文献
