利用尾气分析仪对发酵过程的尾气中的O2、CO2含量进行实时检测,建立了裂殖弧菌发酵生产DHA过程中的呼吸参数在线检测方法,实现了裂殖壶菌补料分批发酵过程及双阶段供氧控制发酵过程中的呼吸参数在线检测分析。通过呼吸参数在线检测分析,从氧消耗机制方面解释了双阶段氧传递控制工艺能获得较高生物量、油脂和DHA含量的原因,从而为该工艺过程提供了理论指导。根据发酵过程中菌体生长不同时期的呼吸参数的变化情况,建立了基于呼吸商变化的在线补料控制方法,设计了一种基于RQ-Stat的补料工艺。RQ-Stat补料方式最终获得的油脂含量、DHA产量和产率比间歇式补料工艺分别提高了11.58%、12.19%和11.40%。 相似文献

Acinar cells in acute pancreatitis (AP) die through apoptosis and necrosis, the impacts of which are quite different. Early clinical interference strategies on preventing the progress of AP to severe acute pancreatitis (SAP) are the elimination of inflammation response and inhibition of necrosis. Muscarinic acetylcholine receptor M3 was encoded by Chrm3 gene. It is one of the best-characterized receptors of pancreatic β cells and regulates insulin secretion, but its function in AP remains unclear. In this study, we explored the function of Chrm3 gene in the regulation of cell death in l -arginine-induced SAP animal models. We found that Chrm3 was upregulated in pancreatitis, and we further confirmed the localization of Chrm3 resided in both pancreatic islets and acinar cell membranes. The reduction of Chrm3 decreased the pathological lesion of SAP and reduced amylase activities in serum. Consistently, Chrm3 can suppress acinar cells necrosis markedly, but has no effect on regulating apoptosis after l -arginine treatment. It was shown that Chrm3 attenuated acinar cells necrosis at least in part by stabilizing caspase-8. Thus, this study indicates that Chrm3 is critical participants in SAP, and regulation of Chrm3 expression might be a useful therapeutic strategy for preventing pathologic necrosis. 相似文献

考察种子培养基中谷氨酸钠质量浓度和NaCl质量浓度(盐度)对Schizochytrium sp. HX-308菌种质量及发酵性能的影响。结果表明:40 g/L谷氨酸钠条件下培养菌种,发酵后DHA质量分数达到(53.25±0.48)%,而其油产量、油脂质量分数和DHA产量分别为(24.9±0.4)g/L、(45.2±0.6)%、(8.6±0.35)g/L; 利用不含谷氨酸钠的种子培养基发酵,其油产量、油脂质量分数和DHA产量分别为(30.1±0.8)g/L、(54.3±0.5)%、(12.1±0.5)g/L,比含40 g/L谷氨酸钠时分别提高了20.88%、20.13%和40.70%,为菌种驯化提供了新的思路和方向。此外,低盐度培养条件有利于菌种生产性能的发挥,6.25 g/L NaCl条件下发酵后油质量分数、DHA产量及DHA质量分数最高,分别为(62.1±0.8)%、(10.0±0.3)g/L、(48.97±0.42)%。 相似文献

考察保护剂、保藏温度及预冷冻方法对Schizochytrium sp.HX-308菌种存活率及发酵性能保持的影响。结果显示:在-80℃低温保藏6个月后,渗透性保护剂的细胞存活率均比非渗透性保护剂高了5%,其中用60%(质量分数)海藻糖的保护剂最终的株细胞存活率达到80.02%,明显优于其他保护剂。采用液氮-196℃保藏菌种(两步预冷冻法、60%海藻糖保护剂),存储6个月后存活率高达90.70%,生物量、油产量和二十二碳六烯酸(DHA)产量分别达到了61.65、26.41和11.10 g/L,为最优的保藏方法,为裂殖壶菌的实验室研究及工业化生产提供了一种长期安全的保藏法。 相似文献

We have investigated the mycelial growth and extracellular polysaccharide (EPS) production of Stropharia rugosoannulata #2 under optimal culture conditions in a 5-L stirred-tank fermenter. Maximum biomass growth (16.35 g/L) was achieved after 5 days of cultivation, whereas EPS reached its maximum level (10.83 g/L) after 8 days. The morphological parameters (i.e., mean diameter, circularity, roughness, compactness) of the fungal pellets and broth viscosity were also characterized. The compactness of the pellets was determined to be significantly and positively correlated with EPS content. The hypoglycemic effect of the polysaccharide, investigated in streptozotocin-induced diabetic rats, included decreases in the plasma concentrations of glucose (37 %), total cholesterol (26 %), and triacylglycerol (24 %) and decreased aspartate aminotransferase activity (20 %). The results indicate the potential of this polysaccharide to prevent hyperglycemia in diabetic patients. 相似文献

As an important method to accurately and timely diagnose stroke and study physiological characteristics and pathological mechanism in it, imaging technology has gone through more than a century of iteration. The interaction of cells densely packed in the brain is three-dimensional (3D), but the flat images brought by traditional visualization methods show only a few cells and ignore connections outside the slices. The increased resolution allows for a more microscopic and underlying view. Today''s intuitive 3D imagings of micron or even nanometer scale are showing its essentiality in stroke. In recent years, 3D imaging technology has gained rapid development. With the overhaul of imaging mediums and the innovation of imaging mode, the resolution has been significantly improved, endowing researchers with the capability of holistic observation of a large volume, real-time monitoring of tiny voxels, and quantitative measurement of spatial parameters.
