An international effort is underway to establish a representative system of marine protected areas (MPAs) in the Southern Ocean to help provide for the long-term conservation of marine biodiversity in the region. Important to this undertaking is knowledge of the distribution of benthic assemblages. Here, our aim is to identify the areas where benthic marine assemblages are likely to differ from each other in the Southern Ocean including near-shore Antarctica. We achieve this by using a hierarchical spatial classification of ecoregions, bathomes and environmental types. Ecoregions are defined according to available data on biogeographic patterns and environmental drivers on dispersal. Bathomes are identified according to depth strata defined by species distributions. Environmental types are uniquely classified according to the geomorphic features found within the bathomes in each ecoregion. We identified 23 ecoregions and nine bathomes. From a set of 28 types of geomorphic features of the seabed, 562 unique environmental types were classified for the Southern Ocean. We applied the environmental types as surrogates of different assemblages of biodiversity to assess the representativeness of existing MPAs. We found that 12 ecoregions are not represented in MPAs and that no ecoregion has their full range of environmental types represented in MPAs. Current MPA planning processes, if implemented, will substantially increase the representation of environmental types particularly within 8 ecoregions. To meet internationally agreed conservation goals, additional MPAs will be needed. To assist with this process, we identified 107 spatially restricted environmental types, which should be considered for inclusion in future MPAs. Abstract

Ethnic and religious minority identity is a subject of intense public debate and academic scrutiny. While assimilation theories anticipate convergence of identity across the generations, discussions of reactive ethnicity, transnational identification and religious revival suggest that there may be a deepening or shifting of minority identity in the second generation. Yet the empirical evidence in support of these different perspectives is far from conclusive. Drawing on a rich data source for the UK, this paper addresses the question of whether minority ethnic groups in Britain show identity assimilation in the second generation. It concludes that both public and private forms of identification with the majority increase across generations, and minority identities tend to become less salient. This is true across ethnic groups, although there are differences in underlying levels and patterns of identity, reflecting variation in contexts of reception and migration.  相似文献

The bead sizes used in approved modified release capsules labeled for sprinkling on food was investigated to generate bead size guidelines for generic products labeled for sprinkling. The conclusions from a survey of FDA databases were corroborated with experimental data obtained by measuring the bead sizes of several reference-listed drugs on the market labeled for administration by sprinkling on food. The experimental data show that majority of the marketed products were found to have bead sizes of less than 1,500 microm (1.5 mm). Based on this information, a bead size of less than 1,500 microm should generally be considered acceptable for use in generic products labeled for sprinkling.  相似文献

The general stress resistance of Escherichia coli is controlled by the RpoS sigma factor (phi(S)), but mutations in rpoS are surprisingly common in natural and laboratory populations. Evidence for the selective advantage of losing rpoS was obtained from experiments with nutrient-limited bacteria at different growth rates. Wild-type bacteria were rapidly displaced by rpoS mutants in both glucose- and nitrogen-limited chemostat populations. Nutrient limitation led to selection and sweeps of rpoS null mutations and loss of general stress resistance. The rate of takeover by rpoS mutants was most rapid (within 10 generations of culture) in slower-growing populations that initially express higher phi(S) levels. Competition for core RNA polymerase is the likeliest explanation for reduced expression from distinct promoters dependent on phi(70) and involved in the hunger response to nutrient limitation. Indeed, the mutation of rpoS led to significantly higher expression of genes contributing to the high-affinity glucose scavenging system required for the hunger response. Hence, rpoS polymorphism in E. coli populations may be viewed as the result of competition between the hunger response, which requires sigma factors other than phi(S) for expression, and the maintenance of the ability to withstand external stresses. The extent of external stress significantly influences the spread of rpoS mutations. When acid stress was simultaneously applied to glucose-limited cultures, both the phenotype and frequency of rpoS mutations were attenuated in line with the level of stress. The conflict between the hunger response and maintenance of stress resistance is a potential weakness in bacterial regulation.  相似文献

Assessing how bats respond to habitat attributes requires an integrative approach to reliably predict direct community-level effects. We focused on hipposiderid and pteropodid bats because of their diverse resource use patterns, body size ranges, and dispersal abilities. We combined an array of bat species-level characteristics with key forest stand characteristics that may covary with habitat use. Twelve stations were sampled in the Lomami and Yangambi landscapes, Democratic Republic of the Congo. We investigated whether species-level flight ability of bats and forest stand characteristics can affect bat commuting flights and community-level estimates of both species detection and habitat occupancy. We captured bats for 108 trap-nights. Three sampling events (early evening, middle of the night, and early morning) were replicated for each survey night. Hipposiderids showed an early evening flight peak, while flight activity of pteropodids was constant throughout the night, but increased around the middle of the night. Species capture probability decreased with higher wing loading in hipposiderids and was negatively correlated with higher wing aspect ratio in pteropodids. Forest occupancy of hipposiderids increased along the gradient towards waterways, while pteropodid occurrence was not directly linked to measured forest stand variables. This suggests a consequence of habitat patterns at larger spatial scales, which would need clarifying through additional data collection. We discuss these findings in terms of resource-use strategies of clutter-tolerant and clutter-intolerant species. We argue that the occurrence of specific bat species and their habitat use patterns can serve as surrogate measures of ecosystem health.  相似文献

Surfactant formulations used in chemical dispersing agents are derived from the same functional components used in numerous household products such as dishwashing soaps and laundry detergents. During the Deep Water Horizon (DWH) oil spill response, a significant volume of chemical dispersant was deployed, causing members of the public and the media to question the role of chemical dispersant (Corexit 9500) usage in mitigating oil spill effects. Consequently, laboratory tests were conducted by regulatory agencies to further evaluate and substantiate the existing aquatic toxicity of Corexit dispersants. To help put dispersant toxicity in context, two independent accredited labs were commissioned to conduct parallel studies that compared the acute toxicity of Corexit 9500 to common household cleaning agents. The results indicate that the acute toxicity of Corexit 9500 to marine aquatic organism is either within the median range or less toxic than the household cleaning agents tested. The median LC50 value for Corexit 9500 exposures to Americamysis bahia was 42.5 mg/L (four products were less toxic and four products were more toxic); whereas, the median LC50 value for Corexit 9500 exposures to Menidia beryllina was 73.1 mg/L (one product was less toxic and seven products were more toxic).  相似文献

The low induction rates of somatic embryogenesis (SE) in Eucalyptus globulus hamper scaling up the process for commercialization. We analyzed the effectiveness of several media (MS, 1/2MS, B5, WPM, DKW and JADS) during SE induction and expression. MS and B5 were the best media for SE induction and embling regeneration. In general, MS was the best medium for expression, independently of the medium previously used during induction. Several anti-browning compounds (ascorbic acid, charcoal, DTE, DTT, PVP, PVPP and silver nitrate) were added to the expression medium (MS), but all decreased SE potential and only DTE, charcoal and silver nitrate reduced explant browning. When added only during the induction period, anti-browning agents reduced accumulation of phenolics but also severely reduced SE potential. Continuous exposure completely inhibited the SE response. The negative impact of anti-browning agents on SE potential raises a question about the role of production/accumulation of phenolics in the SE process.  相似文献

Abstract Site-directed mutagenesis of the lamB gene was used to introduce individual cysteine substitutions at 20 sites in two regions (surface loops L7 and L8) of LamB protein significant in antibody recognition. Characterisation of cysteine mutants involved immunoblotting with three surface-specific monoclonal antibodies (mAb72, mAb302, mAb347) before and after incubation with thiol-specific reagents. In contrast to an earlier study that showed no amino acid changes affecting recognition by all three antibodies, changes at six amino acids were found to influence a common core epitope. These core sites included one residue (T336) in the predicted loop L7 containing amino acids 329–342 and four (Y379, N387, N389, K392, F398) in the large surface loop involving residues 370–412. Individual antibodies made additional but distinct contacts within the two studied regions, with mAb347 binding the most different and affected by seven substitutions in the 328–338 regions. The lamB mutants were also tested for phage λ receptor activity and starch binding before and after thiol modification and were useful in extending previous maps of these ligand binding sites.  相似文献

The yeast translational elongation factor 3 (EF-3) stimulates EF-1 alpha-dependent binding of aminoacyl-tRNA by the ribosome. The requirement for EF-3 is unique to fungi; a functional analog has not been found in prokaryotes or other eukaryotes. We have isolated and characterized the structural gene, YEF3, that encodes EF-3. The YEF3 gene is present in one copy/haploid genome and is essential for vegetative growth. DNA sequence analysis revealed that the YEF3 gene contains an open reading frame of 1044 codons. The deduced amino acid sequence contains two repeats of a nucleotide-binding motif, which is similar to the nucleotide-binding consensus sequences of hydrophilic, membrane-associated ATPases. EF-3 catalyzes ATP hydrolysis in a ribosome-dependent manner. A modified assay procedure has been developed that allows measurement of the ATP hydrolytic activity of EF-3 in cell-free extracts without interference by other nucleotide hydrolyase activities. Using this modified assay, we have shown that the wild-type YEF3 gene restores heat stable EF-3 activity in a yeast strain containing a temperature-sensitive EF-3. Introduction of the YEF3 gene on a high copy number plasmid into yeast strains increases the ribosome-dependent ATPase activity. The level of EF-3 protein is also increased 3-5-fold. Elevated EF-3 protein levels did not cause a significant increase in EF-1 alpha and EF-2 protein. Yeast strains containing elevated EF-3 protein levels are more sensitive to the aminoglycoside antibiotics hygromycin and paromomycin. These drugs are known to increase translational errors. This observation suggests that EF-3 may indirectly affect translational accuracy.  相似文献

The described protocol for repetitive somatic embryogenesis (SE) in Eucalyptus globulus produced more somatic embryos than the primary SE protocol. Primary somatic embryos (induced on MS3NAA) were transferred to the same medium, leading to new cycles of somatic embryos, for at least 2 years. The influence of medium (MS and B5), plant growth regulators (auxins and cytokinins), and light on secondary SE was tested. The MS medium without growth regulators (MSWH) was more efficient for cotyledonary embryo formation and germination than the B5 medium. Reducing auxin (NAA) levels increased the proliferation of globular somatic embryos and allowed SE competence to be maintained on medium free of plant growth regulators. The addition of two cytokinins (BAP and KIN) to the MS medium did not improve proliferation of globular secondary embryos, but was crucial during later stages of the SE process (germination and conversion). Data also show that light may influence the quality of the process, depending on its stage. Darkness should be maintained until the cotyledonary stage is reached, after which exposure to light is recommended.  相似文献
