We conducted three experiments to test the effects of mating history of both sexes and of male body size on mating behaviours in the water strider, Gerris buenoi. Our manipulations influenced the interests of both sexes and, thus, the degree of conflict over mating behaviours. Mating history was a dichotomous variable (deprived/mated), depending on holding conditions in the laboratory. Experiment 1 considered and found independent effects of male and female mating history on latency to copulation and copulation duration. In experiment 2, we manipulated only female mating history, using unsuccessful struggle rates as evidence for female reluctance and conflict over mating. Finally, we investigated the relation between male body size and mating history on copulation duration. We predicted that intersexual conflict over mating would be lowest when females were deprived, because female interests under these conditions should more closely match those of males. Deprived females began mating in half the time of mated females and were twice as likely to mate because of reduced reluctance. Furthermore, copulation duration for deprived males was about one and a half times longer than that for mated males. Although previous studies examining nonrandom mating patterns by size predicted longer copulations for small males, we found that small males prolonged copulation when deprived more than large males. We conclude that females primarily influence copulation frequency, but males primarily influence copulation duration. Our results favour the hypothesis that reduced mating opportunity for small males accounts for their extended copulation duration. Finally, our findings provide evidence for strong effects of male body size on selection mechanisms in water striders, and support the hypothesis of conflicting pre- and postcopulatory selection mechanisms in this group. The oral route has notable advantages to administering dosage forms. One of the most important questions to solve is the poor solubility of most drugs which produces low bioavailability and delivery problems, a major challenge for the pharmaceutical industry. Albendazole is a benzimidazole carbamate extensively used in oral chemotherapy against intestinal parasites, due to its extended spectrum activity and low cost. Nevertheless, the main disadvantage is the poor bioavailability due to its very low solubility in water. The main objective of this study was to prepare microcrystal formulations by the bottom-up technology to increase albendazole dissolution rate, in order to enhance its antiparasitic activity. Thus, 20 novel microstructures based on chitosan, cellulose derivatives, and poloxamer as a surfactant were produced and characterized by their physicochemical properties and in vitro biological activity. To determine the significance of type and concentration of polymer, and presence or absence of surfactant in the crystals, the variables area under the curve, albendazole microcrystal solubility, and drug released (%) at 30 min were analyzed with a three-way ANOVA. This analysis indicated that the microcrystals made with hydroxyethylcellulose or chitosan appear to be the best options to optimize oral absorption of the active pharmaceutical ingredient. The in vitro evaluation of anthelmintic activity on adult forms of Trichinella spiralis identified system S10A as the most effective, of choice for testing therapeutic efficacy in vivo.  相似文献

ABSTRACT

Nanomaterials (Nms) applications and environmental deposition are continuously increasing. Aluminum (Al) and nickel (Ni) fate in soil, both from gamma alumina-based Nms and as chloride salts were evaluted through lysimeters. After 85 days of treatment, which included irrigation and collection of eluates, the soil of each lysimeter was divided into four sections. The metal concentration was analyzed in eluates, soil samples, and extracts. Al and iron (Fe) present in soil eluted from Control lysimeter. Al from Nms suspension treatment was quantified in the eluates since 30 days on. Ni eluted upon solid salt deposition on top of one device. These results indicate that Al and Ni applied under certain conditions on soil, could leach and reach groundwater. The total concentration and bioavailability (extractable metals) of Al and Fe in soils showed similar patterns. Ni was retained only in the soil of devices treated with chloride salts. Bioavailability % results were of concern for Ni under certain conditions of treatment: 15.57% and 11.08% in two chloride salt-treated lysimeters versus 0.55% and 0.47% of those in control and treated with Nms lysimeters. Conducting studies with different kinds of soil and longer treatment periods should be useful to understand Nms-metals fate in the environment. The results presented here constitute important evidences both for significant metal release from Nms and elution and for considerable Ni bioavailability, after deposition on soil in the form of Nms or as a chloride salt, respectively. Then, possible toxic effects could occur through exposure of aquatic and terrestrial organisms.  相似文献

Immunochemical Characterization of Antisera to Rat Neurofilament Subunits   总被引:14,自引:7,他引:7  
Abstract: Antisera raised to the 68,000, 145,000 and 200,000 molecular weight subunits of rat neurofilaments were used for immunochemical staining of polypeptides separated by one- and two-dimensional gel electrophoresis. It was found that each antiserum reacts intensely with its corresponding neurofilament subunit and weakly with the other two subunits. All the antisera also react with a polypeptide of molecular weight 57,000 present in neurofilament-rich preparations from both rat spinal cord and peripheral nerve. This polypeptide is different from either tubulin or vimentin and may represent a neurofilament breakdown product, since it varied in amount from preparation to preparation. The three antisera also reacted with the polypeptide subunits of chicken and goldfish neurofilament despite the considerable difference in molecular weight between these subunits and those of mammalian neurofilament. Phenolic compounds have antioxidant and anti-inflammatory properties and may prevent inflammation and oxidative stress as well as help the athletes to recover from exercise-induced muscle damage (EIMD). Tart cherry (TC) and pomegranate (PG) are two fruits with high content of polyphenols. Their antioxidant and anti-inflammatory properties have recently attracted substantial interest for their potential to reduce strength loss and promote recovery from EIMD. The aims of this review are (1) to summarise the effects of tart cherry and pomegranate supplementation on oxidative stress, inflammation and recovery, and (2) to outline the differences found in supplementation with tart cherries or pomegranates. SPORTDiscus, PubMed, Web of Science and Scopus were searched according to Preferred Reporting Items for Systematic Reviews and Meta-Analysis and 25 studies were included. The existing evidence suggests that both types of supplementation are good strategies to accelerate recovery of functional performance variables, perceptual variables and inflammation but PG supplementation shows better recovery of oxidative stress. However, positive effects are more likely: 1) when supplementation starts some days before muscle damage is induced and finishes some days after, for a total period of at least 8/10 days, 2) with pronounced muscle damage of the muscles involved, and 3) when total phenolic content is at least 1000 mg/day. This review may help to optimise TC or PG supplementation practice to improve post-exercise recovery.  相似文献

In this study, we investigated expression and dimerization of an ER-associated degradation (ERAD) substrate, a null Hong Kong variant of α-1-antitrypsin (NHK) using immunoblotting assay and a novel NanoLuc complementary reporter system called the NanoBiT (NB) assay. This NB-tagged NHK made it possible to monitor the intra- and extracellular status of NHK in living cells. The values for this NB assay fluctuated in response to distinct pharmacological stimuli and co-transfection of several ERAD-related factors. We then focused on mesencephalic astrocyte-derived neurotrophic factor (MANF), an unclarified ATF6/IRE1-downstream target, and established MANF-deficient Neuro2a (N2a) cells using CRISPR/Cas9 system. MANF-deficient N2a significantly elevated OS-9 protein after tunicamycin treatment; however, no specific differences in intra- and extracellular status of NHK protein were observed between wild-type and MANF-deficient cells. Taken together, intrinsic MANF in N2a cells is not strongly associated with the accumulation and clearance of unfolded proteins within the ER under current condition, but this novel NB assay is a useful approach for characterizing the protein status including ERAD substrates.  相似文献

We designed PCR primers by comparison of the deduced amino acid sequences of several ornithine decarboxylase (ODC) genes. They were used to amplify fragments homologous to these genes from several dimorphic fungi. These were sequenced and the deduced amino acid sequences were compared with the corresponding regions of ODCs from different sources. Fungal ODCs fell into a compact group, well separated from the ODCs of other taxa. Sequence homology among fungal enzymes corresponded to their taxonomic position. Interesting patterns of amino acid conservation in ODCs from fungi, distinct from other organisms, were detected. Received: 29 May 1996 / Accepted: 29 June 1996  相似文献

This paper reports the relationship among pico-, nano- and microplanktoniccommunities observed in two different shallow marine hydrothermalenvironments. Seawater samples from five stations in the coastalarea of the ‘Porto di Levante’ (Vulcano Island)and from three stations off the Island of Panarea (Eolian Islands,Italy) were collected in May and July of 1989, respectively.Microbiological investigations were carried out in order todetermine the density of: (i) the total picoplankton (both autotrophicand heterotrophic): (ii) the total picophytoplankton (autofluorescentpicoplanktonic cells); (iii) the larger phytoplankton (>2µm): (iv) the ‘metabolically active’ cellsof total picoplankton and cyanobacteria; (v) the heterotrophicaerobic bacteria. The peak values of picoplankton and picophytoplanktoncomponents, with an order of magnitude of 109 and 107 cells11, respectively, were registered in the wannest water samples(30–75°C) collected from the Vulcano area. At Panarea.eukaryotic picophytoplankton and ‘metabolically active’coccoid cyanobacteria showed an opposite trend. A possible competitionbetween the two groups is to be considered. Cyanobacteria, diatomsand the genus Licmophora. in particular, were prevalent in thewarmest hydrothermal vents of Vulcano. Nano- and microphytoplanktoniccommunities in the offshore waters of the Island of Panareashowed more variability than in Vulcano. Moreover, in the Panareawaters the prevalence of phytoflagellates above the thermoclinewas observed, whereas diatoms were predominant below this layer.  相似文献
