Some reactional parameters as mol ratio (salicylaldehyde:free amino groups), reaction time and temperature were investigated in order to improve the substitution degree (DS) in the preparation of biopolymeric Schiff bases from chitosan. In this case, the reaction of chitosan and salicylaldehyde was used as a probe system in order to produce the Schiff base. The use of 50% (mol/mol) salicylaldehyde excess, reaction time of 18 h and temperature of 55 °C permitted to obtain a DS of 60% without evidences of hydrolysis of the biopolymeric matrix or changes in its acetylation degree. 相似文献

Tiger sharks (Galeocerdo cuvier) are apex predators occurring in most tropical and warm temperate marine ecosystems, but we know relatively little of their patterns of residency and movement over large spatial and temporal scales. We deployed satellite tags on eleven tiger sharks off the north-western coast of Western Australia and used the Brownian Bridge kernel method to calculate home ranges and analyse movement behaviour. One individual recorded one of the largest geographical ranges of movement ever reported for the species, travelling over 4000 km during 517 days of monitoring. Tags on the remainder of the sharks reported for shorter periods (7-191 days). Most of these sharks had restricted movements and long-term (30-188 days) residency in coastal waters in the vicinity of the area where they were tagged. Core home range areas of sharks varied greatly from 1166.9 to 634,944 km2. Tiger sharks spent most of their time in water temperatures between 23°-26°C but experienced temperatures ranging from 6°C to 33°C. One shark displayed seasonal movements among three distinct home range cores spread along most of the coast of Western Australia and generalized linear models showed that this individual had different patterns of temperature and depth occupancy in each region of the coast, with the highest probability of residency occurring in the shallowest areas of the coast with water temperatures above 23°C. These results suggest that tiger sharks can migrate over very large distances and across latitudes ranging from tropical to the cool temperate waters. This study aimed to evaluate protection induced by the vaccine candidate B. ovis ΔabcBA against experimental challenge with wild type B. ovis in rams. Rams were subcutaneously immunized with B. ovis ΔabcBA encapsulated with sterile alginate or with the non encapsulated vaccine strain. Serum, urine, and semen samples were collected during two months after immunization. The rams were then challenged with wild type B. ovis (ATCC25840), and the results were compared to non immunized and experimentally challenged rams. Immunization, particularly with encapsulated B. ovis ΔabcBA, prevented infection, secretion of wild type B. ovis in the semen and urine, shedding of neutrophils in the semen, and the development of clinical changes, gross and microscopic lesions induced by the wild type B. ovis reference strain. Collectively, our data indicates that the B. ovis ΔabcBA strain is an exceptionally good vaccine strain for preventing brucellosis caused by B. ovis infection in rams. 相似文献

Centrosome duplication must remain coordinated with cell cycle progression to ensure the formation of a strictly bipolar mitotic spindle, but the mechanisms that regulate this coordination are poorly understood. Previous work has shown that prolonged S-phase is permissive for centrosome duplication, but prolonging either G2 or M-phase cannot support duplication. To examine whether G1 is permissive for centrosome duplication, we release serum-starved G0 cells into mimosine, which delays the cell cycle in G1. We find that in mimosine, centrosome duplication does occur, albeit slowly compared with cells that progress into S-phase; centrosome duplication in mimosine-treated cells also proceeds in the absence of a rise in Cdk2 kinase activity normally associated with the G1/S transition. CHO cells arrested with mimosine can also assemble more than four centrioles (termed "centrosome amplification"), but the extent of centrosome amplification during prolonged G1 is decreased compared to cells that enter S-phase and activate the Cdk2-cyclin complex. Together, our results suggest a model, which predicts that entry into S-phase and the rise in Cdk2 activity associated with this transition are not absolutely required to initiate centrosome duplication, but rather, serve to entrain the centrosome reproduction cycle with cell cycle progression. 相似文献

A new laccase was purified from Trametes hirsuta IMA2002. The laccase had a molecular mass of 62 kDa and an isoelectric point of pH 7. It had an optimum pH of 3.0 and an optimum temperature of 55°C. The laccase was quite stable at 30°C and pH 4.0 with a half-life of more than 100 hours. On ABTS, syringaldazide, and DMP the laccase showed KM and Kcat values of 75, 12 and 37 μM and 64, 83 and 54 s-1, respectively. The structurally diverse commercial dyes Indigo Carmine, Lanaset Blue 2R, Diamond Black PV 200 and Diamond Fast Brown were oxidized by the laccase. While the rate and extent of decolorization of the latter dye was significantly enhanced by the presence of different types of mediators, the structurally similar azo-dye Tartrazine was not oxidized. Lanaset Blue 2R, a commercial textile dye containing an anthrachinoid structural fragment acted similarly to anthrachinone sulfonic acid by strongly enhancing the rate of the decolorization reaction. Twenty two model azo-dyes based on the molecular framework of 2,7-dihydroxy-1-phenylazonaphtalene-3,6-disulfonic acid were synthesized and the kinetics of their laccase-catalyzed decolorization was studied. Hydroxy-substituted dyes were the most susceptible to enzyme/mediator action. All reactions were well described by Michaelis-Menten-like kinetics and the Hammett free energy linear relationship could be successfully applied to describe the influence of dye structure (substituents on the aromatic ring) on decolorization. Strongly electron withdrawing substituents such as a nitro-group in the meta-position (+0.7) resulted in positive σ-constants whereas electron donating groups such as para-methyl (-0.3) resulted in negative values for σ-constants. 相似文献

Most tumors grow in immunocompetent hosts despite expressing NKG2D ligands (NKG2DLs) such as the MHC class I chain-related genes A and B (MICA/B). However, their participation in tumor cell evasion is still not completely understood. Here we demonstrate that several human melanomas (cell lines and freshly isolated metastases) do not express MICA on the cell surface but have intracellular deposits of this NKG2DL. Susceptibility to NK cell-mediated cytotoxicity correlated with the ratio of NKG2DLs to HLA class I molecules but not with the amounts of MICA on the cell surface of tumor cells. Transfection-mediated overexpression of MICA restored cell surface expression and resulted in an increased in vitro cytotoxicity and IFN-gamma secretion by human NK cells. In xenografted nude mice, these melanomas exhibited a delayed growth and extensive in vivo apoptosis. Retardation of tumor growth was due to NK cell-mediated antitumor activity against MICA-transfected tumors, given that this effect was not observed in NK cell-depleted mice. Also, mouse NK cells killed MICA-overexpressing melanomas in vitro. A mechanistic analysis revealed the retention of MICA in the endoplasmic reticulum, an effect that was associated with accumulation of endoH-sensitive (immature) forms of MICA, retrograde transport to the cytoplasm, and degradation by the proteasome. Our study identifies a novel strategy developed by melanoma cells to evade NK cell-mediated immune surveillance based on the intracellular sequestration of immature forms of MICA in the endoplasmic reticulum. Furthermore, this tumor immune escape strategy can be overcome by gene therapy approaches aimed at overexpressing MICA on tumor cells. 相似文献
