Egg banks in freshwater zooplankton: evolutionary and ecological archives in the sediment

Many representatives of freshwater zooplankton produce at some stage in their life cycle resting stages. A variable portion of the eggs of the previous growing period will hatch at the next occasion while the remaining ones are added to a persistent egg bank, where they can remain viable for decades or longer. The importance of the study of resting eggs and egg banks in general for such different disciplines as taxonomy, ecological biogeography, paleolimnology, nature conservation, evolutionary ecology and community and population ecology is generally appreciated. The major current and expected future developments in this rapidly expanding field of research are presented here. The structure and dynamics of the egg bank are determined by the life history characteristics of the species (or local population), the hatching phenology of their resting stages, and the characteristics of the habitat. The horizontal distribution of dormant stages is generally patchy, with a greater density in the deeper and/or windward parts of a pond or lake. In sediment cores, most viable (responsive) eggs occur in the upper centimeters, although vertical variation related to the history of fish predation or water quality occurs. The accumulation of resting stages of different species, generations and genotypes with variable regeneration niches results in a mixed egg bank with greater potential biodiversity than the active community sampled at any one moment. Through the benthic–pelagic coupling, this dormant reservoir may have considerable impact on the evolutionary potential of the organisms, the ecological dynamics of the community and the distribution of species. Egg banks can be considered the archive of the local habitat, since the pattern of changes in species assemblage and genotypes from the past up to the present reflect changes due to natural or anthropogenic impact that can be used to reconstruct evolutionary processes or even to restore the local habitat. Overlooking the egg bank as an important component of zooplankton communities may lead to erroneous interpretations in the analysis of community and population genetic structure. This review integrates technical and scientific information needed in the study of the structure and function of egg banks in zooplankton with special focus on the fascinating latest developments in the field.     

Effect of manganese on in vitro replication of damaged DNA catalyzed by the herpes simplex virus type-1 DNA polymerase

In vitro bypass of damaged DNA by replicative DNA polymerases is usually blocked by helix-distorting or bulky DNA lesions. In this study, we report that substitution of the divalent metal ion Mg²⁺ with Mn²⁺ promotes quantitative replication of model DNA substrates containing the major cisplatin or N-2-acetylaminofluorene adducts by the catalytic subunit (UL30) of the replicative DNA polymerase of herpes simplex virus. The ability of Mn²⁺ ions to confer bypass of bulky lesions was not observed with other replicative DNA polymerases of the B family, such as bacteriophage T4 or δ polymerases. However, for these enzymes, manganese induced the incorporation of one nucleotide opposite the first (3′) guanine of the d(GpG) intrastrand cisplatin lesion. Translesion replication of the cisplatin adduct by UL30 led to the incorporation of mismatched bases, with the preferential incorporation of dAMP opposite the 3′ guanine of the lesion. Furthermore, substitution of MgCl₂ with MnCl₂ greatly inhibited the 3′ to 5′ exonuclease of UL30 but had a far lesser effect on that of T4 DNA polymerase. Finally, manganese induced a conformational change in the structure of UL30 bound to the platinated substrate. Taken together, the latter findings suggest a mechanism by which manganese might allow UL30 to efficiently promote translesion DNA synthesis in vitro.     

An ovarian follicular epithelium protein of the silkworm (Bombyx mori) that associates with the vitelline membrane and contributes to the structural integrity of the follicle

We have cloned and functionally characterized a novel protein, BmVMP30, which is synthesized by the cells of the follicular epithelium of the ovarian follicles of the domesticated silkworm Bombyx mori, secreted from them and associated with the vitelline membrane. BmVMP30 is a 30 kDa protein that bears limited structural features reminiscent of other insect vitelline membrane proteins. Although BmVMP30 does not share pronounced similarities or signature motifs with other reported proteins, its temporal and spatial expression and its behavior throughout oogenesis suggest that it is a novel member of the insect vitelline membrane protein family. The protein is expressed exclusively in the cells of the follicular epithelium during stages -15 to -1 of vitellogenesis, secreted from them and, ultimately, localized at the junction between the oocyte and the eggshell, where the vitelline membrane is located. Treatment of follicles with an antisense oligonucleotide that encompasses the translation initiation codon results in the production of an N-terminally truncated protein and disruption of the integrity of the follicular epithelium. Antisense oligonucleotide treatment, however, has no effect on the implementation of the developmental program that directs the autonomous progression of ovarian follicles through the last stages of vitellogenesis and choriogenesis.     

Expedient syntheses of neoglycoproteins using phase transfer catalysis and reductive amination as key reactions

Starting from peracetylated chloro- or bromo-glycosyl donors ofN-acetylneurmainic acid,N-acetylglucosamine, glucose and lactose, the correspondingp-formylphenyl glycosides were synthesized stereospecifically under phase transfer catalysed conditions at room temperature in yields of 38–67%. After Zemplén de-O-acetylation, the formyl groups were directly and chemoselectively coupled to the lysine residues of bovine serum albumin by reductive amination using sodium cyanoborohydride. The conjugation reactions were followed as a function of time and under a series of different molar ratios of the reactants to provide glycoconjugates of varying degree of antigenicities. Thus, carbohydrate protein conjugates were made readily available using essentially two key reactions.Presented in part at the 15th International Carbohydrate Symposium, Yokohama, Japan, August 12–17, 1990.     

No evidence for a tumor necrosis factor α stimulated 2-methylaminoisobutyric acid uptake in hepatocyte monolayer

This study investigates the short-tem effects of glucagon and human recombinant tumor necrosis factor α (TNFα) singly and in association on 2-methylaminoisobutyric acid (MeAIB) transport in hepatocyte monolayers. As expected, glucagon induced a time-dependent stimulation of MeAIB transport. In our experimental conditions, TNFα did not induce cytolysis. A 2 hour exposure to TNFα (0.05–500 ng/I) with or without glucagon (10^?9 to 10^?6 M) did not modify the basal or glucagon-stimulated MeAIB transport. Varying the duration of exposure to TNFα 5 ng/I up to 6 h was equally ineffective. The presence of hydrocortisone potentiated the glucagon-stimulated transport, but TNFα remained ineffective. Finally, the association of interferon (IFNγ) with TNFα and/or glucagon was unable to modify the transport activity. These data demonstrate that TNFα does not exert a direct effect on MeAIB transport in hepatocytes, at least on a short-term basis. © 1995 Wiley-Liss, Inc.     

Real time noninvasive assessment of external trunk geometry during surgical correction of adolescent idiopathic scoliosis

Retrospective study.

Objective

To evaluate the radiological results of fusion with segmental pedicle screw fixation in juvenile idiopathic scoliosis with a minimum 5-year follow-up.

Summary of background data

Progression of spinal deformity after posterior instrumentation and fusion in immature patients has been reported by several authors. Segmental pedicle screw fixation has been shown to be effective in controlling both coronal and sagittal plane deformities. However, there is no long term study of fusion with segmental pedicle screw fixation in these group of patients.

Methods

Seven patients with juvenile idiopathic scoliosis treated by segmental pedicle screw fixation and fusion were analyzed. The average age of the patients was 7.4 years (range 5–9 years) at the time of the operation. All the patients were followed up 5 years or more (range 5–8 years) and were all Risser V at the most recent follow up. Three dimensional reconstruction of the radiographs was obtained and 3DStudio Max software was used for combining, evaluating and modifying the technical data derived from both 2d and 3d scan data.

Results

The preoperative thoracic curve of 56 ± 15° was corrected to 24 ± 17° (57% correction) at the latest follow-up. The lumbar curve of 43 ± 14° was corrected to 23 ± 6° (46% correction) at the latest follow-up. The preoperative thoracic kyphosis of 37 ± 13° and the lumbar lordosis of 33 ± 13° were changed to 27 ± 13° and 42 ± 21°, respectively at the latest follow-up. None of the patients showed coronal decompensation at the latest follow-up. Four patients had no evidence of crankshaft phenomenon. In two patients slight increase in Cobb angle at the instrumented segments with a significant increase in AVR suggesting crankshaft phenomenon was seen. One patient had a curve increase in both instrumented and non instrumented segments due to incorrect strategy.

Conclusion

In juvenile idiopathic curves of Risser 0 patients with open triradiate cartilages, routine combined anterior fusion to prevent crankshaft may not be warranted by posterior segmental pedicle screw instrumentation.
