Restriction-map variation associated with the G6PD polymorphism in natural populations of Drosophila melanogaster 总被引:10,自引:0,他引:10
Restriction-map variation was studied in 126 copies of the G6pd region in X
chromosome lines of Drosophila melanogaster from North America, Europe, and
Africa. Special attention was focused on the distribution of variation
relative to the geographically variable polymorphism for two
electrophoretic variants. Nucleotide heterozygosity as determined by eight
six-cutter restriction enzymes for the 13-kb region is estimated, on the
basis of the worldwide sample, to be 0.065%, which is the lowest value
reported for any comparable region in the D. melanogaster genome.
Significant linkage disequilibrium between electrophoretic alleles and
restriction-site variation is observed for several sites. In contrast to
published studies of other genetic regions, there are large insertions that
reach significant frequencies and are found across considerable geographic
distances. There is a clustering of this variation inside the first large
intervening sequence of the G6PD gene.

Cell surface interactions with concanavalin A : Location of bound radiolabeled lectin 总被引:4,自引:0,他引:4
We have developed two improved methods: (1) a procedure for coupling 125Iodine to ConcanavalinA (ConA) that yields intensely labeled and fully active lectin; (2) a procedure that allows studies of lectin binding to be carried out with a minimum of non-specific binding to reaction vessels. We found that BALB/c 3T3 cells, SV3T3 cells, and human red blood cells have 1.3 × 107, 1.5 × 107, and 2.2 × 106 ConA binding sites/cell. More than 99.5% of the radioactivity in the samples counted was associated with the cells; background radioactivity, in the absence of cells, was negligible. We also found that although α-methylmannopyranoside (α-MM) prevented almost all of the ConA from binding to cells, when ConA had first been allowed to bind, α-MM removed only 60 to 80% of the bound ConA. In addition, even after the removal of a portion of bound lectin by α-MM, most, if not all, of the remaining cell-associated ConA was coupled to the plasma membrane. 相似文献

Rhodamine 123, a laser dye, has been demonstrated to inhibit import of the precursor to pyridine dinucleotide transhydrogenase into mitochondria in rat liver cells. When rat hepatocytes were labeled with 35[S] methionine in the presence of 0.4 mM rhodamine 123, the precursor to transhydrogenase was found to have a half-life in the cytoplasm of 15 minutes as opposed to a half-life of 1-2 minutes when cells were radiolabeled in the absence of the dye. To clarify the mechanism of import inhibition, studies were initiated to assess the effect of rhodamine 123 on mitochondrial respiration. Upon addition of the dye to a mitochondrial suspension, respiration was initially enhanced, then inhibited. The inability of FCCP, a classical uncoupler, to enhance respiration during the inhibitory phase suggests that rhodamine 123 is primarily inhibiting respiration through the electron transport system rather than through the ATPase. These results suggest that rhodamine 123 may inhibit import of the transhydrogenase precursor into mitochondria by disrupting components in the mitochondrial membrane necessary for efficient import. 相似文献

The ultrastructure of the gill primary lamellae of juvenile Atlantic salmon was examined during the parr-smolt transformation and for 42 days after smolts were exposed to sea water. Scanning electron microscopy indicated that primary lamellae were characterized by rough convoluted surfaces that became rougher throughout the experimental period and that crypts did begin to form in freshwater fish. Crypt formation increased in sea water.
Transmission electron microscopy indicated that parr preadapt for life in sea water in part by changes in chloride cells. Chloride cells show an elaboration of rough endoplasmic reticulum in fresh water and a decline of rough endoplasmic reticulum after 42 days of sea water exposure. The tubular membrane system becomes well developed in fresh water, and apical vesicles become abundant only after seawater exposure. Mitochondria are both spherical and elongate through the period and contain well developed cristae. No evidence of mitochondrial rupture was observed. The junctions between chloride cells and adjacent cells were characterized in fresh water by long tight junctions with desmosomes. This type of junction continued in sea water and was the norm between chloride cells and accessory cells after 42 days of seawater exposure. While leaky junctions appeared to be forming, no evidence was found of membrane interdigitation between accessory cells and chloride cells after 42 days of seawater exposure. It also appeared that seawater exposure influenced the number of chloride cells exposed to the external milieu.
