A series of new benzolactam derivatives was synthesized and the derivatives were evaluated for their affinities at the dopamine D1, D2, and D3 receptors. Some of these compounds showed high D2 and/or D3 affinity and selectivity over the D1 receptor. The SAR study of these compounds revealed structural characteristics that decisively influenced their D2 and D3 affinities. Structural models of the complexes between some of the most representative compounds of this series and the D2 and D3 receptors were obtained with the aim of rationalizing the observed experimental results. Moreover, selected compounds showed moderate binding affinity on 5-HT2A which could contribute to reducing the occurrence of extrapyramidal side effects as potential antipsychotics. 相似文献
A series of 8 new tetrahydroquinazolinone derivatives was synthesized and evaluated for binding affinity to D2 and 5-HT2A human receptors; in addition, some properties related to blood–brain barrier penetration were calculated. From the results of these assays, three compounds were selected for further binding tests on D1, D3, and 5-HT2C human receptors, which are thought to be involved in schizophrenia. From these data, compound 19b emerged as the most promising candidate based on its good binding affinities for D1, D2, and D3 receptors, high affinity for 5-HT2A, low affinity for 5-HT2C receptors, and a Meltzer’s ratio characteristic of an atypical antipsychotic profile. 相似文献
Microthrombosis is often observed in lupus nephritis (LN) lesions, but its clinical significance is unknown. We evaluated the clinicopathologic correlations of renal microthrombosis and inflammatory markers in LN.
Methods
Kidney biopsies from 58 patients with systemic lupus erythematosus (SLE) proliferative nephritis were analyzed with immunohistochemistry (IHC) for intravascular platelet aggregates (CD61), macrophagic infiltration (CD68), and activated complement deposition (C4d). Clinical data at the time of kidney biopsy and follow-up were analyzed with regard to pathologic IHC data.
Results
Microthrombosis was present in 52% of the tissues. It was significantly more prevalent in patients with antiphospholipid antibodies (aPLs) (62% versus 42%). The presence of microthrombosis significantly correlated with higher macrophagic infiltration. Macrophagic infiltration but not microthrombosis was significantly correlated with C4d deposition. Only macrophagic infiltration showed a correlation with SLE and renal activity (proteinuria and active sediment), whereas neither the presence of CD61+ microthrombi nor the extent of C4d deposition correlated with LN severity or outcome.
Conclusions
Microthrombosis is associated with higher macrophagic infiltration in LN but does not seem to increase independently the severity of renal damage. Macrophagic infiltration was the best marker of SLE and renal activity in this LN series. 相似文献
A number of 1,3-dialkyl-9-deazaxanthines (9-dAXs), bearing a variety of N-substituted benzyloxycarbonylamino substituents at position 8, were prepared and evaluated for their binding affinity to the recombinant human adenosine receptors (hARs), chiefly to the hA2B and hA2A AR subtypes. Several ligands endowed with excellent binding affinity to the hA2B receptors, but low selectivity versus hA2A and hA1 were identified. Among these, 1,3-dimethyl-N-3′-thienyl carbamate 15 resulted as the most potent ligand at hA2B (Ki = 0.8 nM), with a low selectivity versus hA2A (hA2A/hA2B = 12.6) and hA1 (hA1/hA2B = 12.5) and a higher selectivity versus hA3 (hA3/hA2B = 454). When tested in functional assays in vitro, compound 15 exhibited high antagonist activities and efficacies versus both the A2A and A2B receptor subtypes, with pA2 values close to the corresponding pKis. A comparative analysis of structure–affinity and structure–selectivity relationships of the similar analogues 8-N-substituted benzyloxycarbonylamino- and 8-N-substituted phenoxyacetamido-9-dAXs suggested that their binding modes at the hA2B and hA2A ARs may strongly differ. Computational studies help to clarify this striking difference arising from a simple, albeit crucial, structural change, from CH2OCON to OCH2CON, in the para-position of the 8-phenyl ring. 相似文献
Raman images were used to study the effect of the contaminant chlorpyriphos‐oxon on zebrafish eye samples. Multivariate Curve Resolution‐Alternating Least Squares (MCR‐ALS) was used to obtain the distribution maps and spectral signatures of biological components present in the images analyzed. The use of MCRALS spectral signatures as starting information for Partial Least Squares‐Discriminant Analysis allowed statistical assessment of the effect of the contaminant at a specific tissue level. Patterns of chloroplast DNA (cpDNA) and mitochondrial DNA (mtDNA) variation
were studied in 378 populations of oak trees sampled throughout the
southern half of France. Six cpDNA haplotypes detected in a previous
European survey and three new cpDNA haplotypes were found in this region.
Two mitochondrial polymorphisms detected earlier by restriction analysis of
PCR-amplified fragments alone, or in combination with single-strand
conformation polymorphism (SSCP), were compared with the cpDNA data.
Sequencing revealed the nature of the two mitochondrial mutations: a
single-base substitution and a 4-bp inversion associated with a 22-bp
hairpin secondary structure. The single-base substitution was then analyzed
by allele-specific amplification. Results for the two cytoplasmic genomes
were combined, which allowed the identification of 12 cpDNA-mtDNA
haplotypes. The 4-bp mtDNA inversion has appeared independently in
different cpDNA lineages. Given the peculiar nature of this mtDNA mutation,
we suggest that intramolecular recombination leading to repeated inversions
of the 4-bp sequence (rather than paternal leakage of one of the two
genomes) is responsible for this pattern. Furthermore, the geographic
locations of the unusual cpDNA-mtDNA associations (due to the inversion)
usually do not match the zones of contact between divergent haplotypes. In
addition, in southern France, the groupings of populations based on the
mtDNA substitution were strictly congruent with those based on cpDNA.
Because many populations that are polymorphic for both cpDNA and mtDNA have
remained in contact since postglacial recolonization in this area without
producing any new combination of cytoplasms involving the mitochondrial
substitution, we conclude that paternal leakage is not a significant factor
at this timescale. Such results confirm and expand our earlier conclusions
based on controlled crosses.
