We report that human peripheral NK cells expressing high CD56 levels (CD56(+high)) are terminally differentiated cells indistinguishable from mature NK cells recently activated in the presence of IL-12, and not a functionally distinct NK-cell subset or progenitors to mature CD56(+low) NK cells. CD56(+high) NK cells coexpress all differentiation Ags constitutive or inducible in mature (CD56(+)) NK cells, except CD16, present at lower level than on most mature NK cells. Also, activation markers, activating receptors and adhesion molecules, and most inducible receptors are expressed exclusively and constitutively and are inducible at higher levels on CD56(+high) than on CD56(+low) NK cells. Consistent with their activated phenotype, many CD56(+high) NK cells are cycling and mediate heightened effector functions (proliferation, IFN-gamma and IL-10 but not IL-13 production) in response to IL-12 and other NK cell-specific stimuli. Conversely, IL-12 induces on CD56(+low) NK cells all markers constitutively expressed on the CD56(+high) NK cells, concomitantly preventing the IL-2 (and IL-15)-inducible expression of NKp44 and CD16 re-expression after immune complex-induced down-modulation, and CD56(-/+low) NK cells acquire a CD56(+high) NK cell phenotype in short term in vitro culture with IL-12. The significance of these findings to the NK cell-mediated regulation of immune responses and NK cell development is discussed.  相似文献

A series of pyrazolo[1',5':1,6]pyrimido[4,5-d]pyridazin-4(3H)-ones was synthesized and tested in radioligand binding assays to determine their affinities for the human adenosine A(1), A(2A), A(2B) and A(3) receptors. Results indicated that this scaffold is appropriate for adenosine receptor subtype A(1) ligands and that the best arranged groups around this scaffold are 3- and 4-pyridinyl at position 1, benzyl at position 3, hydrogen at position 6 and 3-thienyl or phenyl at position 9. The most interesting compounds showed K(i) for A1 in the nanomolar range and an appreciable selectivity for other receptor subtypes.  相似文献

In an attempt to recognize any ordering of DNA synthesis which might occur in ciliates, the distribution of labelled DNA in the G1 and S periods of the first, second, and third generations of a synchronized population of Euplotes eurystomus was studied by means of autoradiography. The results presented here show that the replicating DNA which at the time of label incorporation is restricted to a morphologically identifiable narrow region of the nucleus (the replication band), becomes dispersed and is evenly distributed throughout the nucleus. This dispersal of labelled DNA occurs previous to division and is observable throughout the G1 period of the following generation. During the S phase of this second generation, this previously labelled DNA once again becomes restricted to a small portion of the nucleus. Now, however, it is present at the tips of the macronucleus independently of the position of the replication band. Again the labelled DNA is found to be dispersed during G1 of the third generation. In the S period of this third generation however the radioactive DNA again appears localized in the replication bands which are found at the same position in the nucleus where they were when the pulse was given two generations earlier. The observations derived from the autoradiographic analysis suggest a non-permanent organization for at least those DNA molecules which are replicated during the first third of the S period. This DNA can be associated in either of two specific locations, replication band or tip, and these two patterns of organization alternate from generation to generation.  相似文献

Sixteen microsatellite loci are described for the two‐spotted spider mite Tetranychus urticae, which is an agricultural pest. The microsatellite loci were obtained through the construction of an enriched library; these loci exhibited polymorphisms (2–5 alleles per locus) and high levels of observed (0.033–0.667, average 0.415) and expected (0.033–0.602, average 0.336) heterozygosities. The isolated microsatellite markers are expected to be useful for the construction of a linkage map of this species.  相似文献

We characterized the nucleic acid‐sensing Toll‐like receptors (TLR) of a New World bat species, the common vampire bat (Desmodus rotundus), and through a comparative molecular evolutionary approach searched for general adaptation patterns among the nucleic acid‐sensing TLRs of eight different bats species belonging to three families (Pteropodidae, Vespertilionidae and Phyllostomidae). We found that the bat TLRs are evolving slowly and mostly under purifying selection and that the divergence pattern of such receptors is overall congruent with the species tree, consistent with the evolution of many other mammalian nuclear genes. However, the chiropteran TLRs exhibited unique mutations fixed in ligand‐binding sites, some of which involved nonconservative amino acid changes and/or targets of positive selection. Such changes could potentially modify protein function and ligand‐binding properties, as some changes were predicted to alter nucleic acid binding motifs in TLR 9. Moreover, evidence for episodic diversifying selection acting specifically upon the bat lineage and sublineages was detected. Thus, the long‐term adaptation of chiropterans to a wide variety of environments and ecological niches with different pathogen profiles is likely to have shaped the evolution of the bat TLRs in an order‐specific manner. The observed evolutionary patterns provide evidence for potential functional differences between bat and other mammalian TLRs in terms of resistance to specific pathogens or recognition of nucleic acids in general.  相似文献

General features of the operation of microwave oscillators based on the Cherenkov resonance interaction of a high-current relativistic electron beam with a preformed plasma are considered. Emphasis is placed on the presence of longitudinal modes of the plasma-beam resonator that make it possible to tune the radiation frequency. Methods by which the radiation frequency can be varied severalfold continuously or in discrete controlled steps and the width of the spectrum of simultaneously generated frequencies can be changed substantially are described.
