The adrenal gland requires stimuli from peptides derived from the ACTH precursor, pro-opiomelanocortin (POMC), to maintain its tonic state. Studies have proposed that a specific postsecretional cleavage of the nonmitogenic N-terminal 16 kDa fragment, also known as pro-gamma-melanotropin (pro-gamma-MSH), is required, releasing shorter fragments that promote adrenal growth. Here, we provide evidence for this hypothesis by the cloning and characterization of a serine protease that is upregulated during growth of the adrenal cortex. It is expressed exclusively in the outer adrenal cortex, the site of cell proliferation, and in the Y1 adrenal cell line. We also show that it is required for growth of Y1 cells, remains bound to the cell surface, and cleaves its substrate, pro-gamma-MSH, at a specific bond.  相似文献

A re-examination of the mode of vegetative nuclear division in Neurospora crassa was facilitated by the availability of the mutant “clock” which produces definite growth bands. Since the growth rhythm is correlated with nuclear divisions, stained mycelial mats of this mutant prepared at intervals from the beginning of a growth period provided a sequence of stages of division. In a 28-hour period the following broad features of nuclear behavior were observed: In the early part of the period during rapid mycelial growth, dividing elongated nuclei predominated. At the end of the period the mycelium contained mostly rounded resting nuclei. In the middle of a growth period nuclear forms of various degrees of annularity occurred along with elongated and rounded nuclei. Elongated and rounded nuclei completed division cycles without change in form, although the corresponding stages of the two types were similar. Elongated nuclei assumed a spiral form at the beginning of division. As division proceeded, relaxation of the nuclear gyres was accompanied by a visible duplication of the chromatin thread and the appearance of chromomere-like bodies on the daughter threads. One of the chromomere-like bodies became displaced and was interpreted to be a chromosome or a segment of a chromosome that acts as a mitotic center. All the chromosomes were found to be interconnected and to act as a unit throughout the division cycle. Only after the separation of the daughter chromatin threads could seven chromosomes be counted. Electron microscopic studies complemented the observations with the light microscope. On the basis of the evidence it was concluded that the vegetative nuclear division in Neurospora differs from the classical mitotic pattern in the following respects: (1) absence of visible centrioles, (2) the presence of interconnected chromosomes, (3) the comparatively late appearance of countable chromosomes, and (4) the frequent presence of interzonal connections between separating chromatin threads.  相似文献

Contrast in levels of metabolic enzymes in human and mouse ova   总被引:2,自引:0,他引:2  
A methodology is described for analyzing single human ova for 8 or 9 different metabolic enzymes, or 4 or 5 enzymes plus as many metabolites. This overcomes an obstacle to the study of human ovum metabolism: the severe limitation of usable material. Results obtained with this methodology, applied to discarded specimens from an in vitro fertilization program, indicate that in spite of imperfections these ova can provide a valid picture of the metabolic characteristics of normal human ova. Data are presented for 17 enzymes from 8 metabolic pathways in human and mouse ova. Relative to size, 10 of the enzymes were substantially higher in human than mouse ova. Most dramatically so were 2 enzymes of fatty acid metabolism (10-fold and 15-fold), hexokinase (9-fold), and aspartate aminotransferase (19-fold). This suggests that major species differences in metabolism are present. The validity of the human data, in spite of restriction to discarded material, is supported by (1) consistency of results among most of the ova, 2] concordance between average levels with those of rare specimens that were discarded because sperm were not available, and (3) the presence of adenosine triphosphate (ATP) concentrations similar to those of normal mouse ova. Surprisingly, both human and mouse ova contain phosphocreatine at levels nearly equal of those of ATP.  相似文献

Pups on San Nicolas Island were counted by two methods; counts by observers on the ground were compared to counts from aerial photographs taken with a 126-mm-format camera with image motion compensation. No difference was detected between photographic counts and ground counts (P= 0.367) when ground counters had unobstructed views. However, ground counts were significantly lower when areas with obstructed views were included in the analysis (P < 0.001). For areas with unobstructed viewing conditions, no difference was detected between counts by the two methods for rock substrates (P= 0.140), sand substrates (P= 0.468), or mixed rock-and-sand substrates (P= 0.968). No differences were found among three replicate aerial photographic censuses (P= 0.432), but a significant difference was found between two replicate ground censuses (P= 0.037). Total counts obtained from the aerial photographs were more precise (CV = 0.042) than counts obtained on the ground (CV = 0.078). Less variability in counts was found between photographic counters than for ground counters.  相似文献
