We developed a functional model for a shrub steppe vegetation of the eastern Monte Phytogeographical Province in Río Negro (Argentina) with six stable states and 12 transitions, based on the woody encroachment degree, characteristics of the herbaceous layer and the soil surface, and biological crust cover. Information was obtained from letters of naturalists and travelers along the region since the late eighteenth century, reports of old settlers, and our own research work. On the Monte, different functional groups can be distinguished. Our work focused on the three most conspicuous to analyze the dynamics of the system: shrubs, grasses and biological crust. The shrub layer is only affected by large-scale disturbances (fire, mechanical clearing) which maintain the balance between grasses and shrubs in the system. Mechanical clearing with soil removal also causes a decrease in grass cover and vegetation diversity. This layer, however, is most frequently affected by intensive and continuous grazing, which reduces the occurrence of fires and shifts the equilibrium toward the shrubs. All disturbances in general deteriorate the biological crust that covers the soil, which offers situations of facilitation for the regeneration of the herbaceous layer. Grazing and mechanical clearing reduce soil cover and promote the development of soil compaction, which reduce water infiltration and germination and/or seedling establishment, and this creates deteriorated states difficult to reverse. Those states can also be produced by tillage for agriculture and stubble grazing. Some of the transitions that we describe can be favored through grassland management strategies, in order to reach higher states of pastoral value and reverse deteriorating situations. 相似文献

The interstitial cells of Cajal (ICC) have been reported to regulate gastrointestinal motility. We investigated the distribution and the morphological and morphometric characteristics of the immunohistochemical reaction against c-kit in the forestomachs of fetal, newborn and adult cows. The anti-c-kit reaction revealed different populations of ICC among age groups and organs. ICC were more numerous and smaller in fetuses. Larger ICC were identified in newborns, except for those in the rumen. During the earliest stages of development, ICC were abundant in the inner layer of the muscularis and were consistently associated with this layer. In all samples, ICC were found in the outer layer of the tunica muscularis. ICC were found between the two muscle layers in the omasum at all ages; however, they were identified only in the rumen of the adult. Galectins, beta-galactoside-binding lectins, are extensively distributed in
the animal kingdom and share some basic molecular properties. Galectin-3, a
member of this family, is generally associated with differentiation,
morphogenesis, and metastasis. In this study, galectin-3 was isolated from
ovine placental cotyledons round the middle of the gestation period by
lactose extraction followed by affinity chromatography on lactosyl-agarose,
and separated from galectin-1 by size exclusion chromatography on a
Superose 12 column. Under native conditions this lectin behaved as a
monomer with an apparent molecular weight of approximately 29,000 and an
isoelectric point of 9.0. The partial amino acid sequence of the peptides
obtained by tryptic digestion of this protein followed by HPLC separation
showed striking homology with other members of the galectin-3 subfamily.
Furthermore, ovine placental galectin-3 exhibited specific mitogenic
activity toward rat spleen mononuclear cells. Besides, this protein
strongly reacted with a rabbit antiserum raised against a chicken galectin.
Results obtained by Western blot analysis showed that its expression was
greatly decreased in term placenta with respect to the middle of the
gestation period, suggesting a regulated expression throughout development.

We purified soluble rat liver chromatin and H1-depleted chromatin and photocrosslinked its DNA with psoralen at pH 7. Digestion of this chromatin with micrococcal nuclease produced a normal nucleosomal repeat. Chromatin was photoreacted in the presence of 0 to 700 mM-NaCl and was fractionated in sucrose gradients containing the same NaCl concentrations. The dissociation of H1 occurred as in the non-crosslinked controls and no preferential dissociation of core histones was observed. The samples between 100 and 500 mM-NaCl showed precipitation. In the electron microscope, the fibers appeared indistinguishable from the controls at low ionic strength. In the presence of 40 mM-NaCl, the fibers of the photoreacted chromatin were slightly more compact than the controls, and at 500 mM-NaCl, despite the complete dissociation of H1, there were still apparently intact fibers at this ionic strength. The disruption of the psoralen-treated chromatin fibers occurred only in 600 mM-NaCl, as opposed to 500 mM-NaCl in controls. The DNA of all the photoreacted samples was spread for electron microscopy under denaturing conditions. They revealed, for all the samples, single-stranded bubbles corresponding to 200 to 400 base-pairs in size. H1-depleted chromatin containing stoichiometric amounts of core histones was photoreacted at pH 10 and very low ionic strength. Under these conditions many of the nucleosomes appeared to be unraveled, although to a variable extent. In the electron microscope, the purified DNA from these samples showed extensive crosslinking when spread under denaturing conditions. These observations show that histone-DNA interactions different from those in intact nucleosomes may be created, which allow extensive access of psoralen to the DNA. 相似文献

Onion (Allium cepa) is an important horticulture crop because of its value as a food with a long shelf life being a relatively non-perishable product. It is very helpful to understand the growth response of the seeded onion crop to conduct appropriate field practices in attaining the highest or optimum yields. A three year field experiment was conducted using a variety of onion Valcatorce INTA, in a randomized block design with five replicates. Treatments were two plant densities and three rates of N application. The bulb growth followed a classical sigmoid curve. During the rapid growing period, the crop had the greatest leaf area (LA) with at least six leaves per plant. Increasing plant density increased yield in kg/ha, but decreased bulb size. Defoliating 40 to 60% of the LA had a significant impact on bulb production only at early growth stages. Late in the growing period, the remaining LA was apparently large enough for producing sufficient amounts of metabolites to feed new leaves, increasing their photosynthesis efficiency for the benefit of bulb production. 相似文献

The mechanisms for the regulation of homeotic genes are poorly understood in most organisms, including plants. We identified BASIC PENTACYSTEINE1 (BPC1) as a regulator of the homeotic Arabidopsis thaliana gene SEEDSTICK (STK), which controls ovule identity, and characterized its mechanism of action. A combination of tethered particle motion analysis and electromobility shift assays revealed that BPC1 is able to induce conformational changes by cooperative binding to purine-rich elements present in the STK regulatory sequence. Analysis of STK expression in the bpc1 mutant showed that STK is upregulated. Our results give insight into the regulation of gene expression in plants and provide the basis for further studies to understand the mechanisms that control ovule identity in Arabidopsis. 相似文献

目的:有效降低石油降解菌株CT-6在工业生产中的培养成本,提高培养速度。方法:利用Minitab软件,采用中心组合设计、响应面分析,以葡萄糖、蔗糖、可溶性淀粉为碳源,以硝酸铵、尿素、氯化铵为氮源,对高效降解石油菌株CT-6的培养基进行了优化。结果:当碳源葡萄糖为5.6818g/L、氮源氯化铵为3.8030g/L时,菌株CT-6培养17h,OD600达到0.615。结论:优化前后,菌体CT-6的OD600分别为0.289和0.615,提高了112.8%,说明该培养基更有利于菌株CT-6的生长。 相似文献
