Pigmentation of Drosophila eyes requires the concerted action of several genes, most of which have been cloned and characterized. Three of them, white, brown, and scarlet, have been directly implicated in the import of pigment precursors into the cells. These three genes encode similar proteins, belonging to the evolutionary conserved family of ATP Binding Cassette transporters. The identification of a novel mouse gene, ABC8, closely related to white is reported here, together with an analysis of its expression profile and its comparative mapping in mouse and human genome. Four field experiments were conducted to study the effect of Sesbania rostrata and Aeschynomene afraspera as rotational and green manure crops on the population dynamics of Hirschmanniella mucronata and H. oryzae, and subsequent rice yields. The sequential cropping of the legumes with rice controlled both nematode species. In two experiments, yield of rice was related to the nematode population denisites at planting and harvesting of the second rice crop (R² = 0.391, P < 0.001, and R² = 0.57, P < 0.001), regardless of the treatments. Rice yield increases were attributed to nutritional effect of the green manure and the reduction of the nematode populations or the modification of a factor(s) linked to the nematode populations induced by their cropping. As the two leguminous crops do not generate direct return, using them to control the rice-root nematodes was not economical, despite the significant yield increase obtained.  相似文献

A procedure for the isolation of plasma-membrane-enriched fractions from bovine 'pars intermedia' and neurohypophysis is described. Various fractions are isolated by differential centrifugation and discontinuous sucrose density gradients. The plasma-membrane-enriched fractions have a density in sucrose of 1.14 and 1.16 and the yields are 1.8 mg and 1.5 mg per gram of tissue for the pars intermedia and neural lobe, respectively. The fractions are characterized by electron microscopy and enzymatic assays. The plasma membrane fractions are mainly vesicular in nature and are free of nuclei, mitochondria, and microsomes when examined by electron microscopy. 5'-Nucleotidase (EC 3.1.3.5) and Mg2+-(Na+ + K+)-ATPase (EC 3.6.1.3) activities are concentrated in the plasma-membrane-enriched fraction. Also, adenylate cyclase (EC 4.61.1) shows a 5 to 10-fold purification in the isolated membrane fraction. NaF (10mM) gives a two to three-fold stimulation of enzymatic activity in all fractions studied The yields of adenylate cyclase, 5'-nucleotidase, and Mg2+-(Na+ +K+)-ATPase are about 6% in the membrane fraction.  相似文献

 We studied the potential of multidimensional flow cytometry to evaluate the frequency and maturation/activation status of dendritic cells in minimally manipulated peripheral blood mononuclear cell preparations (i.e., only separated on Ficoll-Hypaque) of normal donors and cancer patients. A rare subset of HLA-DR+ leukocytes (less than 1% mononuclear cells) was detected in blood of normal donors that displayed all the features of dendritic cells: these cells had high forward-light-scatter characteristics and coexpressed CD4, CD86 and CD54 surface antigens, but lacked the lineage-associated surface markers of T cells, B cells, monocytes, granulocytes or NK i.e. they were CD3, CD19, CD20, CD14, CD11b, CD16, CD56). These physical and phenotypic properties were virtually identical to those of immunomagnetically sorted leukocytes characterized as dendritic-cells on the basis of morphology, phenotype and high stimulatory activity in allogeneic mixed-lymphocyte cultures. Using this flow-cytometric approach we observed that the frequency of dendritic cell-like cells in peripheral blood mononuclear cell specimens of cancer patients receiving chemotherapy alone or those recovering from stem cell transplantation was significantly lower than that of normal individuals (mean ± SE: 0.36 ± 0.05%, 0.14 ± 0.06%, and 0.75 ± 0.04% respectively). Multidimensional flow-cytometric analysis of dendritic cells might represent an important new tool for assessing immunocompetence, and for monitoring the effects of therapeutic regimens on the immune system. Fungal laccases are enzymes that have been studied because of their ability to decolorize and detoxify effluents; they are also used in paper bleaching, synthesis of polymers, bioremediation, etc. In this work we were able to express a laccase from Trametes (Pycnoporus) sanguineus in the filamentous fungus Trichoderma atroviride. For this purpose, a transformation vector was designed to integrate the gene of interest in an intergenic locus near the blu17 terminator region. Although monosporic selection was still necessary, stable integration at the desired locus was achieved. The native signal peptide from T. sanguineus laccase was successful to secrete the recombinant protein into the culture medium. The purified, heterologously expressed laccase maintained similar properties to those observed in the native enzyme (Km and kcat and kcat/km values for ABTS, thermostability, substrate range, pH optimum, etc). To determine the bioremediation potential of this modified strain, the laccase-overexpressing Trichoderma strain was used to remove xenobiotic compounds. Phenolic compounds present in industrial wastewater and bisphenol A (an endocrine disruptor) from the culture medium were more efficiently removed by this modified strain than with the wild type. In addition, the heterologously expressed laccase was able to decolorize different dyes as well as remove benzo[α]pyrene and phenanthrene in vitro, showing its potential for xenobiotic compound degradation.  相似文献

Two novel type I ribosome-inactivating proteins (RIPs) were found in the storage roots of Mirabilis expansa, an underutilized Andean root crop. The two RIPs, named ME1 and ME2, were purified to homogeneity by ammonium sulfate precipitation, cation-exchange perfusion chromatography, and C4 reverse-phase chromatography. The two proteins were found to be similar in size (27 and 27.5 kD) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and their isoelectric points were determined to be greater than pH 10.0. Amino acid N-terminal sequencing revealed that both ME1 and ME2 had conserved residues characteristic of RIPs. Amino acid composition and western-blot analysis further suggested a structural similarity between ME1 and ME2. ME2 showed high similarity to the Mirabilis jalapa antiviral protein, a type I RIP. Depurination of yeast 26S rRNA by ME1 and ME2 demonstrated their ribosome-inactivating activity. Because these two proteins were isolated from roots, their antimicrobial activity was tested against root-rot microorganisms, among others. ME1 and ME2 were active against several fungi, including Pythium irregulare, Fusarium oxysporum solani, Alternaria solani, Trichoderma reesei, and Trichoderma harzianum, and an additive antifungal effect of ME1 and ME2 was observed. Antibacterial activity of both ME1 and ME2 was observed against Pseudomonas syringae, Agrobacterium tumefaciens, Agrobacterium radiobacter, and others.  相似文献

X-linked adrenoleukodystrophy (X-ALD) is a neurodegenerative disease due to a defect in the ABCD1 (ALD) gene. ABCD1, and the two close homologues ABCD2 (ALDR) and ABCD3 (PMP70), are genes encoding ATP-binding cassette half-transporters of the peroxisomal membrane. As overexpression of the ABCD2 or ABCD3 gene can reverse the biochemical phenotype of X-ALD (reduced beta-oxidation of very-long-chain fatty acids), pharmacological induction of these partially redundant genes may represent a therapeutic approach to X-ALD. We previously reported that the ABCD2 and ABCD3 genes could be strongly induced by fibrates, which are hypolipidaemic drugs and peroxisome-proliferators in rodents. We provide evidence that the induction is dependent on peroxisome proliferator-activated receptor (PPARalpha) as both genes were not induced in fenofibrate-treated PPARalpha -/- knock-out mice. To further characterize the PPARalpha pathway, we cloned and analysed the promoter of the ABCD2 gene, the closest homologue of the ABCD1 gene. The proximal region (2 kb) of the rat promoter displayed a high conservation with the human and mouse cognate sequences suggesting an important role of the region in regulation of the ABCD2 gene. Classically, fibrate-induction involves interaction of PPARalpha with a response element (PPRE) characterized by a direct repeat of the AGGTCA-like motif. Putative PPRE motifs of the rat ABCD2 promoter were studied in the isolated form or in their promoter context by gel-shift assay and transfection of COS-7 cells. We failed to characterize a functional PPRE, suggesting a different mechanism for the PPARalpha-dependent regulation of the ABCD2 gene.  相似文献

We tested the effect of 2-amino-5-bromo-6 phenyl-4 pyrimidinol (ABPP), an interferon (IFN) inducer, on NK cell cytotoxicity in various tissues of athymic and euthymic rats. Significant augmentation of NK cell cytotoxicity was observed in SP, PB, PE, lung, and liver of adult rats. Cytotoxic potential was also induced by ABPP in young rats, not exhibiting cytotoxicity before the treatment. Cytotoxic cells were detected in nylon wool-filtered fraction, and were not removed by carbonyl iron treatment. Separation on Percoll gradient indicated that ABPP-stimulated cytotoxic SPC resided in LGL-enriched fractions and some of them exhibited less dense characteristics than SPC from untreated rats. The ABPP-mediated cytotoxicity was thymus-independent; both athymic and euthymic rats were equally augmented. The observation that only tissues displaying NK cell augmentation produced significant levels of IFN suggest possible involvement of IFN in ABPP-mediated augmentation.  相似文献
