Dendroecological methods that use growth releases to reconstruct the history of canopy disturbances are most useful when calibrated for specific species in specific forest types. In this study, we calibrate the radial-growth averaging method to detect growth releases of western redcedar (Thuja plicata Donn ex D. Don), western hemlock (Tsuga heterophylla (Raf.) Sarg.), and Pacific silver fir (Abies amabilis Dougl. ex J. Forbes) following fine-scale canopy disturbances in old-growth forests of coastal British Columbia, Canada. Our goal is to establish a version of the method that will capture the full range of growth increases that occur for the study species following natural, fine-scale canopy gaps, yet will account for the effects of climatic variability on radial growth and growth increases. We used traditional dendroclimatological techniques and the radial-growth averaging method to examine how climatic and regional-scale factors contributed to radial growth and growth increases. In addition, we did a sensitivity analysis, using both ring widths and basal area increments, to explore how varying the values of three parameters of the radial-growth averaging method (threshold, moving average, and window) influenced the proportion of trees that showed a release pattern following the formation of canopy gaps of known timing of origin. Although radial growth of western redcedar and western hemlock was significantly associated with climate, percent-growth change derived from residual chronologies rarely exceeded 25%, which defined our minimum threshold for a release. For the sensitivity analysis, two general trends were common to all three species: (1) as threshold increased, the proportion of trees that showed a release pattern decreased, particularly for western redcedar and (2) a higher proportion of trees showed a release pattern using a 10-year versus a 5-year window, particularly for thresholds <100%. The greatest proportion of trees showed a release pattern using a 25% threshold, 5-year moving average, and 10-year window for both ring widths and basal area increments. Overall, a higher proportion of trees showed a release pattern using basal area increments as opposed to ring widths. Therefore, basal area increments are better suited to assess releases in these old-growth stands that have large inter- and intra-species variability in tree size. By establishing these empirically-based criteria, we have achieved the first step towards quantifying attributes of growth releases of trees in the study stands, allowing future studies to capture the variability of past disturbance events and predict changes in forest structure and composition over time. 相似文献

Mammalian α4 phosphoprotein, the homolog of yeast Tap42, is a component of the mammalian target‐of‐rapamycin (mTOR) pathway that regulates ribogenesis, the initiation of translation, and cell‐cycle progression. α4 is known to interact with the catalytic subunit of protein phosphatase 2A (PP2Ac) and to regulate PP2A activity. Using α4 as bait in yeast two‐hybrid screening of a human K562 erythroleukemia cDNA library, EDD (E3 isolated by differential display) E3 ubiquitin ligase was identified as a new protein partner of α4. EDD is the mammalian ortholog of Drosophila hyperplastic discs gene (hyd) that controls cell proliferation during development. The EDD protein contains a PABC domain that is present in poly(A)‐binding protein (PABP), suggesting that PABP may also interact with α4. PABP recruits translation factors to the poly(A)‐tails of mRNAs. In the present study, immunoprecipitation/immunoblotting (IP/IB) analyses showed a physical interaction between α4 and EDD in rat Nb2 T‐lymphoma and human MCF‐7 breast cancer cell lines. α4 also interacted with PABP in Nb2, MCF‐7 and the human Jurkat T‐leukemic and K562 myeloma cell lines. COS‐1 cells, transfected with Flag‐tagged‐pSG5‐EDD, gave a (Flag)‐EDD–α4 immunocomplex. Furthermore, deletion mutants of α4 were constructed to determine the binding site for EDD. IP/IB analysis showed that EDD bound to the C‐terminal region of α4, independent of the α4‐PP2Ac binding site. Therefore, in addition to PP2Ac, α4 interacts with EDD and PABP, suggesting its involvement in multiple steps in the mTOR pathway that leads to translation initiation and cell‐cycle progression. J. We assessed infrared thermography (IRT) as a tool for evaluating spatial and temporal surface temperature patterns in juvenile female harbor seals (Phoca vitulina, n=6) and adult female Steller sea lions (Eumetopias jubatus, n=2). Following a technical assessment of the influence of environmental parameters on the specific camera to be used, we identified regional and seasonal variability of surface temperatures. Variation was observed in several seasonal transitions (winter, reproductive, molt) in ten monitored body regions. Spatially and temporally consistent thermal patterns in the shoulder, axillae, foreflipper and hindflipper suggest thermal windows in both species. 相似文献

Growth hormone (GH) in rhesus macaque in vitro oocyte maturation (IVM) has been shown to increase cumulus expansion and development of embryos to the 9–16 cell stage in response to 100 ng/ml recombinant human GH (r‐hGH) supplementation during IVM. Although developmental endpoints for metaphase II (MII) oocytes and embryos are limited in the macaque, gene expression analysis can provide a mechanism to explore GH action on IVM. In addition, gene expression analysis may allow molecular events associated with improved cytoplasmic maturation to be detected. In this study, gene expression of specific mRNAs in MII oocytes and cumulus cells that have or have not been exposed to r‐hGH during IVM was compared. In addition, mRNA expression was compared between in vitro and in vivo‐matured metaphase II (MII) oocytes and germinal vesicle (GV)‐stage oocytes. Only 2 of 17 genes, insulin‐like growth factor 2 (IGF2) and steroidogenic acute regulator (STAR), showed increased mRNA expression in MII oocytes from the 100 ng/ml r‐hGH treatment group compared with other IVM treatment groups, implicating insulin‐like growth factor (IGF) and steroidogenesis pathways in the oocyte response to GH. Forward head and rounded shoulder posture (FHRSP) is theorized to contribute to alterations in scapular kinematics and muscle activity leading to the development of shoulder pain. However, reported differences in scapular kinematics and muscle activity in those with forward head and rounded shoulder posture are confounded by the presence of shoulder pain. Therefore, the purpose of this study was to compare scapular kinematics and muscle activity in individuals free from shoulder pain, with and without FHRSP. Eighty volunteers were classified as having FHRSP or ideal posture. Scapular kinematics were collected concurrently with muscle activity from the upper and lower trapezius as well as the serratus anterior muscles during a loaded flexion and overhead reaching task using an electromagnetic tracking system and surface electromyography. Separate mixed model analyses of variance were used to compare three-dimensional scapular kinematics and muscle activity during the ascending phases of both tasks. Individuals with FHRSP displayed significantly greater scapular internal rotation with less serratus anterior activity, during both tasks as well as greater scapular upward rotation, anterior tilting during the flexion task when compared with the ideal posture group. These results provide support for the clinical hypothesis that FHRSP impacts shoulder mechanics independent of shoulder pain. 相似文献

The spatial and temporal coordination of chromosome segregation with cytokinesis is essential to ensure that each daughter cell receives the correct complement of chromosomal and cytoplasmic material. In yeast, mitotic exit and cytokinesis are coordinated by signaling cascades whose terminal components include a nuclear Dbf2-related family kinase and a noncatalytic subunit, Mps one binding (Mob) 1. There are five human Mob1 isoforms, all of which display redundant localization patterns at the spindle poles and kinetochores in early mitosis, and the spindle midzone during cytokinesis. Mob1 shares similar localization patterns to Polo-like kinase (Plk1) and the chromosomal passenger complex (CPC), and although depletion of Plk1 resulted in a loss of Mob1 from the spindle poles, Mob1 recruitment to kinetochores was unaffected. Conversely, disruption of CPC signaling resulted in a loss of Mob1 from kinetochores without disrupting recruitment to the spindle poles. In Mob1-depleted cells, the relocalization of the CPC and mitotic kinesin-like protein (MKLP) 2 to the spindle midzone was delayed during early anaphase, and as a consequence, the midzone recruitment of MKLP1 also was affected. Together, these results suggest that Mob1 and the other mammalian orthologues of the mitotic exit network regulate mitotic progression by facilitating the timely mobilization of the CPC to the spindle midzone. 相似文献

When invasive species establish in new environments, they may disrupt existing or create new interactions with resident species.
Understanding of the functioning of invaded ecosystems will benefit from careful investigation of resulting species-level
interactions. We manipulated ant visitation to compare how invasive ant mutualisms affect two common plants, one native and
one invasive, on a sub-tropical Indian Ocean island. Technomyrmex albipes, an introduced species, was the most common and abundant ant visitor to the plants. T. albipes were attracted to extrafloral nectaries on the invasive tree (Leucaena
leucocephala) and deterred the plant’s primary herbivore, the Leucaena psyllid (Heteropsylla cubana). Ant exclusion from L. leucocephala resulted in decreased plant growth and seed production by 22% and 35%, respectively. In contrast, on the native shrub (Scaevola taccada), T. albipes frequently tended sap-sucking hemipterans, and ant exclusion resulted in 30% and 23% increases in growth and fruit production,
respectively. Stable isotope analysis confirmed the more predacious and herbivorous diets of T. albipes on the invasive and native plants, respectively. Thus the ants’ interactions protect the invasive plant from its main herbivore
while also exacerbating the effects of herbivores on the native plant. Ultimately, the negative effects on the native plant
and positive effects on the invasive plant may work in concert to facilitate invasion by the invasive plant. Our findings
underscore the importance of investigating facilitative interactions in a community context and the multiple and diverse interactions
