The Envelope protein (E) is one of the four structural proteins encoded by the genome of SARS‐CoV and SARS‐CoV‐2 Coronaviruses. It is an integral membrane protein, highly expressed in the host cell, which is known to have an important role in Coronaviruses maturation, assembly and virulence. The E protein presents a PDZ‐binding motif at its C‐terminus. One of the key interactors of the E protein in the intracellular environment is the PDZ containing protein PALS1. This interaction is known to play a key role in the SARS‐CoV pathology and suspected to affect the integrity of the lung epithelia. In this paper we measured and compared the affinity of peptides mimicking the E protein from SARS‐CoV and SARS‐CoV‐2 for the PDZ domain of PALS1, through equilibrium and kinetic binding experiments. Our results support the hypothesis that the increased virulence of SARS‐CoV‐2 compared to SARS‐CoV may rely on the increased affinity of its Envelope protein for PALS1. 相似文献
The length–weight relationships (LWR) were estimated for five selected demersal marine fishes from the relatively undisturbed shelves and slopes of the Cabo Verde Islands. Most of them were of ecological and/or commercial interest. Occasional sampling was carried out during different fisheries research projects. LWRs were taken for the following bycatch species for which no such estimates exists in the science literature: Myroconger compressus, Synaphobranchus affinis, Physiculus cyanostrophus, Physiculus caboverdensis and Canthidermis maculata. Total length and fork length (to the nearest 0.1 cm) and total weight (to the nearest 0.1 g) were determined. The resulting length–weight equations are characterized by a high accompanying coefficient of correlation. The present data help improving the knowledge base needed for further fish biology studies and fisheries management. 相似文献

Background aimsMetastasis to different organs is the major cause of death in breast cancer patients. The poor clinical prognosis and lack of successful treatments for metastatic breast cancer patients demand the development of new tumor-selective therapies. Thus, it is necessary to develop treatments capable of releasing therapeutic agents to both primary tumors and metastases that avoid toxic side effects in normal tissue, and neural stem cells are an attractive vehicle for tracking tumor cells and delivering anti-cancer agents. The authorspreviously demonstrated that a soluble form of growth arrest specific 1 (GAS1) inhibits the growth of triple-negative breast tumors and glioblastoma.MethodsIn this study, the authors engineered ReNcell CX (EMD Millipore, Temecula, CA, USA) neural progenitor cells to express truncated GAS1 (tGAS1) under a tetracycline/on inducible system using lentiviral vectors.ResultsHere the authors show that treatment with ReNcell-tGAS1 in combination with tetracycline decreased primary tumor growth and inhibited the formation of metastases in tumor-bearing mice by diminishing the phosphorylation of AKT and ERK1/2 in orthotopic mammary gland tumors. Moreover, the authors observed that ReNcell-tGAS1 prolonged the survival of 4T1 tumor-bearing mice.ConclusionsThese data suggest that the delivery of tGAS1 by ReNcell cells could be an effective adjuvant for the treatment of triple-negative breast cancer. 相似文献
The anthropogenic movement of species has favoured the introduction of invasive plants worldwide. Invasive plants are frequently released from their natural enemies; however, new associations with generalist herbivores may induce defence mechanisms of non-native plants. Defensive traits are often directly related to the highly competitive ability, but also to potential antagonisms and mutualisms that they can establish with soil microorganisms. Here, we examined whether the intraspecific competition and soil microorganisms influence the morphological and physiological traits of Carpobrotus edulis when is being attacked by the native generalist snail Theba pisana. To achieve this, we grew two C. edulis individuals in separate and same pots filled with live or sterile sand, and with or without T. pisana. Our results indicated that herbivory induced an increase of shoot biomass in attacked C. edulis individuals (i.e., treated donor plants), as well as in un-attacked neighbouring individuals co-growing in the same pot (i.e., untreated recipient plants). Nevertheless, intraspecific competition nor soil microorganisms did not affect the growth of C. edulis despite reduced physiological activity and damage caused by the herbivore. Overall, our findings revealed that C. edulis individuals tolerate snail attack by inducing a compensatory growth response. We conclude that phenotypic plasticity of invasive C. edulis favours tolerance against herbivores, but we also suggest that plant-plant interactions probably determine the plant growth of un-attacked neighbouring C. edulis individuals, thus favouring their invasion mechanisms.
A plant’s eventual size depends on the integration of its genetic program with environmental cues, which vary on a daily basis. Both efficient carbon metabolism and the plant hormone gibberellin are required to guarantee optimal plant growth. Yet, little is known about the interplay between carbon metabolism and gibberellins that modulates plant growth. Here, we show that sugar starvation in Arabidopsis thaliana arising from inefficient starch metabolism at night strongly reduces the expression of ent-kaurene synthase, a key regulatory enzyme for gibberellin synthesis, the following day. Our results demonstrate that plants integrate the efficiency of photosynthesis over a period of days, which is transduced into a daily rate of gibberellin biosynthesis. This enables a plant to grow to a size that is compatible with its environment. 相似文献
Abstract A 3′, 5′-cyclic-AMP phosphodiesterase (PDE) was detected and measured in the lichen Evernia prunastri. The percentage of hydrolysis of tritiated 3′, 5′-cyclic-adenosine monophosphate ([3H]-cAMP) and 3′, 5′-cyclic-guanosine monophosphate ([3H]-cGMP) by the PDE enzyme into tritiated 5′-adenosine-monophospahte ([3H]-AMP) and tritiated 5′-guanosine-monophospahte ([3H]-GMP) was measured by treating the PDE products with a 5′-nucleotidase enzyme present in snake venom. The lysate fraction (L) (plasma membranes and cell walls) and the supernatant (S) (soluble fraction of the cells) were tested. In both fractions, competition of unlabelled cAMP, but not unlabelled cGMP, was revealed. Specific competitive PDE inhibitors such as IBMX inhibited enzymatic activity. Although it is thought that in this species cAMP is regulated by red/far red light through PDE activity, this is the first report that seems to suggest the presence of a PDE activity specific for cAMP in lichenized fungi. However, this work is at a preliminary stage and despite the high levels of enzymatic activity with cAMP found in both fractions, data are still insufficient to state the absolute specificity for this nucleotide. 相似文献
Abstract The first unambiguous documentation of hybridism in the genus Doronicum (Senecioneae – Asteraceae) is reported. All our morphological, karyological and molecular data concur to indicate that Doronicum × minutilloi Peruzzi hybr. nov. (2n = 60) is a hybrid growing in Monti Aurunci (Central Italy), originated from the spontaneous crossing D. orientale Hoffm. (2n = 60) × D. columnae Ten. (2n = 60). This new hybrid shows a slightly higher morphological, karyotypic and ribotypic affinity with D. columnae, but shares a trnL-trnF IGS haplotype with D. orientale, and co-occurs with the latter species only; it has reduced fertility and a high potential for vegetative propagation through rhizome fragmentation. Our results led us to suspect in fieri homoploid hybrid speciation. 相似文献
