Phosphorylation of the contact site A glycoprotein (gp80) of Dictyostelium discoideum 总被引:5,自引:0,他引:5
Developmentally regulated cohesion of Dictyostelium discoideum can be blocked by the Fab fragment of antiserum prepared against a glycoprotein of about 80,000 daltons, gp80, purified from the membranes of developing amoebae. Immunoprecipitation of gp80 with this serum, from 32P-labeled cell extracts of aggregating D. discoideum amoebae showed it to be a phosphoprotein. Serine phosphate was found in the molecule. All multiple isoelectric forms of gp80 were phosphorylated. Synthesis of the phosphorylated form of gp80 was found to be limited to the period of aggregation and coincided with the period of incorporation of [35S]methionine into gp80. Phosphorylation could be rapidly inhibited by cycloheximide suggesting that phosphorylation occurs only on newly made gp80. No unphosphorylated gp80 could be detected in cell extracts. 相似文献

Metmyoglobin promotes arachidonic acid peroxidation at acid pH 总被引:1,自引:0,他引:1
The ability of metmyoglobin and other heme proteins to promote peroxidation of arachidonic acid under acidic conditions was investigated. Incubation of metmyoglobin with arachidonic acid resulted in a pH-dependent increase in lipid peroxidation as measured by the formation of thiobarbituric acid reactive products and oxygen consumption. Increased peroxidation was observed at pH levels below 6.0, reaching a plateau between pH 5.5 and 5.0. At comparable heme concentrations, metmyoglobin was more efficient than oxymyoglobin, methemoglobin, or ferricytochrome c in promoting arachidonic acid peroxidation. Metmyoglobin also promoted peroxidation of 1-palmityl-2-arachidonyl phosphatidylcholine and methylarachidonate but at significantly lower rates than arachidonic acid. Addition of fatty acid-free albumin inhibited arachidonic acid peroxidation in a molar ratio of 6 to 1 (arachidonic acid:albumin). Both ionic and non-ionic detergents inhibited metmyoglobin-dependent arachidonic acid peroxidation under acidic conditions. The anti-oxidants butylated hydroxytoluene and nordihydroguaiaretic acid and low molecular weight compounds with reduced sulfhydryl groups inhibited the reaction. However, mannitol, benzoic acid, and deferoxamine were without significant effect. Visible absorption spectra of metmyoglobin following reaction with arachidonic acid showed minimal changes consistent with a low level of degradation of the heme protein during the reaction. These observations support the hypothesis that metmyoglobin and other heme proteins can promote significant peroxidation of unsaturated fatty acids under conditions of mildly acidic pH such as may occur at sites of inflammation and during myocardial ischemia and reperfusion. The volatile oil from peppermint (Mentha piperita L.) is composed primarily of monoterpenes with less than 2% sesquiterpenes. However, radioactivity from mevalonate-2-14C is incorporated into caryophyllene and other sesquiterpene hydrocarbons much more extensively than into monoterpenes by peppermint cuttings. Both mono- and sesquiterpenes show maximum incorporation of label after 6 hr (0·03% vs. 0·33% of the physiological isomer) and lose 75% of the incorporated label after an additional 6 hr. Caryophyllene derived from mevalonate-2-14C after 6 hr of incorporation was chemically degraded. The isoprenoid origin of caryophyllene was confirmed, and preferential labelling of the isopentenyl pyrophosphate derived portions of the molecule was noted. On the basis of such evidence it appears that separate sites may exist for the biosynthesis of mono- and sesquiterpenes and that an endogenous dimethylallyl pyrophosphate pool may participate in the biosynthesis of sesquiterpenes in peppermint. 相似文献

The construction of a simple multiple pipet is outlined and its operation in conjunction with multitest culture trays and an efficient replicator is described. Sugar beets (Beta vulgaris L.) infected with the Beet Yellows Virus exhibit lower rates of net photosynthesis at light saturation than do healthy plants. These Pn reductions were correlated with increases in leaf resistance to water vapor loss. Theoretical analyses demonstrated that, although the leaf resistance to water vapor loss increases could account for a major part of the net photosynthesis decreases, some other aspect of leaf functioning also was debilitated by infection. Both the levels and the activities of ribulose-1, 5-diP carboxylase were less on a leaf area basis in extracts from infected leaves than from healthy ones. Soluble carbohydrates accumulate in Beet Yellows Virus-infected leaves, but inhibiting translocation in several ways provided no evidence in support of the hypothesis that the accumulation of photosynthates in leaves has a direct, short term, feed-back effect upon the photosynthetic rate. 相似文献
