Compared with the standard method of manual fertilizer broadcasting (MFB), mechanized hill-drilling direct-seeding with deep application of slow-release nitrogen fertilizer (MHDDF) is an efficient method to integrate both fertilization and seeding. However, there are few studies that combine the use of slow-release fertilizer with MHDDF. We sought to explore the combined effect of MHDDF with slow-release fertilizer on rice yield and nitrogen, phosphorus, and potassium utilization, compared to MFB. We compared three different MHDDF methods (D30: 450 kg ha?1, D40: 600 kg ha?1, D50: 750 kg ha?1), with one MFB method (B50: 750 kg ha?1), and one control (CK: 0 kg ha?1). We found that the yield of all MHDDF method was higher than that of both the MFB method. Yield was the highest in the D50 treatment and was 14.14–46.03% higher than that in B50 treatment. Biomass accumulation, nutrient accumulation, and nutrient use efficiency were similarly higher in MHDDF method than both MFB and CK. Compared to B50, the D50 treatment increased nitrogen recovery efficiency by 170.53–231.50%, phosphorus recovery efficiency by 480.00–724.25%, and potassium recovery efficiency by 201.55–169.59%. Overall, we found that combining MHDDF with slow-release fertilizer was an effective method to increase rice yield and nutrient use efficiency compared with MFB.
Microtubule dynamics and organization are important for plant cell morphogenesis and development. The microtubule-based motor protein kinesins are mainly responsible for the transport of some organelles and vesicles, although several have also been shown to regulate microtubule organization. The ARMADILLO REPEAT KINESIN (ARK) family is a plant-specific motor protein subfamily that consists of three members (ARK1, ARK2, and ARK3) in Arabidopsis thaliana. ARK2 has been shown to participate in root epidermal cell morphogenesis. However, whether and how ARK2 associates with microtubules needs further elucidation. Here, we demonstrated that ARK2 co-localizes with microtubules and facilitates microtubule bundling in vitro and in vivo. Pharmacological assays and microtubule dynamics analyses indicated that ARK2 stabilizes cortical microtubules. Live-cell imaging revealed that ARK2 moves along cortical microtubules in a processive mode and localizes both at the plus-end and the sidewall of microtubules. ARK2 therefore tracks and stabilizes the growing plus-ends of microtubules, which facilitates the formation of parallel microtubule bundles. 相似文献
Serine esterases (SEs) are hydrolases that catalyze the conversion of carboxylic esters into acids and alcohols. Lipases and carboxylesterases constitute two major groups of SEs. Although over a hundred of insect genomes are known, systematic identification and classification of SEs are rarely performed, likely due to large size and complex composition of the gene family in each species. Considering their key roles in lipid metabolism and other physiological processes, we have categorized 144 M. sexta SEs and SE homologs (SEHs), 114 of which contain a motif of GXSXG. Multiple sequence alignment and phylogenetic tree analysis have revealed 39 neutral lipases (NLs), 3 neutral lipase homologs (NLHs), 11 acidic lipases (ALs), 3 acidic lipase homologs (ALHs), a lipase-3, a triglyceride lipase, a monoglyceride lipase, a hormone-sensitive lipase, and a GDSL lipase. Eighty-three carboxylesterase genes encode 29 α-esterases (AEs), 12 AEHs (e.g., SEH4-1–3), 20 feruloyl esterases (FEs), 2 FEHs, 2 β-esterases (BEs), 2 integument esterases (IEs), 1 IEH, 4 juvenile hormone esterases, 2 acetylcholinesterases, gliotactin, 6 neuroligins, neurotactin, and an uncharacteristic esterase homolog. In addition to these GXSXG proteins, we have identified 26 phospholipases and 13 thioesterases. Expression profiling of these genes in specific tissues and stages has provided insights into their functions including digestion, detoxification, hormone processing, neurotransmission, reproduction, and developmental regulation. In summary, we have established a framework of information on SEs and related proteins in M. sexta to stimulate their research in the model species and comparative investigations in agricultural pests or disease vectors. 相似文献
The morphological effects of population density in locusts in the production of phases is now well established and in recent years somewhat parallel effects have been observed in the larvae of Lepidoptera. Unlike locusts Lepidoptera pass through a quiescent pupal period but nevertheless the adult may be affected. In both P. gamma and P. brassicae adults from crowded larval cultures were smaller in both size and weight although the effect was counteracted to some extent by approaching pupal diapause in P. brassicae. The effect was similar in many cases to that produced by an increase in temperature except that it disturbed the relationships between the lengths of the wing and hind femur with the live weight. The ratio of these lengths (equivalent to E/F ratio in locusts) was significantly increased but was not a satisfactory index of larval population density. No effects were observed in the wing pattern and colouration in either species.
Zusammenfassung Die Einwirkung der larvalen Populationsdichte auf die Morphologie der jungen Falter wurde bei Plusia gamma und Pieris brassicae durch Vergleich frischgeschlüpfter Falter aus Einzel- und aus Massenzuchten bestimmt.Bei P. gamma wogen Puppen aus Massenkulturen etwas weniger und verloren während des Puppenstadiums mehr an Gewicht. Die entstehenden Falter waren gewichtsmäßig deutlich leichter und hatten etwas kürzere Flügel. Größe und Gewicht der Männchen waren größer als die der Weibchen, und da die Massenzucht die meßbaren Größen bei den Weibchen stärker beeinflußt, verstärkte sie den Unterschied zwischen den Geschlechtern. Jahreszeitliche Unterschiede wurden beobachtet, wobei Gewicht und Größen in der warmen Jahreszeit geringer waren, doch blieb der jahreszeitliche Einfluß geringer als der durch Übervölkerung hervorgerufene.Bei P. brassicae hatte die Massenzucht eine ähnliche Wirkung auf die Puppen- und Erwachsenengewichte und auch auf die Maße, außer daß der Flügel kürzer und schmaler war. Die Unterschiede zwischen den Geschlechtern wurden durch die Massenaufzucht nicht verschärft. Lebendgewicht und Femurlänge waren beim Männchen größer, währen das Weibchen größere Flügel hatte. Bei Herannahen der Diapause wurde die Wirkung der Massenkultur auf Puppen- und Imaginalgewicht kompensiert, aber die Wirkungen auf die Erwachsenenmaße blieben noch erhalten, wenn auch in geringerem Ausmaß als sie bei nichtdiapausierenden Generationen zu beobachten waren. In der wärmeren Jahreszeit wurden Variationen erzielt, die in etwa den durch Übervölkerung bewirkten ähnlich waren, außer daß die Flügel abweichende Form aufwiesen und daß Übervölkerung bei den Männchen die Wirkung stark reduzierte.In Einzelkulturen beider Arten wurden positive, aber nicht stark signifikante Korrelationen in der Beziehung von Costa- und Hinterschenkellänge mit dem Lebendgewicht gefunden. Diese Beziehungen wurden durch Massenkultur gestört.In beiden Arten wurde das Costa-Schenkel-Verhältnis durch Massenkultur signifikant vergrößert (cf. E/F-Rate bei Heuschrecken), ergab aber keinen befriedigenden Index für die larvale Populationsdichte.In der Flügelzeichnung und Färbung beider Arten wurden durch larvale Massenkultur keine Wirkungen beobachtet.
Triple-helix formation of the peptide (Pro-Hyp-Gly)10 was monitored by nmr and CD spectroscopy. The two-dimensional nmr spectra indicated that the Gly C alpha H and Pro C delta H proton resonances shift upfield in going from the nonhelical to helical form, while hydroxy-proline resonances are unchanged. The integrated areas of the helical and nonhelical resonances could be monitored in the one-dimensional nmr spectrum, and indicate that in the (Pro-Hyp-Gly)10 about 90% of the residues are in a defined triple-helical conformation. The introduction of a glycine to alanine substitution or the deletion of a single hydroxyproline residue in the stable triple-helical peptide (Pro-Hyp-Gly)10 still allows trimers to be formed, but the trimers show a substantial loss of triple helix and decreased thermal stability compared with (Pro-Hyp-Gly)10. Two computer models were generated for the Gly----Ala peptide, one with the Ala side chains packed inside the helix and the other with the region containing the alanines forming a beta-bend that loops out from the helix. The alien addition line TAI-27 contains a pair of chromosomes of Thinopyrum intermedium that carry resistance against barley yellow dwarf virus (BYDV). A subtractive library was constructed using the leaves of TAI-27, which were infected by Schizaphis graminum carrying the GAV strain of BYDV, and the control at the three-leaf stage. Nine differentially expressed genes were identified from 100 randomly picked clones and sequenced. Two of the nine clones were highly homologous with known genes. Of the remaining seven cDNA clones, five clones matched with known expressed sequence tag (EST) sequences from wheat and (or) barley whereas the other two clones were unknown. Five of the nine differentially expressed sequences (WTJ9, WTJ11, WTJ15, WTJ19, and WTJ32) were highly homologous (identities >94%) with ESTs from wheat or barley challenged with pathogens. These five sequences and another one (WTJ18) were also highly homologous (identities >86%) with abiotic stress induced ESTs in wheat or barley. Reverse Northern hybridization showed that seven of the nine differentially expressed cDNA sequences hybridized with cDNA of T. intermedium infected by BYDV. Three of these also hybridized with cDNA of line 3B-2 (a parent of TAI-27) infected by BYDV. The alien chromosome in TAI-27 was microdissected. The second round linker adaptor mediated PCR products of the alien chromosomal DNA were labeled with digoxygenin and used as the probe to hybridize with the nine differentially expressed genes. The analysis showed that seven differentially expressed genes were homologous with the alien chromosome of TAI-27. These seven differentially expressed sequences could be used as ESTs of the alien chromosome of TAI-27. This research laid the foundation for screening and cloning of new specific functional genes conferring resistance to BYDV and probably other pathogens. 相似文献
To evaluate the role of bacterial symbionts (Buchnera spp.) in the black bean aphids (Aphis craccivora Koch), the aphids were treated with the antibiotic, rifampicin, to eliminate their intracellular symbiotic bacteria. Analysis of protein and amino acid concentration in 7‐day‐old of aposymbiotic aphids showed that the total protein content per mg fresh weight was significantly reduced by 29%, but free amino acid titers were increased by 17%. The ratio of the essential amino acids was in general only around 20% essential amino acids in phloem sap of broad bean, whereas it was 44% and 37% in symbiotic and aposymbiotic aphids, respectively, suggesting that the composition of the free amino acids was unbalanced. For example, the essential amino acid, threonine represented 21.6% of essential amino acids in symbiotic aphids, but it was only 16.7% in aposymbiotic aphids. Likewise, two nonessential amino acids, tyrosine and serine, represented 8.9% and 5.6% of total amino acids in symbiontic aphids, respectively, but they enhanced to 21.1% and 13.6% in aposymbiotic aphids. It seems likely that the elevated free amino acid concentration in aposymbiotic aphids was caused by the limited protein anabolism as the result of the unbalanced amino acid composition. 相似文献
