We compared the efficacy of four plant essential oils to repel onion thrips, Thrips tabaci (Lindeman) (Thysanoptera: Thripidae), in the presence of an attractive odour, ethyl iso‐nicotinate in a pasture field. Four horizontal white sticky plates were placed adjacent to (directions: N, S, E, W) a central horizontal white plate (C). After 24 h, in the treatment combination where the four plates were sprayed with essential oil surrounding a central sticky plate sprayed with ethyl iso‐nicotinate, fewer onion thrips were found on the plates treated with sweet marjoram [Origanum majorana L. (Labiatae)] or clove basil [Ocimum gratissimum L. (Labiatae)] (87 and 71% less, respectively) compared to the control treatment of four water‐sprayed plates surrounding a central plate with ethyl iso‐nicotinate. We also compared the distribution of onion thrips on the plates. Relative thrips numbers on each plate were compared with similar (N, S, E, W, and C) plates in the control treatment. There were relatively lower thrips numbers on the south (23% reduction) and west (26% reduction) O. majorana‐treated plates and higher numbers (37% increase) on the central attractant‐treated plate indicating a short‐distance push–pull effect. When four plates were sprayed with the thrips attractant surrounding a central sticky plate sprayed with an essential oil or water (control), only O. majorana reduced the number of thrips on the attractant‐sprayed plates (62% reduction). The distribution of thrips on the different plates within this treatment combination did not change substantially when compared to the distribution in the water‐control treatment. Other essential oils tested (wormwood [Artemisia arborescens L. (Compositae)]) and tea tree [Melaleuca alternifolia (Maiden. & Betche.) Cheel. (Myrtaceae)]) were not effective in any of the treatments. It appears that O. majorana is a promising thrips repellent which could be used for further testing in a push–pull system with the attractant ethyl iso‐nicotinate. The field setup used also proved to be a valuable tool for evaluating the potential of repellents to control onion thrips.  相似文献

With the increased attention to bioenergy and especially cellulosic ethanol, there are concerns regarding potential competition for available land between biofuels and feeds/foods. Ammonia fiber expansion (AFEX), a pretreatment process for cellulosic ethanol, may also be used to improve ruminant digestibility of feedstuffs not traditionally used as forages. Eleven forages – including traditional forages, agricultural residues, and dedicated energy crops – were AFEX treated and digested in vitro with rumen inoculum. AFEX treatment improved 48-h neutral detergent fiber (aNDFom) digestion for several moderately indigestible forages compared to untreated samples, but showed no improvement for highly digestible samples. Of particular interest are corn stover and late-harvest switchgrass, as AFEX treatment improved digestibility by 52% and 128% over untreated material, whereas the improvement was 74% and 70% over conventional ammonia treatment, respectively. The crude protein content of all treated samples increased to more than 100 g/kg dry forage. This research strongly suggests that AFEX-treated feedstuffs can be competitive with traditional forages, and thus offer expanded options for ruminant feeding.  相似文献

Dry phospholipid bilayers are known to undergo transient changes in permeability during rehydration. In this review, we present evidence from which we suggest that this permeability change is due to a gel to liquid-crystaline phase transition accompanying rehydration. If the transition is avoided, as in lipids that remain in gel phase whether dry or rehydrated, the problem of leakage during rehydration is obviated, at least in part. Further, the evidence that the transition temperature for dry bilayers can be depressed by certain sugars is discussed. Finally, we show that these principles can be extended to intact cells. Using pollen grains as a model, we have measured the transition temperature for membrane phospholipids and show that the transition is correlated with physiological measurements including permeability changes and subsequent germination. From theT m values taken from pollen grains at different water contents, we have constructed a phase diagram for the intact pollen that has high predictive value for physiological properties.  相似文献

The Salton Sea (Sea) is a eutrophic to hypereutrophic lake characterized by high nutrient concentrations, low water clarity, and high biological productivity. Based on dissolved phosphorus (P) and nitrogen (N) concentrations and N:P ratios, P is typically the limiting nutrient in the Sea and, therefore, should be the primary nutrient of concern when considering management efforts. Flows in the major tributaries to the Sea have been measured since 1965, whereas total P (TP) concentrations were only measured intermittently by various agencies since 1968. These data were used to estimate annual P loading from 1965 to 2002. Annual loads have increased steadily from ∼940,000 kg around 1968 to ∼1,450,000 kg in 2002 (∼55% increase), primarily a result of increased TP concentrations and loads in the New River. Although the eutrophic condition of the Salton Sea is of great concern, only limited nutrient data are available for the Sea. It is difficult to determine whether the eutrophic state of the Sea has degraded or possibly even improved slightly in response to the change in P loading because of variability in the data and changes in the sampling and analytical methodologies. Pannexin1 (Panx1) subunits oligomerize to form large-pore channels between the intracellular and extracellular milieu that have been shown to regulate proliferation, differentiation and cell death mechanisms. These key cellular responses are ultimately necessary for normal tissue development and function but the role of Panx1 in development, differentiation and function in many tissues remains unexplored, including that of the breast. Panx1 was identified to be expressed in the mammary gland through western blot and immunofluorescent analysis and is dynamically upregulated during pregnancy and lactation. In order to evaluate the role of Panx1 in the context of mammary gland development and function, Panx1-/- mice were evaluated in comparison to wild-type mice in the mammary glands of virgin, lactating and involuting mice. Our results revealed that Panx1 ablation did not affect virgin or involuting mammary glands following histological and whole mount analysis. Panx1 was necessary for timely alveolar development during early lactation based on a decreased number of alveolar lumen following histological analysis and reduced proliferation following Ki67 immunofluorescent labelling. Importantly, the loss of Panx1 in lactating mammary glands did not overtly affect epithelial or secretory differentiation of the mammary gland suggesting that Panx1 is not critical in normal mammary gland function. In addition, PANX1 mRNA expression was correlated with negative clinical outcomes in patients with breast cancer using in silico arrays. Together, our results suggest that Panx1 is necessary for timely alveolar development following the transition from pregnancy to lactation, which may have implications extending to patients with breast cancer.  相似文献

XRCC4 plays a crucial role in the nonhomologous end joining (NHEJ) pathway of DNA double-strand break repair acting as a scaffold protein that recruits other NHEJ proteins to double-strand breaks. Phosphorylation of XRCC4 by protein kinase CK2 promotes a high affinity interaction with the forkhead-associated domain of the end-processing enzyme polynucleotide kinase/phosphatase (PNKP). Here we reveal that unphosphorylated XRCC4 also interacts with PNKP through a lower affinity interaction site within the catalytic domain and that this interaction stimulates the turnover of PNKP. Unexpectedly, CK2-phosphorylated XRCC4 inhibited PNKP activity. Moreover, the XRCC4·DNA ligase IV complex also stimulated PNKP enzyme turnover, and this effect was independent of the phosphorylation of XRCC4 at threonine 233. Our results reveal that CK2-mediated phosphorylation of XRCC4 can have different effects on PNKP activity, with implications for the roles of XRCC4 and PNKP in NHEJ.  相似文献

To test if otoliths can be used to track fish migration in polluted areas, fish sampled from Onondaga Lake, heavily polluted with mercury, were used in an assay to determine whether mercury was detectable in the fishes’ otoliths using synchrotron-based scanning X-ray fluorescence microscopy (SXFM). Mercury was undetectable, but selenium, rarely reported in otoliths and known as a physiological antagonist to mercury, was. Strontium was also present but appeared to be taken up independently of selenium, and thus these serve as independent biogeochemical markers. Both selenium and mercury were detected in fish tissues, but selenium was below levels considered toxic. Selenium was low in otoliths of fishes collected in nearby Oneida Lake. Synoptic surveys of water chemistry revealed that Se is regionally highest in Onondaga Lake and in particular its main inlet, Onondaga Creek. SXFM appears to be a sensitive method for detecting selenium in otoliths.  相似文献

Maternal licking in rats affects the development of the spinal nucleus of the bulbocavernosus (SNB), a sexually dimorphic motor nucleus that controls penile reflexes involved with copulation. Reduced maternal licking produces decreased motoneuron number, size, and dendritic length in the rostral portion of the adult SNB as well as deficits in adult male copulatory behavior. Previous research suggests that decreases in perineal tactile stimulation may be responsible for these effects. To determine whether the regional effects of maternal licking on SNB morphology are driven by sensory afferent innervation of the lumbosacral spinal cord, we used WGA‐HRP to reconstruct the location of sensory afferent fibers from the perineal skin. We found that these fibers are caudally concentrated relative to the area of the SNB dendritic field, with the rostral dendritic arbor receiving little perineal afferent innervation. We also assessed Fos expression following perineal tactile stimulation to determine whether it increased local spinal cord activity in the SNB dendritic field. Sixty seconds of licking‐like perineal stimulation produced a transient 115% increase in Fos expression in the area of the SNB dendritic field. This effect was driven by a significant increase in Fos in the caudal portion of the SNB dendritic field, matching the pattern of perineal afferent fiber labeling. Perineal tactile stimulation also produced significantly greater Fos expression in male pups than in female pups. Together, these results suggest that perineal sensory afferent activity mediates the effects of early maternal care on the masculinization of the SNB and resultant male copulatory behavior. © 2009 Wiley Periodicals, Inc.
