Blockade of the adrenergic receptors or the injection of Bordetella pertussis vaccine prevented the normal hyperglycemic response of CFW mice to exogenous cyclic 3',5'-adenosine phosphate (3',5'-AMP). The cyclic nucleotide was also ineffective in offsetting the histamine hypersensitivity of the beta-adrenergically blocked and the pertussis-sensitized groups. These observations suggest that the autonomic malfunction occurs at a point(s) subsequent to release of 3',5'-AMP. Neither the hyperglycemic effect of epinephrine or 3',5'-AMP, or both, nor the histamine sensitivity of pertussis-pretreated animals was influenced by the subcutaneous administration of theophylline. 相似文献

The nature of the phage typing reactions on Brucella species was determined by rates of adsorption and infection, one-step growth experiments, and susceptibility to lysis from without. The highest rates of adsorption and infection were obtained on smooth B. abortus cultures, and large clear plaques were produced. One or a few phage particles per B. neotomae cell killed about one-half of the cells, but some went through an infective cycle and released mature phage that resulted in production of small clear plaques. With B. suis, more phage particles per cell were required to kill, replication did not occur, and plaques were not observed. Still greater numbers of phage particles were required to cause some inhibition of growth of B. melitensis lawns. Rough Brucella cultures and species, such as B. ovis and B. canis, were not affected by the highest concentrations of phage. B. abortus cultures of intermediate colonial morphology adsorbed phage, but only a few infected cells (after a delayed latent period) released mature phage. An infected culture or colony appeared normal until spontaneous phage mutants appeared which could penetrate the cell wall more effectively than the parent phage. Somatic mutations were induced in maize embryos in order to follow the albino-tissue patterns in mature plants. A reconstruction of cellular development in the shoot apex has been attempted. Two strains of maize were employed, wd/Yg2 and pastel-8549/y1 for seed irradiation with gamma rays. After mature plants had developed from this radiated seed, the sectored plants were analyzed in detail for their patterns of albino tissue. The location and frequency of these patterns were correlated with cell number at various sites of the initial shoot apex in order to deduce the number of cells contributing to each frequency class. Various lines of evidence lead to the conclusion that the cellular differentiation in the shoot apex is organized and a relatively stable process. Apparently a few cells in the apical dome provided daughter tissue for the upper half of the maize plant. Various sector patterns are diagrammed and the position of their albino tissue is explained in relation to the location of a specific cell in the apex. 相似文献

Activity of myofibrillar adenosinetriphosphatase was demonstrated histochemically at a fine structural level in isolated, unfixed or hydroxyadipaldehyde-fixed cardiac myofibrils in the rat, using a lead precipitation technique and either Ca++ or Mg++ as activating ion. Activity in relaxed myofibrils was found in the A band, but not the H, I, or Z bands. Deposits of final product frequently exhibited an axial periodicity of near 365 A, and bore a close relationship to filaments within the A band. Several patterns of distribution occurred in contracted myofibrils. In myofibrils which had shortened to the point of disappearance of the I band, final product was distributed throughout the sarcomere, except for the unreactive Z band. A second type of distribution occurred in strongly contracted fibers in which there was intensification of activity in the center of the sarcomere. These findings are discussed in the light of the recent morphological evidence and it is suggested that the distribution of final product is consistent with localization of enzyme activity to the cross-bridges between the thick and thin filaments. 相似文献

The chloroplasts of three genera of marine algae, Dictyota, Padina, and Bryopsis, were labeled with tritiated-thymidine for various time periods during culture in "Erd-Schreiber's" solution. Autoradiographs were prepared from both smeared and sectioned material. They revealed that almost all of the radioactivity was in the cytoplasm and associated with the chloroplasts, as detected in the overlying silver halide crystals. Deoxyribonuclease, ribonuclease, and hot trichloracetic acid treatments indicated that the loss of radioactivity corresponded to the removal of DNA and not RNA. Quantitative studies of silver grain distribution suggested that the radioactivity of the labeled DNA originated from the edge of the pyrenoids on either side in the longitudinal direction of Bryopsis chloroplasts. Nuclei did not incorporate H3-thymidine even though cells were dividing rapidly in the three genera examined. It is postulated that the enzyme, thymidine kinase, is absent as a coding sequence of nuclear DNA in algae, but is present in chloroplast DNA. When the chloroplasts of Dictyota and Padina in various stages of division were scored for labeling, there appeared to be a DNA synthesis period, analogous to S period in cell division. This chloroplast-labeling period occurred just previous to fission. Many of the criteria seem to have been satisfied to establish the self-reproducing and semi-autonomous nature of chloroplasts, especially when combined with the chemical, genetic, and morphological evidence. 相似文献

The nuclear changes accompanying spermatid elongation have been studied in two species of grasshopper, Dissosteira carolina and Melanoplus femur-rubrum. Testes were fixed in 1 per cent buffered OsO4, imbedded in butyl methacrylate, and examined as thin sections in the electron microscope. In both species nuclear changes during spermatid development involve (1) an early period, during which the nuclear contents are predominately fibrous; (2) a middle period, characterized by the lateral association of the nuclear fibers to form plates or lamellae which are oriented longitudinally in the major axis of the elongated nucleus; and (3) a late period, involving coalescence of the lamellae into a crystalline body which eventually becomes so dense that all resolvable detail is lost. The fibers seen in the early spermatid nucleus are about 150 A in diameter and so are similar to fibers described from other types of nuclei. The thickness of the lamellae varies from about 150 A when first formed to 70 A during the later stages. The lack of evident chromosomal boundaries in the spermatid nucleus makes it difficult to relate either the fibers or lamellae to more familiar aspects of chromosome structure. We see no apparent reason to consider that the fiber alignment described here is related to conventional chromosome pairing. 相似文献

Brucella antigens capable of sensitizing normal and tanned sheep red blood cells for indirect hemagglutination were compared with antigens involved in agglutination, gel diffusion, and immunoelectrophoresis. Hyperimmune rabbit sera, before and after absorption with various antigenic preparations from smooth and rough B. abortus, were used in the tests. Normal erythrocytes could be sensitized with an NaOH-treated ether-water extract (EW-T) of smooth Brucella. Tanned erythrocytes could be sensitized with a water-soluble extract from ultrasonically disrupted smooth or rough Brucella. The EW-T produced a single precipitation band and the water-soluble antigens produce 6 to 23 bands in immunoelectrophoresis with unabsorbed sera. After absorption of antisera with water-soluble extracts from smooth or rough Brucella cells or from smooth or rough cell walls, the hemagglutinins for sensitized tanned erythrocytes and the precipitins for water-soluble antigens were removed. Absorption with living smooth or rough Brucella cells or with EW-T did not remove these antibodies. The precipitins and hemagglutinins for the antigen EW-T, and agglutinins for smooth cells, were absorbed by smooth antigens but not by rough antigens. It appears that the antigens which sensitize tanned erythrocytes and diffuse through agar gels are present in both smooth and rough forms and may be situated in the cytoplasm or in the internal part of the cell wall, whereas the agglutinogen and the antigen which attaches to normal erythrocytes are surface antigens found only on the smooth Brucella cell. 相似文献

Immune sera were prepared in rabbits by the injection of living and acetone-killed cells of Brucella ovis and smooth and rough B. melitensis. The use of whole-cell antigens in agglutination and agglutinin-absorption tests revealed little relationship between B. ovis and smooth B. melitensis, although there was extensive cross-agglutination between B. ovis and rough B. melitensis. The use of water-soluble antigens prepared from ultrasonically treated cells of the three strains revealed extensive cross-reactions in indirect hemagglutination, agar gel precipitation, and immunoelectrophoresis tests, as well as in allergic skin tests in rabbits. The most definitive results were obtained with the immunoelectrophoresis technique. B. ovis antigen produced at least 11 lines with its homologous serum. All were removed by absorption of the serum with rough B. melitensis antigen. All but three were removed by absorption with smooth B. melitensis antigen. Smooth B. melitensis antigen produced 11 lines with its homologous serum, and all but 3 were removed by absorption with B. ovis antigen. Rough B. melitensis produced nine lines with its homologous serum, and eight were removed by B. ovis antigen. The extensive cross-reactions between soluble antigens of B. ovis and B. melitensis are added evidence that B. ovis belongs in the genus Brucella. 相似文献
