To determine the mechanism by which gastric inhibitory polypeptide (GIP) and insulin stimulate the in vitro fatty acid incorporation into adipose tissue (FIAT), we measured the cyclic AMP variations and FIAT in epididymal fat pads of lean Fa/-- and obese fa/fa Zucker rats. GIP was used at 1, 2 and 4 ng/ml and insulin at a concentration of 100 microU/ml. There was no significant variation of cAMP when FIAT was stimulated either by GIP, either by insulin or by both hormones. There was no correlation at all between FIAT increases and cAMP levels. We conclude that GIP and insulin act through cAMP independent mechanisms in adipose tissue. The modification by GIP of insulin binding to adipocytes or an easier passage of fatty acids through the membrane could constitute alternative solutions for such mechanisms.  相似文献

We have examined the nuclear localization of isoprenylated proteins in CHO-K1 cells labeled with [14C]mevalonate. Nuclear proteins of 68, 70, and 74 kD, posttranslationally modified by an isoprenoid, are also components of a nuclear matrix-intermediate filament preparation from CHO cells. Furthermore, the 68-, 70-, and 74-kD isoprenylated polypeptides are immunoprecipitated from cell extracts with two different anti-lamin antisera. Based on exact two-dimensional comigration with lamin B, both from rat liver lamin and CHO nuclear matrix-intermediate filament preparations, and its immunoprecipitation with anti-lamin antisera, we conclude that the 68-kD isoprenylated protein found in nuclei from [14C]mevalonate-labeled CHO cells is lamin B. The more basic 74-kD isoprenylated nuclear protein is similar in molecular mass and isoelectric pH variants to the lamin A precursor polypeptide reported by others. Starving cells for mevalonate results in a dramatic accumulation of a polypeptide that comigrates on two-dimensional, non-equilibrium pH gradient electrophoresis (NEPHGE) gels with the 74-kD isoprenylated protein. The 70-kD isoprenylated protein, which is resolved on NEPHGE gels as being higher in molecular mass and slightly more basic than lamin B, has not yet been identified.  相似文献

Vinpocetine prevents ischemic cell damage in rat hippocampus   总被引:3,自引:0,他引:3  
The effects of vinpocetine on hippocampal cell damage and local cerebral blood flow (LCBF) were measured in a rat model of forebrain ischemia (2-vessel occlusion and hypotension). Duration of ischemia was 10 min. LCBF was determined after 2 min of recirculation using the 14C-iodoantipyrine technique. Hippocampal cell loss was quantified histologically 7 days post-ischemia. Intraperitoneal application of vinpocetine (10 mg/kg) 15 min prior to ischemia significantly reduced neuronal cell loss in hippocampal CA 1 sector from 60% to 28%. The drug led to a marked increase in blood flow in cortical areas, whereas LCBF remained unchanged in hippocampus and all other structures measured. It is suggested that the protective effect of vinpocetine does not depend on increased postischemic blood flow.  相似文献

The national cervical cytology registry being developed in the Netherlands is described. A large-scale screening program for cervical cancer has been in effect since 1975 in the region of the cities of Nijmegen, Utrecht and Rotterdam. At the start of the pilot projects, laboratories agreed upon a uniform protocol for reporting cytologic findings and recommendations for follow-up examinations in cases of abnormalities. Based on the results of the three pilot projects, in 1985 the Dutch government decided to organize a nationwide screening program for cervical cancer. All pathology laboratories involved in this national screening program are using the same screening protocol and the same coding system for cytologic and histologic diagnoses. By the end of 1989, all pathology laboratories will be linked to a central pathology diagnosis data base (PALGA). Linkage of screening results to previous and follow-up cytologic and histologic findings will enable epidemiologic studies on a regional or national level. Each physician who has submitted specimens will, next to the cytology reports, periodically receive reviews of the number of smears submitted, the cellular composition (quality) of those smears and the follow-up findings. The execution of requests for follow-up examination will be supervised by the participating pathology laboratories. The national cervical cytology registry will enable registration of all relevant cytologic and histologic diagnoses in a uniform way, but will also establish a unique high-quality national data bank, which will be of great value in the analysis of the effectiveness of the national screening program for cervical cancer. It will enable measurement of the impact of various screening protocols and give insight into the behavior of cervical cancer and the progressive or regressive character of its early stages. It will also offer the opportunity to initiate and evaluate quality control protocols.  相似文献

Summary 14CO2 at ambient concentration was administered to a section of an upper branch of 8-year-old Scots pines and the import of radiocarbon into the stem and roots was determined after various chase periods. 14CO2 fixation was performed in October when export of carbon into the stems and roots was maximal. In the short-term experiments the trees were harvested 1 h, 2 days and 5 days after a 3-h 14C pulse, while chase periods of 5 or 8 months were used in the long-term experiments. Loss of 14C was initially substantial, and even after a 5-day chase had not come down to a rate which indicated decrease only by respiration. After 5 days, more than 10% of the recovered radiocarbon (53% of the 14C translocated into the stem) had entered the roots and approximately the same amount was found in the stem. Extension of the chase period beyond 5 months did not result in a further significant loss of 14C by respiration, and the bulk of the label could be localized in the cell-wall fraction. No substantial redistribution of radiocarbon prior and subsequent to the formation of the new shoots could be observed, thus indicating that the stored material was utilized for thickening the stem and roots. Radioautography of stem cross-sections revealed a narrow helical strip of 14C from the feeding branch to the root in the phloem region. In the tree harvested after bud break the utilization of the 14C-labelled material stored in the stem for the production of the first layers of earlywood and the corresponding phloem was apparent.  相似文献

Summary A number of parameters characteristic of the wing margin precursor in imaginal discs of Drosophila are known: the zone of non-proliferating cells or ZNC (O'Brochta and Bryant 1985), aldehyde oxidase (AO) and other enzyme staining patterns (Sprey et al. 1982), E1C antigen localization in a narrow band along the margin (Piovant and Lena 1988). To test our hypothesis that such parameters, and others, act in concert to determine margin identity and the positional information that specifies the bristles and hairs appropriate to the anterior, posterior and distal margins, we have examined these parameters in the dominant mutant Lyra, in which much of the anterior and posterior margins is missing. After establishing that Lyra phenotype is already evident in the early pupal wing, we tested the known imaginal disc parameters and found that only Mab E1C (Piovant and Lena 1988) distribution differs from wild type, suggesting that E1C antigen may be a component of positional information. Sibatani's (1983) model for specification of positional information (PI) applied to wing discs predicts the Lyra adult wing shape as well as the reduced distribution of E1C antigen in Lyra wing discs. The model is based on the assumption that specification of positional information depends on interactions of multiple, independent factors. Clonal analysis with shaggy (Simpson et al. 1988 and Ripoll et al. 1988) indicates that factors in addition to E1C antigen contribute to margin PI in Lyra wings and should allow us to test the multi-component hypothesis further.  相似文献

The standard of obstetrics care by general practitioners in Bradford was assessed by reviewing the case records of all women who in 1988 were booked for delivery under their general practitioner but subsequently required transfer to consultant care. A total of 5885 women were delivered in Bradford during 1988. Of 1289 booked under their general practitioner, 637 required transfer to consultant care. In 259 cases transfer occurred during labour; only 37 of these women were visited by their general practitioner. Many of the problems that precipitated transfer were predictable and some were considered preventable: 263 of the women transferred were considered unsuitable for booking by general practitioners. The perinatal mortality among women booked under their general practitioner was 10.1/1000 and the stillbirth rate 7.8/1000. These figures are high and suggest a need for tighter controls over the qualifications and experience of doctors participating in a fully integrated system of obstetric care.  相似文献

A number of cell-cycle-specific temperature-sensitive (ts) mutants have been isolated from animal cells, especially Syrian hamster cells. These ts mutants, like cell cycle ts mutants of yeast, can be complemented by specific genes, some of which have been molecularly cloned. We have isolated a cDNA clone that complements TK-ts13 cells, but only temporarily. This clone, called B1, differs from a previously isolated clone (Sekiguchi et al.: EMBO Journal 7:1683-1687, 1988) that specifically complements ts13 cells. In addition, B1 also complemented temporarily three other ts mutants of the cell cycle, tsAF8, ts694, and ts550C cells. These mutants have different mutations since, in cell fusion experiments, they complement each other. Sequencing of the B1 cDNA clone revealed that it was a mutant of human ADP/ATP translocase in which some human sequences at the 5' end have been replaced by SV40 sequences. The wild-type translocase was less effective but could still increase the survival time of cell cycle ts mutants at the restrictive temperature. Using the polymerase chain reaction, it was possible to demonstrate that the B1 plasmid is expressed in TK-ts13 cells undergoing temporary complementation.  相似文献
