A series of Plectranthus spp. plant extracts (aqueous, acetonic, methanolic and ethyl acetic) obtained from eight different species, and previously isolated compounds (ranging from polyphenols, diterpenes and triterpenes), were assayed for in vitro inhibition of the skin-related enzymes tyrosinase, collagenase and elastase, and for studying their antioxidant properties. The ethyl acetic extracts of P. grandidentatus and P. ecklonii registered the highest antioxidant activity, whereas acetonic, methanolic and ethyl acetic extracts of P. ecklonii, P. grandidentatus, P. madagascariensis and P. saccatus concerning the enzymatic inhibition assays revealed high anti-tyrosinase and anti-collagenase activities. From the isolated compounds tested, abietane diterpenes and triterpenes were highly active against tyrosinase and elastase activity. Overall, the experimental results showed the powerful antioxidant and inhibitory action on skin-related enzymes tyrosinase, collagenase and elastase of Plectranthus spp. extracts and/or isolated compounds, supporting their further research as bioactive metabolites against skin sagging and hyperpigmentation in cosmetic and pharmaceutical formulations. 相似文献
Parkinson''s disease (PD) is a neurodegenerative disorder that affects adult people whose treatment is palliative. Thus, we decided to test three dammarane triterpenes 1, 1a, 1b, and we determined that 1 and 1a inhibit β-aggregation through thioflavine T rather than 1b. Since compound 1 was most active, we determined the interaction between α-synuclein and 1 at 50 µM (Kd) through microscale thermophoresis. Also, we observed differences in height and diameter of aggregates, and α-synuclein remains unfolded in the presence of 1. Also, aggregates treated with 1 do not provoke neurites'' retraction in N2a cells previously induced by retinoic acid. Finally, we studied the potential sites of interaction between 1 with α-synuclein fibrils using molecular modelling. Docking experiments suggest that 1 preferably interact with the site 2 of α-synuclein through hydrogen bonds with residues Y39 and T44. 相似文献
Intrinsic protein disorder is an interesting structural feature where fully functional proteins lack a three-dimensional structure in solution. In this work, we estimated the relative content of intrinsic protein disorder in 96 plant proteomes including monocots and eudicots. In this analysis, we found variation in the relative abundance of intrinsic protein disorder among these major clades; the relative level of disorder is higher in monocots than eudicots. In turn, there is an inverse relationship between the degree of intrinsic protein disorder and protein length, with smaller proteins being more disordered. The relative abundance of amino acids depends on intrinsic disorder and also varies among clades. Within the nucleus, intrinsically disordered proteins are more abundant than ordered proteins. Intrinsically disordered proteins are specialized in regulatory functions, nucleic acid binding, RNA processing, and in response to environmental stimuli. The implications of this on plants’ responses to their environment are discussed. 相似文献
Castilleja tenuiflora is a facultative root hemiparasitic plant that has colonized a disturbed lava field in central Mexico. To determine the effects of hemiparasitism on the population dynamics of the parasite, we identified a set of potential hosts and quantified their effects on the vital rates of C. tenuiflora during 2016–2018. Connections between the roots of the hemiparasite and the hosts were confirmed with a scanning electron microscope. Annual matrices considering two conditions (with and without potential hosts) were built based on vital rates for each year, and annual stochastic finite rate growth rates (λs) were calculated. Plants produced more reproductive structures with hosts than without hosts. A Life Table Response Experiment (LTRE) was performed to compare the contributions of vital rates between conditions. We identified 19 species of potential hosts for this generalist hemiparasite. Stochastic lambda with hosts λs = 1.02 (CI = 0.9999, 1.1) tended to be higher than without them λs = 0.9503 (CI = 0.9055, 0.9981). The highest elasticity values correspond to survival. LTRE indicated that the most important parameters are survival and fecundity; the total contribution of fecundity (0.0192) to the difference in growth was three times lower than that of survival (0.0603). Piqueria trinervia was the most abundant host, and C. tenuiflora had a higher lambda with it than with other species. The anthropogenic movement of species has favoured the introduction of invasive plants worldwide. Invasive plants are frequently released from their natural enemies; however, new associations with generalist herbivores may induce defence mechanisms of non-native plants. Defensive traits are often directly related to the highly competitive ability, but also to potential antagonisms and mutualisms that they can establish with soil microorganisms. Here, we examined whether the intraspecific competition and soil microorganisms influence the morphological and physiological traits of Carpobrotus edulis when is being attacked by the native generalist snail Theba pisana. To achieve this, we grew two C. edulis individuals in separate and same pots filled with live or sterile sand, and with or without T. pisana. Our results indicated that herbivory induced an increase of shoot biomass in attacked C. edulis individuals (i.e., treated donor plants), as well as in un-attacked neighbouring individuals co-growing in the same pot (i.e., untreated recipient plants). Nevertheless, intraspecific competition nor soil microorganisms did not affect the growth of C. edulis despite reduced physiological activity and damage caused by the herbivore. Overall, our findings revealed that C. edulis individuals tolerate snail attack by inducing a compensatory growth response. We conclude that phenotypic plasticity of invasive C. edulis favours tolerance against herbivores, but we also suggest that plant-plant interactions probably determine the plant growth of un-attacked neighbouring C. edulis individuals, thus favouring their invasion mechanisms.

Saliva is a biofluid that maintains the health of oral tissues and the homeostasis of oral microbiota. Studies have demonstrated that Oral squamous cell carcinoma (OSCC) patients have different salivary microbiota than healthy individuals. However, the relationship between these microbial differences and clinicopathological outcomes is still far from conclusive. Herein, we investigate the capability of using metagenomic and metaproteomic saliva profiles to distinguish between Control (C), OSCC without active lesion (L0), and OSCC with active lesion (L1) patients. The results show that there are significantly distinct taxonomies and functional changes in L1 patients compared to C and L0 patients, suggesting compositional modulation of the oral microbiome, as the relative abundances of Centipeda, Veillonella, and Gemella suggested by metagenomics are correlated with tumor size, clinical stage, and active lesion. Metagenomics results also demonstrated that poor overall patient survival is associated with a higher relative abundance of Stenophotromonas, Staphylococcus, Centipeda, Selenomonas, Alloscordovia, and Acitenobacter. Finally, compositional and functional differences in the saliva content by metaproteomics analysis can distinguish healthy individuals from OSCC patients. In summary, our study suggests that oral microbiota and their protein abundance have potential diagnosis and prognosis value for oral cancer patients. Further studies are necessary to understand the role of uniquely detected metaproteins in the microbiota of healthy and OSCC patients as well as the crosstalk between saliva host proteins and the oral microbiome present in OSCC. 相似文献
Microtubule plus-end depolymerization rate is a potentially important target of physiological regulation, but it has been challenging to measure, so its role in spatial organization is poorly understood. Here we apply a method for tracking plus ends based on time difference imaging to measure depolymerization rates in large interphase asters growing in Xenopus egg extract. We observed strong spatial regulation of depolymerization rates, which were higher in the aster interior compared with the periphery, and much less regulation of polymerization or catastrophe rates. We interpret these data in terms of a limiting component model, where aster growth results in lower levels of soluble tubulin and microtubule-associated proteins (MAPs) in the interior cytosol compared with that at the periphery. The steady-state polymer fraction of tubulin was ∼30%, so tubulin is not strongly depleted in the aster interior. We propose that the limiting component for microtubule assembly is a MAP that inhibits depolymerization, and that egg asters are tuned to low microtubule density. 相似文献
