Maintenance of cellular proteostasis relies on efficient clearance of defective gene products. For misfolded secretory proteins, this involves dislocation from the endoplasmic reticulum (ER) into the cytosol followed by proteasomal degradation. However, polypeptide aggregation prevents cytosolic dislocation and instead activates ill‐defined lysosomal catabolic pathways. Here, we describe an ER‐to‐lysosome‐associated degradation pathway (ERLAD) for proteasome‐resistant polymers of alpha1‐antitrypsin Z (ATZ). ERLAD involves the ER‐chaperone calnexin (CNX) and the engagement of the LC3 lipidation machinery by the ER‐resident ER‐phagy receptor FAM134B, echoing the initiation of starvation‐induced, receptor‐mediated ER‐phagy. However, in striking contrast to ER‐phagy, ATZ polymer delivery from the ER lumen to LAMP1/RAB7‐positive endolysosomes for clearance does not require ER capture within autophagosomes. Rather, it relies on vesicular transport where single‐membrane, ER‐derived, ATZ‐containing vesicles release their luminal content within endolysosomes upon membrane:membrane fusion events mediated by the ER‐resident SNARE STX17 and the endolysosomal SNARE VAMP8. These results may help explain the lack of benefits of pharmacologic macroautophagy enhancement that has been reported for some luminal aggregopathies. 相似文献

Total litter fall, over a period of more than 22 months, was 4.22, 3.84 and 4.32 g dry wt. m−2 day−1 in the Avicennia, Sonneratia and Rhizophora forests, respectively. Leaves comprised 40–67% of the total. The Avicennia alba Bl. forest, however, showed a distinct peak from August to October when fruits comprised 50–65% of the total litter. Correlations of total and leaf component litters with the four meterological parameters of wind run, mean temperature, evaporation and rainfall accounted for less than 50% of the variation in litter fall. Wind run had the greatest influence in the seaward Avicennia forest while mean temperature had the greatest influence on the landward Rhizophora forest. 相似文献

Immunofluorescent staining, dot blot hybridization, PCR, random amplified polymorphic DNA (RAPD) markers, and restriction fragment length polymorphism wee used to study the genetic relatedness among mycoplasmalike organisms (MLOs) associated with several geographically diverse grapevine yellows diseases (CA1, CH1, SA1, and SA2 from Bologna, Italy; GYU from Udine, Italy; GYR from Rome, Italy; and GYG from Germany). The relationship between these and MLOs associated with clover phyllody diseases in Italy (CPhB and CPhC) and Canada (CPhCa) was also examined. Two monoclonal antibodies reacted with MLOs of GYU-, CPhB-, and CPhC-infected periwinkles. Dot blot hybridization with two cloned GYU DNA fragments, GYD-1 and GYD-2 inserts, showed that both hybridized with DNAs of GYU-, CPhB-, and CPhC-infected periwinkles but not with those of GYR and CPhCa. In addition, GYD-1 insert hybridized with DNAs of CA1, CH1, SA1, SA2, and GYG. Three primer pairs were developed in PCR experiments for this study. By using primer set GYD2P1F and GYD2P1R, a 600-bp DNA fragment was amplified only when DNAs from GYU-, CPhB-, and CPhC-infected plants were used as templates. With the primer pair GYD2P1F and GYD2P2R, a 550-bp DNA fragment was amplified from GYU, CPhB, CPhC, and GYG. Ovarian activity, assessed by the plasma progesterone concentrations, were studied in four groups of Sardinian breed sheep with different lambing season. The resumption of ovarian activity was shorter and similarly the proportion of ewes which had resumed ovarian activity in response to introduction of rams was greater in sheep with November and December rather than January and March lambings. This study reports 12 novel mutations of the Wilson disease (WD) gene which have been detected by the molecular analysis
of 29 patients of Mediterranean descent carrying uncommon chromosomal haplotypes at the WD locus. These mutations include
two nonsense, one splice site and nine missense. The missense mutations lie in regions of the WD gene critical for its function,
such as the transmembrane region, the transduction domain and the ATP loop and ATP-binding domain, indicating that they are
disease-causing mutations. These new findings improve our knowledge for the role played by functional domains on the ATP7B
function.
