A new family of insecticidal crystal proteins was discovered by screening sporulated Bacillus thuringiensis cultures for oral activity against western corn rootworm (WCR) larvae. B. thuringiensis isolates PS80JJ1, PS149B1, and PS167H2 have WCR insecticidal activity attributable to parasporal inclusion bodies containing proteins with molecular masses of ca. 14 and 44 kDa. The genes encoding these polypeptides reside in apparent operons, and the 14-kDa protein open reading frame (ORF) precedes the 44-kDa protein ORF. Mutagenesis of either gene in the apparent operons dramatically reduced insecticidal activity of the corresponding recombinant B. thuringiensis strain. Bioassays performed with separately expressed, biochemically purified 14- and 44-kDa polypeptides also demonstrated that both proteins are required for WCR mortality. Sequence comparisons with other known B. thuringiensis insecticidal proteins failed to reveal homology with previously described Cry, Cyt, or Vip proteins. However, there is evidence that the 44-kDa polypeptide and the 41.9- and 51.4-kDa binary dipteran insecticidal proteins from Bacillus sphaericus are evolutionarily related. The 14- and 44-kDa polypeptides from isolates PS80JJ1, PS149B1, and PS167H2 have been designated Cry34Aa1, Cry34Ab1, and Cry34Ac1, respectively, and the 44-kDa polypeptides from these isolates have been designated Cry35Aa1, Cry35Ab1, and Cry35Ac1, respectively.  相似文献

A bacteriocin produced by Pseudomonas syringae pv. ciccaronei, used at different purification levels and concentrations in culture and in planta, inhibited the multiplication of P. syringae subsp. savastanoi, the causal agent of olive knot disease, and affected the epiphytic survival of the pathogen on the leaves and twigs of treated olive plants. Treatments with bacteriocin from P. syringae pv. ciccaronei inhibited the formation of overgrowths on olive plants caused by P. syringae subsp. savastanoi strains PVBa229 and PVBa304 inoculated on V-shaped slits and on leaf scars at concentrations of 10(5) and 10(8) CFU ml(-1), respectively. In particular, the application of 6,000 arbitrary units (AU) of crude bacteriocin (dialyzed ammonium sulfate precipitate of culture supernatant) ml(-1) at the inoculated V-shaped slits and leaf scars resulted in the formation of knots with weight values reduced by 81 and 51%, respectively, compared to the control, depending on the strains and inoculation method used. Crude bacteriocin (6,000 AU ml(-1)) was also effective in controlling the multiplication of epiphytic populations of the pathogen. In particular, the bacterial populations recovered after 30 days were at least 350 and 20 times lower than the control populations on twigs and on leaves, respectively. These results suggest that bacteriocin from P. syringae pv. ciccaronei can be used effectively to control the survival of the causal agent of olive knot disease and to prevent its multiplication at inoculation sites.  相似文献

CD40-mediated activation of NF-kappa B in airway epithelial cells   总被引:2,自引:0,他引:2  
We have reported previously that airway epithelial cells (AEC) express CD40 and that activation of this molecule stimulates the expression of inflammatory mediators, including the chemokine RANTES (regulated on activation normal T cell expressed and secreted). Because NF-kappaB regulates the expression of many inflammatory mediators, such as RANTES, we utilized CD40-mediated induction of RANTES expression to investigate the mechanisms that underlie CD40-mediated activation of NF-kappaB in AEC. Results demonstrate that, in AEC, intact NF-kappaB sites were required for CD40-mediated activation of the RANTES promoter. To examine activation of NF-kappaB binding directly, electrophoretic mobility shift analyses were performed. These analyses revealed that CD40 ligation stimulated NF-kappaB binding and that the activated NF-kappaB complexes were composed of p65 subunits. Additional studies focused on the CD40-triggered signaling pathways that facilitate NF-kappaB activation. Findings show that CD40 engagement activated the IkappaB kinases IKK-alpha and IKK-beta and stimulated IkappaBalpha phosphorylation. Analyses also examined the role of tumor necrosis factor-associated factor (TRAF) molecules in CD40-mediated NF-kappaB activation within AEC. Stable transfectants expressing wild-type or mutant forms of the cytoplasmic domain of CD40 suggested that TRAF3, but not TRAF2, binding was essential for CD40-mediated RANTES expression. Further studies indicated that exogenous expression of wild-type TRAF3 enhanced activation of the RANTES promoter, whereas exogenous expression of wild-type TRAF2 inhibited this activation; TRAF3-mediated enhancement was dependent upon NF-kappaB. Together, these findings suggest that, in AEC, ligation of CD40 regulates the expression of inflammatory mediators, such as RANTES, via activation of NF-kappaB. Moreover, these results suggest that CD40-mediated signaling in AEC differs with previously reported findings observed in other cell models, such as B lymphocytes.  相似文献

Neurons of the mammalian central nervous system (CNS) are an essential and largely nonrenewable cell population. Thus, virus infections that result in neuronal depletion, either by virus-mediated cell death or by induction of the cytolytic immune response, could cause permanent neurological impairment of the host. In a transgenic mouse model of measles virus (MV) infection of neurons, we have previously shown that the host T-cell response was required for resolution of infection in susceptible adult mice. In this report, we show that this protective response did not result in neuronal death, even during the peak of T-cell infiltration into the brain parenchyma. When susceptible mice were intercrossed with specific immune knockout mice, a critical role for gamma interferon (IFN-gamma) was identified in protection against MV infection and CNS disease. Moreover, the addition of previously activated splenocytes or recombinant murine IFN-gamma to MV-infected primary neurons resulted in the inhibition of viral replication in the absence of neuronal death. Together, these data support the hypothesis that the host immune response can promote viral clearance without concomitant neuronal loss, a process that appears to be mediated by cytokines.  相似文献

In the majority of patients with Pelizaeus-Merzbacher disease, duplication of the proteolipid protein gene PLP1 is responsible, whereas deletion of PLP1 is infrequent. Genomic mechanisms for these submicroscopic chromosomal rearrangements remain unknown. We identified three families with PLP1 deletions (including one family described elsewhere) that arose by three distinct processes. In one family, PLP1 deletion resulted from a maternal balanced submicroscopic insertional translocation of the entire PLP1 gene to the telomere of chromosome 19. PLP1 on the 19qtel is probably inactive by virtue of a position effect, because a healthy male sibling carries the same der(19) chromosome along with a normal X chromosome. Genomic mapping of the deleted segments revealed that the deletions are smaller than most of the PLP1 duplications and involve only two other genes. We hypothesize that the deletion is infrequent, because only the smaller deletions can avoid causing either infertility or lethality. Analyses of the DNA sequence flanking the deletion breakpoints revealed Alu-Alu recombination in the family with translocation. In the other two families, no homologous sequence flanking the breakpoints was found, but the distal breakpoints were embedded in novel low-copy repeats, suggesting the potential involvement of genome architecture in stimulating these rearrangements. In one family, junction sequences revealed a complex recombination event. Our data suggest that PLP1 deletions are likely caused by nonhomologous end joining.  相似文献

Sexual selection for divergent female preferences has been proposed to stimulate speciation. We tested this basic model by selecting for divergence in the courtship repertoire of the house fly Musca domestica L. Specifically, we subjected replicate strains to artificial selection for differentiation along the first two principal components of the phenotypic intercorrelation structure of five courtship traits. Highly significant differentiation in courtship repertoire resulted, and the magnitude of the selection response was highest along the first principal component (representing the ‘size’, or general intensity, of courtship). Videotaped matings of the crosses between divergent lines (i.e. males of one strain mating with females from a different line) showed that the selection responses in the intensities of male performances were due to shifts in female preferences. In particular, the males were able to accommodate the demands of ‘foreign’ females (as well as their own) in the no-choice situation (i.e. only one male and one female per mating chamber). In contrast to this plasticity of the males, the females were consistent in their differential resistance responses, regardless of the type of male involved in the courtship. Multiple-choice mate choice tests revealed significant reproductive isolation among some lines, although the effect was asymmetrical. The patterns of nonrandom mating were largely due to females from more genetically healthy lines (i.e. with overall high mating propensity) discriminating against males from populations with more inbreeding depression. We suggest that the inability to achieve true (symmetrical) reproductive isolation could have been due to low evolutionary potential in the ‘shape’ of courtship, as defined by the second principal component. The morphology-performance-fitness paradigm is usually exploredby determining whether natural or "phenotypically engineered"variation among individuals in morphology (physiology) or performancecovaries with an index of fitness such as survival. Here westudy between-line covariation between performance and fitnessfor 44 lines of flies that had undergone mutation accumulation(in the absence of natural selection) on the second chromosomefor 62 generations, plus 13 control lines. These mutation accumulation(MA) lines were known to have reduced competitive fitness andlife history scores, and to have positive between-line covariancesamong life history traits. We measured several performance traitsof larvae and adults (and a life history trait), examined covariancesamong those trait means, and also examined covariances of traitswith competitive fitness. MA lines had significantly lower performancesthan did control lines in most traits. However, because controllines had been unknowingly contaminated, a conclusion that MAreduces performance must be tentative. Correlations among performancetraits were highly variable in sign, suggesting that MA doesnot negatively affect all traits equivalently. Even so, correlationmatrices for MA and for control lines were very similar. Inbivariate comparisons, only one performance trait (a "get-a-gripindex," which measures the ability of a falling fly to catchitself on baffles) was positively correlated with competitivefitness. Multivariate analyses again suggested the importanceprimarily of get-a-grip. Two main patterns emerge from thisstudy. First, MA negatively affects diverse aspects of physiologicalperformance, but does so differentially across traits. Second,except for GAG, MA-induced variation in performance is at bestweakly correlated with competitive fitness.  相似文献

The societal benefits of hydropower systems (e.g., relatively clean electrical power, water supply, flood control, and recreation) come with a cost to native stream fishes. We reviewed and synthesized the literature on hydropower-related pulsed flows to guide resource managers in addressing significant impacts while avoiding unnecessary curtailment of hydropower operations. Dams may release pulsed flows in response to needs for peaking power, recreational flows, reservoir storage adjustment for flood control, or to mimic natural peaks in the hydrograph. Depending on timing, frequency, duration, and magnitude, pulsed flows can have adverse or beneficial short and long-term effects on resident or migratory stream fishes. Adverse effects include direct impacts to fish populations due to (1) stranding of fishes along the changing channel margins, (2) downstream displacement of fishes, and (3) reduced spawning and rearing success due to redd/nest dewatering and untimely or obstructed migration. Beneficial effects include: (1) maintenance of habitat for spawning and rearing, and (2) biological cues to trigger spawning, hatching, and migration. We developed a basic conceptual model to predict the effects of different types of pulsed flow, identified gaps in knowledge, and identified research activities to address these gaps. There is a clear need for a quantitative framework incorporating mathematical representations of field and laboratory results on flow, temperature, habitat structure, fish life stages by season, fish population dynamics, and multiple fish species, which can be used to predict outcomes and design mitigation strategies in other regulated streams experiencing pulsed flows.  相似文献

The identification, production, and potential electron conductivity of bacterial extracellular nanofilaments is an area of great study, specifically in Shewanella oneidensis MR-1. While some studies focus on nanofilaments attached to the cellular body, many studies require the removal of these nanofilaments for downstream applications. The removal of nanofilaments from S. oneidensis MR-1 for further study requires not only that the nanofilaments be detached, but also for the cell bodies to remain intact. This is a study to both qualitatively (AFM) and quantitatively (LC/MS-MS) assess several nanofilament shearing methods and determine the optimal procedure. The best method for nanofilament removal, as judged by maximizing extracellular filamentous proteins and minimizing membrane and intracellular proteins, is vortexing a washed cell culture for 10 min.  相似文献
