Potent and durable vaccine responses will be required for control of malaria caused by Plasmodium falciparum (Pf). RTS,S/AS01 is the first, and to date, the only vaccine that has demonstrated significant reduction of clinical and severe malaria in endemic cohorts in Phase 3 trials. Although the vaccine is protective, efficacy declines over time with kinetics paralleling the decline in antibody responses to the Pf circumsporozoite protein (PfCSP). Although most attention has focused on antibodies to repeat motifs on PfCSP, antibodies to other regions may play a role in protection. Here, we expressed and characterized seven monoclonal antibodies to the C-terminal domain of CSP (ctCSP) from volunteers immunized with RTS,S/AS01. Competition and crystal structure studies indicated that the antibodies target two different sites on opposite faces of ctCSP. One site contains a polymorphic region (denoted α-ctCSP) and has been previously characterized, whereas the second is a previously undescribed site on the conserved β-sheet face of the ctCSP (denoted β-ctCSP). Antibodies to the β-ctCSP site exhibited broad reactivity with a diverse panel of ctCSP peptides whose sequences were derived from field isolates of P. falciparum whereas antibodies to the α-ctCSP site showed very limited cross reactivity. Importantly, an antibody to the β-site demonstrated inhibition activity against malaria infection in a murine model. This study identifies a previously unidentified conserved epitope on CSP that could be targeted by prophylactic antibodies and exploited in structure-based vaccine design.  相似文献

Predicting broad-scale patterns of biodiversity is challenging, particularly in ecosystems where traditional methods of quantifying habitat structure fail to capture subtle but potentially important variation within habitat types. With the unprecedented rate at which global biodiversity is declining, there is a strong need for improvement in methods for discerning broad-scale differences in habitat quality. Here, we test the importance of habitat structure (i.e. fine-scale spatial variability in plant growth forms) and plant productivity (i.e. amount of green biomass) for predicting avian biodiversity. We used image texture (i.e. a surrogate for habitat structure) and vegetation indices (i.e., surrogates for plant productivity) derived from Landsat Thematic Mapper (TM) data for predicting bird species richness patterns in the northern Chihuahuan Desert of New Mexico. Bird species richness was summarized for forty-two 108 ha plots in the McGregor Range of Fort Bliss Military Reserve between 1996 and 1998. Six Landsat TM bands and the normalized difference vegetation index (NDVI) were used to calculate first-order and second-order image textures measures. The relationship between bird species richness versus image texture and productivity (mean NDVI) was assessed using Bayesian model averaging. The predictive ability of the models was evaluated using leave-one-out cross-validation. Texture of NDVI predicted bird species richness better than texture of individual Landsat TM bands and accounted for up to 82.3% of the variability in species richness. Combining habitat structure and productivity measures accounted for up to 87.4% of the variability in bird species richness. Our results highlight that texture measures from Landsat TM imagery were useful for predicting patterns of bird species richness in semi-arid ecosystems and that image texture is a promising tool when assessing broad-scale patterns of biodiversity using remotely sensed data.  相似文献

The regulation of population processes for most organisms depends upon the strength and rate of feedback between resources and consumers. We conducted an experimental manipulation of leaf packs in stream channels, a patchy and ephemeral resource, which is consumed by a number of detritivorous invertebrates. We reduced the number of available food patches (red alder leaf packs) by half and then measured a variety of community responses, including emigration rate, aggregation on remaining food patches, decomposition rate of food patches, and species-specific differences in these responses. Replacement of removed leaf packs with polyester mimics resulted in no statistical difference in emigration rates or aggregation on remaining resources when compared to those removal channels without replacement. These results indicate that leaf packs are not used primarily for refuge. In the removal channels (including those with leaf pack mimics) emigration rate nearly doubled relative to control channels. Those invertebrates that did not emigrate from removal channels aggregated on remaining leaf packs, which led to more rapid decomposition of leaf packs relative to control channels. The increase in emigration rate only became apparent 2-3 days after the manipulation, presumably because animals colonized the remaining leaf packs and did not emigrate until food patch value per individual had been reduced by higher densities or due to increased discharge. Discharge through the channels increased slightly starting 3 days after the manipulation, resulting in increased emigration rates in all channels. Despite the increase in discharge, the effect of the manipulation remained strong. These results show that stream invertebrates colonizing leaf packs responded in predictable ways to a short-term reduction in food resources which would be adaptive in a system which is heterogeneous in space and time.  相似文献

The distribution of phenoloxidase and polyphenols have been studied during cuticle formation at the 4th to 5th molt in Colpodes ethius. Cuticular phenoloxidases arise in the epidermis in cisternae of the rough endoplasmic reticulum, pass through the Golgi complex and are transported to the apical face in secretory vesicles. From the cuticular environment some enzyme is pinocytosed and broken down in the apical multivesicular bodies. Phenoloxidase and polyphenols are present during the formation of the cuticulin layer which also reacts as if it were at least partly composed of a phenoloxidase. The rest of the epicuticle incorporates phenoloxidase as it is deposited, particularly that over the dorsal tubercles which later melanize. Polyphenols do not appear until shortly before ecdysis. They are associated with the epicuticular filaments in both epicuticle and presumptive epocuticle. It is proposed that the epicuticular filaments may arise as liquid crystals with a protein component which becomes stabilized like the rest of the cuticle. These structures could provide a channel for the movement of both lipids and quinones to the surface. Phenoloxidases may pass through fibrous cuticle to be deposited as part of the epicuticle but are incorporated in fibrous cuticle scheduled for sclerotization. The time of stabilization is determined by the availability of polyphenols.  相似文献

The apical plasma membranes of Calpodes epidermal cells have small fattened areas or plaques with an extra density upon their cytoplasmic face. The plaques are typically at the tips of microvilli. The are present during the deposition of fibrous cuticle and the cuticulin layer. Since the plaques are close (less than 15nm) to the sites where these kinds of cuticle first appear, they are presumed to have a role in their synthesis and/or deposition and orientation. When fifth stage larval cuticle deposition ceases prior to pupation, the plaques are lost as the area of the apical plasma membrane is reduced. The plaques pass from the surface into pinocytosis vesicles and multivesicular bodies where they are presumably digested. The loss of plaques occurs as the blood level of moulting hormone reaches a peak at the critical period after which the prothoracic glands are no longer needed for pupation. Apolysis or separation of the epidermis from the old cuticle is the stage when plaques are absent, the old ones have been lost but the new ones have yet to form. After the critical period, the epidermis prepared for pupation with a phase of elevated RNA synthesis at the end of which plaques and microvilli reform in time to secrete the new cuticulin layer and later the fibrous cuticle of the pharate pupa. There is a new generation of plaques for each moult and succeeding intermoult and each generation is involved in two kinds of cuticle deposition before involution and redifferentiation.  相似文献

In the present study, a dynamic mathematical model for the growth of granulocyte progenitor cells in the hematopoietic process is developed based on the principles of diffusion and chemical reaction. This model simulates granulocyte progenitor cell growth and oxygen consumption in a three-dimensional (3-D) perfusion bioreactor. Material balances on cells are coupled to the nutrient balances in 3-D matrices to determine the effects of transport limitations on cell growth. The method of volume averaging is used to formulate the material balances for the cells and the nutrients in the porous matrix containing the cells. All model parameters are obtained from the literature. The maximum cell volume fraction reached when oxygen is depleted in the cell layer at 15 days and is nearly 0.63, corresponding to a cell density of 2.25 x 10(8) cells/mL. The substrate inhibition kinetics for cell growth lead to complex effects with respect to the roles of oxygen concentration and supply by convection and diffusion on cell growth. Variation in the height of the liquid layer above the cell matrix where nutrient supply is introduced affected the relative and absolute amounts of oxygen supply by hydrodynamic flow and by diffusion across a gas permeable FEP membrane. Mass transfer restrictions of the FEP membrane are considerable, and the supply of oxygen by convection is essential to achieve higher levels of cell growth. A maximum growth rate occurs at a specific flow rate. For flow rates higher than this optimal, the high oxygen concentration led to growth inhibition and for lower flow rates growth limitations occur due to insufficient oxygen supply. Because of the nonlinear effects of the autocatalytic substrate inhibition growth kinetics coupled to the convective transport, the rate of growth at this optimal flow rate is higher than that in a corresponding well-mixed reactor where oxygen concentration is set at the maximum indicated by the inhibitory kinetics.  相似文献

Most studies of reinforcement have focused on the evolution of either female choice or male mating cues, following the long-held view in sexual selection theory that mating mistakes are typically more costly for females than for males. However, factors such as conspecific sperm precedence can buffer females against the cost of mating mistakes, suggesting that in some hybrid zones mating mistakes may be more costly for males than for females. Thus, the historical bias in reinforcement research may underestimate its frequency. In this study, we present evidence that reinforcement has driven the evolution of male choice in a hybrid zone between the highly promiscuous leaf beetles Chrysochus cobaltinus and C. auratus, the hybrids of which have extremely low fitness. In addition, there is evidence for male choice in these beetles and that male mating mistakes may be costly, due to reduced opportunities to mate with conspecific females. The present study combines laboratory and field methods to quantify the strength of sexual isolation, test the hypothesis of reproductive character displacement, and assess the link between relative abundance and the strength of selection against hybridization. We document that, while sexual isolation is weak, it is sufficient to produce positive assortative mating. In addition, reproductive character displacement was only detected in the relatively rare species. The strong postzygotic barriers in this system are sufficient to generate the bimodality that characterizes this hybrid zone, but the weak sexual isolation is not, calling into question whether strong prezygotic isolation is necessary for the maintenance of bimodality. Growing evidence that the cost of mating mistakes is sufficient to shape the evolution of male mate choice suggests that the reinforcement of male mate choice may prove to be a widespread occurrence.  相似文献
