AIM: Identification of the predominant lactic acid bacteria (LAB) involved in spontaneous fermentations of Almagro eggplants, and evaluation of the biodiversity by molecular typing. METHODS AND RESULTS: Almagro eggplant fermentations in three factories (A, B and C) enjoying Protected Designation of Origin (PDO) status were monitored by chemical and microbiological analysis of brines. LAB isolates from brines were identified by phenotypic analysis and by species-specific PCR reactions and typed by randomly amplified polymorphic DNA (RAPD)-PCR. All isolates from factories A and C belonged to the genus Lactobacillus (Lact.), whereas isolates from factory B belonged to Lactobacillus (50%), Leuconostoc (Ln.) (25%) and Lactococcus (Lc.) (25%); 1.9% of this microbiota was considered cosmopolitan. The genera Leuconostoc and Lactococcus and the species Lact. acidophilus and Lact. paracasei had never previously been reported in Almagro eggplant fermentations. CONCLUSION: Considerable differences in the composition of the lactic acid microbiota participating in the Almagro eggplant fermentations exist. Brine NaCl concentration has a notable influence both in number and in the species participating. SIGNIFICANCE AND IMPACT OF THE STUDY: The original aspect of this work consists of an ecological study of the LAB taking part in spontaneous Almagro eggplant fermentations from different factories. Participation of Leuconostoc and Lactococcus species and of Lact. acidophilus and Lact. paracasei, which had never before been described for this pickle, and the evidence that a lactic fermentation does not always take place, were the most relevant results.  相似文献

Abstract We conducted a 2-year in situ experiment to test the capacity of iron additions to reverse the decline experienced by a Posidonia oceanica meadow colonizing carbonate, iron poor sediment. Iron additions improved the sediment conditions that support seagrass growth by decreasing the sediment sulfide concentration and sulfate reduction rates, and decreased sulfide intrusion into the plants. Iron additions for 2 years did not significantly change survivorship of shoots present at the onset of the experiment, but significantly increased shoot recruitment and survivorship of shoots recruited during the experiment. After 2 years, iron additions reversed seagrass decline and yielded positive growth rates of shoots relative to control populations where seagrass continued to decline. This research demonstrates that seagrass decline in carbonate sediments may be reversed by targeting critical processes such are sediment sulfide pools and seagrass nutritional status, controlling the functioning of the ecosystem.  相似文献

RepB initiates plasmid rolling‐circle replication by binding to a triple 11‐bp direct repeat (bind locus) and cleaving the DNA at a specific distant site located in a hairpin loop within the nic locus of the origin. The structure of native full‐length RepB reveals a hexameric ring molecule, where each protomer has two domains. The origin‐binding and catalytic domains show a three‐layer α–β–α sandwich fold. The active site is positioned at one of the faces of the β‐sheet and coordinates a Mn2+ ion at short distance from the essential nucleophilic Y99. The oligomerization domains (ODs), each consisting of four α‐helices, together define a compact ring with a central channel, a feature found in ring helicases. The toroidal arrangement of RepB suggests that, similar to ring helicases, it encircles one of the DNA strands during replication to confer processivity to the replisome complex. The catalytic domains appear to be highly mobile with respect to ODs. This mobility may account for the adaptation of the protein to two distinct DNA recognition sites.  相似文献

Launaea cervicornis (Boiss.) Font Quer & Rothmaler, an endemic of the islands of Majorca and Minorca (Balearic Islands), is a keystone species of the cushion-like plant communities of the coastal rocky places. The study of its reproductive biology shows the existence of differential strategies, with reference to other species of the genus. The principal features are: (1) the absence of continuous iteroparity; (2) short average duration of the flowering period (36.8 days); (3) high value of intrapopulational flowering overlap index (0.86); (4) high levels of aptitude to produce fruits having atelechory as preferential mechanism of dispersion; and (5) germination capacity is increased during the autumn of the first year (fruits are not capable of joining the seedbank of the soil). These characteristics are considered to be determinants for its adaptation to the peculiar habitat in which it prospers.  相似文献

To help clarify the control of arginine synthesis in Thermotoga maritima, the putative gene (argB) for N-acetyl-L-glutamate kinase (NAGK) from this microorganism was cloned and overexpressed, and the resulting protein was purified and shown to be a highly thermostable and specific NAGK that is potently and selectively inhibited by arginine. Therefore, NAGK is in T. maritima the feedback control point of arginine synthesis, a process that in this organism involves acetyl group recycling and appears not to involve classical acetylglutamate synthase. The inhibition of NAGK by arginine was found to be pH independent and to depend sigmoidally on the concentration of arginine, with a Hill coefficient (N) of approximately 4, and the 50% inhibitory arginine concentration (I0.5) was shown to increase with temperature, approaching above 65 degrees C the I0.50 observed at 37 degrees C with the mesophilic NAGK of Pseudomonas aeruginosa (the best-studied arginine-inhibitable NAGK). At 75 degrees C, the inhibition by arginine of T. maritima NAGK was due to a large increase in the Km for acetylglutamate triggered by the inhibitor, but at 37 degrees C arginine also substantially decreased the Vmax of the enzyme. The NAGKs of T. maritima and P. aeruginosa behaved in gel filtration as hexamers, justifying the sigmoidicity and high Hill coefficient of arginine inhibition, and arginine or the substrates failed to disaggregate these enzymes. In contrast, Escherichia coli NAGK is not inhibited by arginine and is dimeric, and thus the hexameric architecture may be an important determinant of arginine sensitivity. Potential thermostability determinants of T. maritima NAGK are also discussed.  相似文献

OBJECTIVE: To analyze the relationship between the peroxisome proliferator-activated receptor-gamma (PPARgamma2) Pro12Ala variant and type-2 diabetes mellitus and its correlation with some cytokine determinants of insulin resistance such as tumor necrosis factor (TNF)-alpha and leptin. METHODS: The PPARgamma2 Pro12Ala genetic polymorphism was studied in 167 type-2 diabetic patients and 63 healthy controls. Serum leptin and plasma-soluble TNF-R2 were measured. RESULTS: Women carriers of the Pro12Ala mutation exhibited higher leptin levels than women non-carriers (median 31.4 vs. 17.5 ng/ml; p < 0.005). sTNF-R2 levels did not show differences between the two genotypes. Analysis by the multiple linear regression model of leptin-body mass index controlled by the PPARgamma2 genotype showed that leptin levels were determined by the Pro12Ala mutation in type-2 diabetic women but not in men. CONCLUSIONS: PPARgamma2 seems to be implicated in leptin homeostasis in type-2 diabetic women.  相似文献

The cytokeratin intermediate filaments have a relevant role in the proliferation and differentiation processes of epithelial cells. To provide information about the role of K8 cytokeratin during the auditory receptor differentiation, two groups of adult mice were used: TGK8-4 transgenic and control animals. The TGK8-4 transgenic mice contained 12 kb of K8 human cytokeratin (HK8) locus (Casanova et al., 1995, 1999). The functional activity of the auditory receptor was analyzed by auditory thresholds. Morphological studies demonstrate that the auditory receptors of the TGK8-4 transgenic mice are highly immature. Immunocytochemical studies were made by using two monoclonal antibodies: CAM 5-2 (recognizing K8 human cytokeratin) and Troma-1 (recognizing both mouse and human K8 cytokeratin). These demonstrated significant differences between the auditory receptors of the transgenic mice and the control mice. These functional and morphological differences clearly suggest that K8 cytokeratin has a relevant role during the differentiation and tridimensional organization of the sensory and the supporting cells of the auditory receptor.  相似文献

Variations in standard length ( L S), gape size ( S G) and jaw length ( L J) were studied in larval and juvenile gadoids (cod Gadus morhua , haddock Melanogrammus aeglefinus and whiting Merlangius merlangus ) from 4 to 70 mm. The increase in S G and L J was not linear with respect to L S. The relationship was best described by segmented regression lines in all three species, with an inflection point at c . 10·5 mm. The S G and L J increased more rapidly in relation to larval L S for individuals smaller than this inflection point size. The rates of increase slowed significantly post-inflection, an effect more noticeable in S G data compared to L J data. In each case, the inflection point fell in the intermediate period of development between the larval and juvenile stages, which could be considered as metamorphosis. Published equations that have been used to predict S G from L J lead to the overestimation of gape. New relationships are presented, which may be used to predict S G from measurements of either L S or upper jaw length in cod, haddock and whiting.  相似文献
