Summary Using straw columns colonized by the lignocellulytic fungus Pleurotus cornucopiae, translocation of 109Cd and 203Hg in the substrate-mycelium complex and via the substrate-mycelium complex into the fruiting bodies was studied. The translocation patterns generated were metal specific and were influenced by the temperature and the physiological conditions of the mycelium (growing mycelium, established mycelium, reproductive stage).Under all conditions, generally more mercury than cadmium was translocated. In growing mycelia, for instance, an average of about seven times more mercury than cadmium was translocated. Translocation was greatly enhanced, when fruiting bodies were present. Up to 7% and 20% (average: 3.5% and 12%) of the applied cadmium and mercury, respectively, were found in the fruiting bodies. In old columns bearing fruiting bodies (colonized for more than 50 days by the fungus) considerably more heavy metal (up to 45% of the applied radioactivity) was released from the point of application than in younger columns.With one exception, no substantial differences in the translocation patterns of the label in relation to the direction of mycelial growth could be detected. 相似文献

Summary Linear measurement of blocks of constitutive heterochromatin and the euchromatin portion 1q-h in three members of a family was used to study the dependence of the size of C blocks on the degree of chromosomal contraction. The results demonstrate that the size of heterochromatin portions decrease regularly with an increases of the degree of euchromatin contraction. The dependence was found to be linear, except for mitoses with an extremely high or low degree of contraction. Summary A comparsion of the Feulgen hydrolysis curves and the chromatin compactness of the liver cell nuclei of young and old rats was made. It was found that the rate of DNA depurination and chromatin compactness are higher in the liver cell nuclei of old rats, both in di-and tetraploidal cells. By using the choline starvation process it is possible to deplete the membranes of Neurospora crassa choline auxotroph of phosphatidylcholine, without affecting the viability of germinated spores or whole mycelium. Spin label probes were used to examine the possible dependence of the physical state of cellular lipids on the presence of phosphatidylcholine in the membranes.Increased freedom of rotational motion of lipid soluble probes was regularly detected in choline-starved mycelium. The accumulation of neutral lipids (mostly triglycerides) in bulk form was also observed during the choline starvation process. The experiments with isolated and separated lipid classes indicated that the observed increase in fluidity of lipids in choline-starved mycelium is partly due to the difference in physical properties between bulk lipids and membrane lipids. Zooplankton of the fish pond reveals a two-year periodicity, which is induced by the two-year cycle of fishery management. In the first year of each cycle, the biomass of large cladocerans prevailed — mainly adult specimens of the species Daphnia pulicaria Forbes, which were absent in the second year. The species Daphnia galeata Sars was present in both years, however, it is more numerous in the second year. We have characterized the occurrence and expression of multiple acyl carrier protein (ACP) isoforms in Arabidopsis thaliana (L.) Heynh ecotype Columbia. Immunoblot analysis of ACPs from Arabidopsis tissues separated by native polyacrylamide gel electrophoresis and 1 molar urea polyacrylamide gel electrophoresis revealed a complex pattern of multiple ACP isoforms. All tissues examined (leaves, roots, and seeds) expressed at least three forms of ACP. The immunoblot identifications of ACP bands were confirmed by acylation of ACP extracts with Escherichia coli acyl-ACP synthetase. A full-length cDNA clone has been isolated that has 70% identity with a previously characterized Arabidopsis genomic ACP clone (ACP-1) (MA Post-Beittenmiller, A Hloušek-Radojčić, JB Ohlrogge [1989] Nucleic Acids Res 17: 1777). Based on RNA blot analysis, the cDNA clone represents an ACP that is expressed in leaves, seeds, and roots. In order to identify the protein products of each known ACP gene, their mature coding sequences have been expressed in E. coli. Using polymerase chain reactions, exons II and III of the genomic ACP-1 clone and the mature coding sequences of the ACP-2 cDNA clone were subcloned into E. coli expression vectors. Site-directed mutagenesis was used to convert the amino acid sequence of the ACP-2 cDNA clone to that of the A2 clone of Lamppa and Jacks ([1991] Plant Mol Biol 16: 469-474), ACP-3. The three E. coli-expressed proteins have different mobilities on polyacrylamide gel electrophoresis gels and each comigrates with a different Arabidopsis ACP isoform expressed in leaves, seeds, and roots. Thus, all of the three cloned ACPs appear to be constitutively expressed Arabidopsis ACPs. The selenium (Se) contents in common cereals in endemic and nonendemic areas in Serbia are very low. Plasma Se levels of both
patients and healthy subjects, were also low, reflecting low Se intakes. Patients with Balkan endemic nephropathy (BEN) had
significantly lower (p<0.05) plasma Se levels than healthy individuals, both from regions close to endemic areas, and from Belgrade. Mean plasma
Se of BEN patients was slightly but insignificantly higher in samples taken immediately after dialysis than in those taken
before, suggesting that very little of the Se present in plasma is dialyzable. Plasma SeGSH-Px activities before and after
hemodialysis in both BEN and Nonendemic chronic renal failure (NCRF) patients were not significantly different, but BEN patients
had lower enzyme activities than those with NCRF and healthy controls. In BEN patients, a significant correlation between
plasma Se and SeGSH-Px activity was found.
