We present evidence for the coexistence and coevolution of antibiotic resistance and biosynthesis genes in soil bacteria. The distribution of the streptomycin (strA) and viomycin (vph) resistance genes was examined in Streptomyces isolates. strA and vph were found either within a biosynthetic gene cluster or independently. Streptomyces griseus strains possessing the streptomycin cluster formed part of a clonal complex. All S. griseus strains possessing solely strA belonged to two clades; both were closely related to the streptomycin producers. Other more distantly related S. griseus strains did not contain strA. S. griseus strains with only vph also formed two clades, but they were more distantly related to the producers and to one another. The expression of the strA gene was constitutive in a resistance‐only strain whereas streptomycin producers showed peak strA expression in late log phase that correlates with the switch on of streptomycin biosynthesis. While there is evidence that antibiotics have diverse roles in nature, our data clearly support the coevolution of resistance in the presence of antibiotic biosynthetic capability within closely related soil dwelling bacteria. This reinforces the view that, for some antibiotics at least, the primary role is one of antibiosis during competition in soil for resources.  相似文献

  1. Landscape ecology and its emphasis on relationships between spatial heterogeneity, scale, and ecological processes can be applied to manage stream ecosystems as riverscapes. Hierarchy theory, a central tenet of riverscape ecology, allows for investigating ecological relationships between aquatic organisms and habitat by exploring the nested interactions inherent in reaches, segments, and basins. Arkansas darter Etheostoma cragini is a Great Plains fish endemic to the Arkansas River basin and is listed as in need of conservation in all five states within its range in North America. The aim of this study was to develop empirical models describing relationships between Arkansas darter abundance and local habitats, and identify the appropriate scale(s) for assessing and managing populations.

We used Arkansas darter abundance data collected from stream sample units as a fixed grain size (102 m) and compiled datasets at three hierarchical spatial extents, including reach (103 m), nested within segment (104 m), nested within basin (105 m) in southcentral Kansas. We fit generalised linear mixed models (GLMMs) to assess scale‐specific relationships between Arkansas darter abundance and local habitat dimensions (stream depth and width) and heterogeneity (canopy cover, overhanging vegetation, and woody structure).
  3. Habitat parameters included in best‐supported GLMMs differed among extents. Variation in Arkansas darter abundance was explained by canopy cover and channel depth at the reach (54% explained) and segment (56%) extents, and stream width and depth at the basin extent (37%). Partial dependence plots from multiple regression GLMMs revealed that Arkansas darter abundance was negatively correlated with increasing channel depth across all extents, increasing canopy cover at reach and segment extents, and increasing channel width at the basin extent.
  4. Our results highlight scale‐specific relationships between Arkansas darter abundance and local habitats across multiple spatial extents. These findings provide direction for assessing stream fish ecology pattern‐process relationships at the appropriate spatial scales and benefit native fish conservation by highlighting the application of hierarchy theory to address management challenges.

Pioneer species are fast-growing, short-lived gap exploiters. They are prime candidates for neutral dynamics because they contain ecologically similar species whose low adult density is likely to cause widespread recruitment limitation, which slows competitive dynamics. However, many pioneer guilds appear to be differentiated according to seed size. In this paper, we compare predictions from a neutral model of community structure with three niche-based models in which trade-offs involving seed size form the basis of niche differentiation. We test these predictions using sowing experiments with a guild of seven pioneer species from chalk grassland. We find strong evidence for niche structure based on seed size: specifically large-seeded species produce fewer seeds but have a greater chance of establishing on a per-seed basis. Their advantage in establishment arises because there are more microsites suitable for their germination and early establishment and not directly through competition with other seedlings. In fact, seedling densities of all species were equally suppressed by the addition of competitors' seeds. By the adult stage, despite using very high sowing densities, there were no detectable effects of interspecific competition on any species. The lack of interspecific effects indicates that niche differentiation, rather than neutrality, prevails.  相似文献

5-Lipoxygenase (5-lipox) has been purified to homogeneity from the 20,000 xg supernatant of sonicated rat basophilic leukemia (RBL-1) cells using a 4-step procedure. Purification was achieved primarily through the use of anion-exchange HPLC on two different media. Using the supernatant from 1 × 10 9 cells, approximately 33 μg of the enzyme can be routinely isolated with an estimated net yield of 5–10%. Purified 5-lipox consists of a single Mr 73,000 band on SDS gels (reduced or unreduced). When the purified enzyme was incubated with radiolabeled arachidonic acid and products analyzed by both straight phase and reversed phase HPLC, 5-hydroperoxyeicosate-traenoic acid (5-HPETE) was the only enzymatic product detected. The purified enzyme exhibits the same characteristic lag phase and premature cessation of reaction as does the 5-lipox activity seen in crude cell homogenates.  相似文献

Nonalcoholic fatty liver disease (NAFLD) has been associated with reduced growth hormone levels and signaling. Such hormonal changes also occur in metabolic acidosis. Since mild metabolic acidosis can be diet induced, diet-induced acid load may constitute a nutritional factor with possible influence on NAFLD development. This study explored whether a higher diet-induced acid load is associated with an increased likelihood of NAFLD. Apparently healthy Chinese adults (330 male, 463 female) aged 19-72 years were recruited through population screening between 2008 and 2010 in a cross-sectional population-based study in Hong Kong. Estimated net endogenous acid production (NEAP) was calculated using Frassetto’s method and potential renal acid load (PRAL) was calculated using Remer’s method based on dietary data from a food frequency questionnaire. NAFLD was defined as intrahepatic triglyceride content at >5% by proton-magnetic resonance spectroscopy. Possible advanced fibrosis was defined as liver stiffness at >7.9 kPa by transient elastography. Multivariate logistic regression models were used to examine the association between each measure of dietary acid load and prevalent NAFLD or possible advanced fibrosis with adjustment for potential anthropometric and lifestyle factors. 220 subjects (27.7%) were diagnosed with NAFLD. Estimated NEAP was positively associated with the likelihood of having NAFLD after adjustment for age, sex, body mass index, current drinker status and the presence of metabolic syndrome [OR (95% CI) = 1.25 (1.02-1.52), p = 0.022]. The association was slightly attenuated but remained significant when the model was further adjusted for other dietary variables. No association between PRAL and NAFLD prevalence was observed. Both estimated NEAP and PRAL were not associated with the presence of possible advance fibrosis. Our findings suggest that there may be a modest association between diet-induced acid load and NAFLD. More studies are needed to ascertain the link between diet-induced acid load and NAFLD and to investigate the underlying mechanisms.  相似文献

Closing the gap between the health and well-being status of Indigenous people living in remote areas of northern Australia and non-Indigenous Australians has long been a major target of federal health policy. With climate projections suggesting large increases in hot spells in desert regions and more extremes in rainfall in other areas of the north, direct and indirect impacts resulting from these changes are likely to further entrench this health and well-being disparity. This paper argues that it is time to explicitly draw on Indigenous definitions of health, which directly address the need to connect individual and community health to the health of their country, in order to develop effective climate adaptation and health strategies. We detail how current health policies overlook this ‘missing’ dimension of Indigenous connection to country, and why that is likely to be detrimental to the health and well-being of people living in remote communities in a climate-changed future.  相似文献

This study correlated assessment tools for evaluating the severity of skin, nail, and joint symptoms in patients with psoriasis (Pso) and psoriatic arthritis (PsA). Adults with plaque Pso (with or without PsA) were enrolled from four U.S. institutions. Patients were evaluated using a novel 10-area Linear Psoriasis Area and Severity Index (XL-PASI), Psoriatic Arthritis Assessment (PsAA), Psoriatic Arthritis Screening and Evaluation Questionnaire (PASE), Nail Assessment (NA) and Joint Assessment (JA) tools, Psoriasis Weighted Extent and Severity Index (PWESI), and Lattice Physician Global Assessment (LS-PGA). Correlations between assessment tools and individual items in the assessment tools were performed. Data from 180 patients (55 with PsA) were analyzed. Highest correlations between tools (r = 0.77–0.88) were between the XL-PASI, PWESI and LS-PGA. Individual items in the XL-PASI correlated with items in the PWESI for extent skin symptoms, but not for all body areas. Overall, correlations were seen between hands and feet, between face and scalp, and between buttocks, chest, and back. Only low correlation was seen between items assessing joint symptoms with items assessing skin symptoms. These data support the notion that the complex phenotype of psoriatic disease requires instruments that assess the severity of skin, nails, and joints separately.  相似文献
