The receptor tyrosine kinases (RTKs) family is well-recognized as vital targets for the treatment of hepatocarcinoma cancer (HCC) clinically, whereas the survival benefit of target therapy sorafenib is not satisfactory for liver cancer patients due to metastasis. EGFR and MET are two molecules of the RTK family that were related to the survival time of liver cancer patients and resistance to targeted therapy in clinical reports. However, the mechanism and clinical therapeutic value of EGFR/MET in HCC metastasis are still not completely clarified. The study confirmed that EGFR/MET was highly expressed in HCC cells and tissues and the phosphorylation was stable after metastasis. The expression of EGFR/MET was up-regulated in circulating tumor microemboli (CTM) to accelerate IL-8 production and resistance to the lethal effect of leukocytes. Meanwhile, highly expressed EGFR/MET effectively regulated the Ras/MAPK pathway and stabilized suspended HCC cells by facilitating proliferation and inhibiting apoptosis. Moreover, EGFR/MET promoted phosphorylation of hetero-RTKs, which was dependent on high-energy phosphoric acid compounds rather than their direct interactions. In conclusion, highly expressed EGFR/MET could be used in CTM identification and suitable for preventing metastasis of HCC in clinical practice.Subject terms: Liver cancer, Metastasis  相似文献

The study of genomic structural evolution associated with accelerated evolutionary rates that result in avoidance of meltdown and increase biodiversity is becoming ever more possible as the number of available plastomes increases. To more comprehensively analyze rate heterogeneity among monocots and within Poaceae, we sequenced plastomes from four Poaceae species, combined them with publicly available data from ~200 plastomes, and conducted comparative analyses to quantify the pattern of rate heterogeneity between different lineages, functional groups, and periods of evolutionary time. We compared structural differences across the Poaceae to quantify how changes in plastome size correspond to different genomic subunits and the evolution of IR–SC junction boundaries. The substitution rates among ancestral Poaceae were inferred to be exceptionally rapid compared to other monocots but slowed after divergence into extant lineages, which could not be sufficiently explained by positive selection. As such, rapid rates in the ancestral lineage leading to Poaceae might be more closely linked to large-scale structural changes like the loss of ycf1 and ycf2. The total increase in plastome size across Poaceae was positively correlated with the total length of intergenic spacers, tandem repeats, and dispersed repeats as well as large single copy, and inverted repeats (IRs). The continuous evolution of IR–SC junction boundaries was asynchronous with sizes of total genome and subunits across Poaceae. Future work is needed to better understand what factors in ancestral Poaceae evolved to harness such rapid rates of plastome evolution, avoid a mutational meltdown, and escape the stagnation of strong purifying selection as well as if these factors could be utilized to synthetically control rates.  相似文献

在克隆了马铃薯X病毒(PVX)、马铃薯Y 病毒(PVY)和马铃薯卷叶病毒(PLRV)的外壳蛋白基因的基础上,构建同时包含PVX和PVY 与PVY 和PLRV 两个外壳蛋白基因植物表达框架的表达载体,通过农杆菌(Agrobacterium tumefaciens)介导转化烟草(Nicotianatabacum )和生产上常用的几个马铃薯(Solanum tuberosum )优良品种:“Favorita”、“虎头”、“克4”。经PCR检测证明外源基因已整合到植物的染色体上,得到批量转基因植株。在转PVX+PVY 外壳蛋白基因的烟草上接种PVX (5 μg/m L)、PVY(20 μg/m L)病毒,得到有一定抗性的植株  相似文献

The physical locations of the 5S and 45S rDNA sequences were examined in three types of teosinte, Zea mays ssp. mexicana (2n = 20), Zea diploperennis (2n = 20) and Zea perennis (2n = 40) by biotinylated fluorescence in situ hybridization (FISH). The tested materials only showed one hybridization site of 5S rDNA on their genomes, but they were different in the position of the signals. The hybridization site of Zea mays ssp. mexicana was located on the long arm of chromosome 2, indicating that it is the same as the cultivated maize in the position of 5S rDNA, while the sites of Zea diploperennis and Zea perennis were on the short arms of other chromosomes. For 45S rDNA, one hybridization site was detected at secondary constriction region of the satellite chromosomes in Zea mays ssp. mexicana and Zea diploperennis, while in Zea perennis, besides the site located at the secondary constriction region, a second site on the short arm of another chromosome pair was observed. Our results provide additional evidence for Zea mays ssp. mexicana being a subspecies of Zea mays.  相似文献

本文对地处中亚热带安徽黟县的次生灌丛和灌草丛以及常绿阔叶幼林的地上部分生产力进行了测定。结果表明:黟县常绿阔叶幼林、次生灌丛和灌草丛的地上部分净生产力分别为11.94,6.84和3.80(t/ha·a),次生灌丛和灌草丛的生产力较低,对亚热带优越的水热资源利用不充分。所调查的九个样地的地上部分生物量和净生产力可回归成线性方程ŷ=4.27+0.15x,净生产力随着生物量的增加而增加,即群落所处的演替阶段越高,净生产力越大,所以利用处于较高演替阶段的植被,会得到更多的收获。  相似文献

The Ziwuling black goat is an indigenously in China, their offspring are frequently affected by congenital cryptorchidism. The extracellular matrix (ECM) contains cytokines and growth factors that regulate the development of the testis, and component changes often result in pathological changes. Cryptorchidism is closely related to structural changes in ECM. In this study, the histochemical staining, immunohistochemical, immunofluorescence and Western blot combined with semi-quantitative analysis was used to describe the distribution of the important ECM components Collagen type IV (Col IV), laminin (LN)and heparan sulfate proteoglycans (HSPG) in the normal and cryptorchid testes of Ziwuling black goats. Results showed that: The histochemical staining showed that the dysplasia of seminiferous tubules and decreased number of Sertoli cells in cryptorchidism, as well as sparse collagen fiber. Meanwhile, the distribution of reticular fibers is relatively rich. Furthermore, the PAS and AB staining in the interstitial vessels and lamina propria of seminiferous tubules is weak. The immunohistochemical and immunofluorescence revealed that Col IV, LN was strongly expressed in Leydig, Sertoli cells of normal testes and moderately positive in the spermatogonia and spermatids, but HSPG was not expressed in the spermatogonia. However, cryptorchidism, the expression of Col IV, LN and HPSG in Leydig, Sertoli cells significantly decreased, as well as the expression of Col IV and LN in capillary endothelial cells, but HSPG was moderately expressed in spermatogonia. Based on these data, the underdevelopment of spermatogenic epithelium, decreased synthesis function of collagen fibers and Leydig cells develop usually in the cryptorchidism were shown to be closely related to the abnormal metabolism of Col IV and LN. The positive expressed of HSPG in the spermatogonia of cryptorchid testes is related to the compensatory development of spermatogonia.  相似文献

利用常规石蜡制片法,对生长于北戴河海滨沙地的9种沙生植物的营养器官的形态结构进行比较观察。结果表明:(1)叶结构出现各种旱生和盐生植物特征,如一些植物叶的栅栏组织发达,海绵组织退化;多数植物的叶片较小;多数植物支脉和侧脉多等;(2)轴器官具有发达的表皮和机械组织,有异常结构的出现。说明北戴河海滨沙滩生长的9种植物的营养器官存在一系列的旱生结构特征,这些结构在减少蒸腾作用,增强光合作用,储存水分,增强器官的机械强度方面具有积极的作用。  相似文献

长链非编码RNA(long non-coding RNA,IncDNA)是指长度超过200个核苷酸、具有调控基因表达作用的非编码RNA。近年来研究表明,长链非编码RNA在肿瘤的发生、发展过程中发挥着促癌或抑癌作用,它们参与了细胞凋亡调控、肿瘤浸润与转移等过程;另外,它们还通过表观遗传调控的方式影响肿瘤细胞的生长。它们有希望成为新型肿瘤标志物和肿瘤治疗的靶点,在肿瘤诊断和治疗方面显示出良好的临床应用前号。  相似文献
