Apoplastic Sugars, Fructans, Fructan Exohydrolase, and Invertase
in Winter Oat: Responses to Second-Phase Cold Hardening 总被引:8,自引:1,他引:8
Changes in apoplastic carbohydrate concentrations and activities of carbohydrate-degrading enzymes were determined in crown tissues of oat (Avena sativa L., cv Wintok) during cold hardening. During second-phase hardening (−3°C for 3 d) levels of fructan, sucrose, glucose, and fructose in the apoplast increased significantly above that in nonhardened and first-phase-hardened plants. The extent of the increase in apoplastic fructan during second-phase hardening varied with the degree of fructan polymerization (DP) (e.g. DP3 and DP4 increased to a greater extent than DP7 and DP > 7). Activities of invertase and fructan exohydrolase in the crown apoplast increased approximately 4-fold over nonhardened and first-phase-hardened plants. Apoplastic fluid extracted from nonhardened, first-phase-hardened, and second-phase-hardened crown tissues had low levels, of symplastic contamination, as determined by malate dehydrogenase activity. The significance of these results in relation to increases in freezing tolerance from second-phase hardening is discussed. 相似文献

We have shown the immunogenicity and safety of synthetic carbohydrate vaccines when conjugated to the carrier keyhole limpet
hemocyanin (KLH) and given with the adjuvant, QS-21, in patients with biochemically relapsed prostate cancer. To determine
whether immune response could be further enhanced with stimulation by multiple antigens, a hexavalent vaccine was prepared
using previously determined doses and administered in a Phase II setting to 30 high-risk patients. The hexavalent vaccine
included GM2, Globo H, Lewisy, glycosylated MUC-1-32mer and Tn and TF in a clustered formation, conjugated to KLH and mixed with QS-21. Eight vaccinations
were administered over 13 months. All 30 patients had significant elevations in antibody titers to at least two of the six
antigens; 22 patients had increased reactivity with FACS. These serologic responses were lower than that seen previously in
patients treated with the respective monovalent vaccines. The reciprocal median combined IgM and IgG antibody titers with
ELISA against MUC1, Tn, TF, globo H and GM2 for these 30 patients were 640, 80, 120, 40 and 0, compared to 1280, 640, 1280,
320 and 160 seen in patients receiving individual monovalent vaccines. This hexavalent vaccine of synthetic “self” antigens
broke immunologic tolerance against two or more antigens in all 30 vaccinated patients, was safe, but antibody titers against
several of the antigens were lower than those seen in individual monovalent trials. No impact on PSA slope was detected. We
address the relevance of the multivalent approach for prostate cancer treatment.

Modulation of cAMP levels has been linked to insulin secretion in preclinical animal models and in humans. The high expression of PDE-10A in pancreatic islets suggested that inhibition of this enzyme may provide the necessary modulation to elicit increased insulin secretion. Using an HTS approach, we have identified quinoline-based PDE-10A inhibitors as insulin secretagogues in vitro. Optimized compounds were evaluated in vivo where improvements in glucose tolerance and increases in insulin secretion were measured. 相似文献

We study the effect of micro-scale electric fields on voltage-gated ion channels in mammalian cell membranes. Such micro- and nano-scale electric fields mimic the effects of multiferroic nanoparticles that were recently proposed [1] as a novel way of controlling the function of voltage-sensing biomolecules such as ion channels. This article describes experimental procedures and initial results that reveal the effect of the electric field, in close proximity of cells, on the ion transport through voltage-gated ion channels. We present two configurations of the whole-cell patch-clamping apparatus that were used to detect the effect of external stimulation on ionic currents and discuss preliminary results that indicate modulation of the ionic currents consistent with the applied stimulus. 相似文献

Several commercially available pharmaceutical compounds have been shown to block the IKr current of the cardiac action potential. This effect can cause a prolongation of the electrocardiogram QT interval and a delay in ventricular repolarization. The Food and Drug Administration recommends that all new potential drug candidates be assessed for IKr block to avoid a potentially lethal cardiac arrhythmia known as torsades de pointes. Direct compound interaction with the human ether-a-go-go- related gene (hERG) product, a delayed rectifier potassium channel, has been identified as a molecular mechanism of IKr block. One strategy to identify compounds with hERG liability is to monitor hERG current inhibition using electrophysiology techniques. The authors describe the Ion Works HT instrument as a tool for screening cell lines expressing hERG channels. Based on current amplitude and stability criteria, a cell line was selected and used to perform a 300-compound screen. The screen was able to identify compounds with hERG activity within projects that spanned different therapeutic areas. The cell line selection and optimization, as well as the screening abilities of the Ion Works HT system, provide a powerful means of assessing hERGactive compounds early in the drug discovery pipeline. 相似文献

ABSTRACT. Previous studies using thermal imaging cameras (TI) have used target size as an indicator of target altitude when radar was not available, but this approach may lead to errors if birds that differ greatly in size are actually flying at the same altitude. To overcome this potential difficulty and obtain more accurate measures of the flight altitudes and numbers of individual migrants, we have developed a technique that combines a vertically pointed stationary radar beam and a vertically pointed thermal imaging camera (VERTRAD/TI). The TI provides accurate counts of the birds passing through a fixed, circular sampling area in the TI display, and the radar provides accurate data on their flight altitudes. We analyzed samples of VERTRAD/TI video data collected during nocturnal fall migration in 2000 and 2003 and during the arrival of spring trans-Gulf migration during the daytime in 2003. We used a video peak store (VPS) to make time exposures of target tracks in the video record of the TI and developed criteria to distinguish birds, foraging bats, and insects based on characteristics of the tracks in the VPS images and the altitude of the targets. The TI worked equally well during daytime and nighttime observations and best when skies were clear, because thermal radiance from cloud heat often obscured targets. The VERTRAD/TI system, though costly, is a valuable tool for measuring accurate bird migration traffic rates (the number of birds crossing 1609.34 m [1 statute mile] of front per hour) for different altitudinal strata above 25 m. The technique can be used to estimate the potential risk of migrating birds colliding with man-made obstacles of various heights (e.g., communication and broadcast towers and wind turbines)—a subject of increasing importance to conservation biologists. 相似文献

Herpes simplex virus type 1 (HSV-1) DNA replication occurs in replication compartments that form in the nucleus by an ordered process involving a series of protein scaffold intermediates. Following entry of viral genomes into the nucleus, nucleoprotein complexes containing ICP4 can be detected at a position adjacent to nuclear domain 10 (ND10)-like bodies. ND10s are then disrupted by the viral E3 ubiquitin ligase ICP0. We have previously reported that after the dissociation of ND10-like bodies, ICP8 could be observed in a diffuse staining pattern; however, using more sensitive staining methods, we now report that in addition to diffuse staining, ICP8 can be detected in tiny foci adjacent to ICP4 foci. ICP8 microfoci contain UL9 and components of the helicase-primase complex. HSV infection also results in the reorganization of the heat shock cognate protein 70 (Hsc70) and the 20S proteasome into virus-induced chaperone-enriched (VICE) domains. In this report we show that VICE domains are distinct but adjacent to the ICP4 nucleoprotein complexes and the ICP8 microfoci. In cells infected with an ICP4 mutant virus encoding a mutant protein that cannot oligomerize on DNA, ICP8 microfoci are not detected; however, VICE domains could still be formed. These results suggest that oligomerization of ICP4 on viral DNA may be essential for the formation of ICP8 microfoci but not for the reorganization of host cell chaperones into VICE domains. 相似文献
