Targeting of many transmembrane proteins to post-Golgi compartments is dependent on cytoplasmically exposed sorting signals. The most widely used signals conform to the tyrosine- or the leucine-based motifs. Both types of signals have been implicated in protein localization to the same intracellular compartments, but previous results from both cell-free experiments and studies of transfected cell lines have indicated that the two types of signals interact with separate components of the sorting machinery. We have overexpressed several transmembrane proteins in stably transfected Madin-Darby canine kidney cells using an inducible promoter system. Overexpression of proteins containing tyrosine- or leucine-based sorting signals resulted in reduced internalization of the transferrin receptor, whereas recycling and polarized distribution was not influenced. Our results indicate that proteins with tyrosine- and leucine-based sorting signals can be transported along common saturable pathways.  相似文献

In order to study the occurrence of mycoplasmas among Norwegian sheep, lungs from a great number of different herds were collected at 3 abattoirs in Southern Norway. The presence of fermenting mycoplasmas and bacteria was examined in both normal and pneumonic lungs to determine whether recovery of these agents could be related to pneumonic changes. Pneumonic lungs demonstrated lesions typical of the condition described as subacute or chronic pneumonia. Mycoplasma ovipneumoniae was found in 87 % of the 126 pneumonic lungs and in 6 % of the 83 normal lungs. Bacteria, mostly Pasteurella haemolytica, were less frequently encountered in the pneumonic lungs, and usually in combination with M. ovipneumoniae. It is concluded that M. ovipneumoniae is widespread among sheep in Southern Norway and can be considered to have etiological significance in subacute or chronic pneumonia, whereas bacteria probably occur mainly as secondary invaders. Changes resulting from moderate invasion by lungworm were found in about half of the lungs, but just as frequently in normal as in pneumonic lungs, and accordingly did not appear to contribute to the pneumonia investigated.  相似文献

NMR was used to monitor the binding to neurophysin of oxytocin and 8-arginine-vasopressin, 15N labeling being used to identify specific backbone 15N and 1H signals. The most significant effects of binding were large downfield shifts in the amino nitrogen resonance of Phe-3 of vasopressin and in its associated proton, providing evidence that the peptide bond between residues 2 and 3 of the hormones is hydrogen-bonded to the protein within hormone-neurophysin complexes. Suggestive evidence of hydrogen bonding of the amino nitrogen of Tyr-2 was also obtained in the form of decreased proton exchange rates on binding; however, the chemical shift changes of this nitrogen and its associated proton indicated that such hydrogen bonding, if present, is probably weak. Shifts in the amino nitrogen of Asn-5 and in the -NH protons of both Asn-5 and Cys-6 demonstrated that these residues are significantly perturbed by binding, suggesting conformational changes of the ring on binding and/or the presence of binding sites on the hormone outside the 1-3 region. No support was obtained for the thesis that there is a significant second binding site for vasopressin on each neurophysin chain. The behavior of both oxytocin and vasopressin on binding was consistent with formation of 1:1 complexes in slow exchange with the free state under most pH conditions. At low pH there was evidence of an increased exchange rate. Additionally, broadening of 15N resonances in the bound state at low pH occurred without a corresponding change in the resonances of equilibrating free hormone.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Corticotropin releasing factor (CRF) or saline was administered i.p. to rats aged either 100 or 220 days, followed by either brief handling or water immersion restraint. Plasma corticosterone was measured 75 min. later. Age of the animals in itself was not a significant factor either for basal levels of plasma corticosterone or for extent of restraint induced gastric pathology. However after CRF administration, young but not older animals revealed a significant increase in plasma corticosterone levels, and post restraint gastric ulcerations were more severe in older than young animals. CRF significantly decreased the number of restraint induced ulcers in young rats, while the cumulative ulcer length was increased in older animals.  相似文献

The relationship between ATP and energy charge was studied in individuals of Cirolana borealis under heavy metabolic stress caused by anoxia or exposure to toluene. Prolonged anoxia led to a lowering of the ATP content to about 10% after 4 days, with a simultaneous decrease in energy charge to about 0.25. A lowering of the total adenylate pool reduced the fall in energy charge somewhat, but this effect was marked only in late anoxia when the individuals had become inactive. Exposure to 0.14 mM toluene for 8 days led to a similar decrease in ATP and energy charge. Exposure to 1.4 mM toluene for 24 h led to only slight changes in the adenine nucleotide pool, although the individuals became narcotized within a few hours. The energy charge associated with moribund individuals thus varied much. The mechanism of energy charge stabilization through reduction of the adenine nucleotide pool as ATP declined seemed to be of little significance for the survival of the individuals.  相似文献

As part of a project aimed at assessing the available energy in a pelagic system and the efficiency of its transfer, monthly biomass estimates of the 23 main macroplanktonic and mesoplanktonic species have been made in Korsfjorden (Norway). Sampling was carried out over approximately 3 years (1971–1974) and results are presented as mg dry weight in a 690 m3 water column. The total annual stocks of the various species were not constant in the 3 years of sampling. Principal components analyses of the monthly stocks in each of the 3 years show striking differences in the correlation matrices from year to year. On the basis of known and supposed feeding habits, the species have been classified as herbivores, omnivores, or carnivores. The monthly biomass estimates of these groups show a consistent pattern from year to year. This, in contrast to the variable correlations between individual species, indicates that there can be considerable flexibility in the specific composition of a planktonic community without correspondingly marked effects on its trophic structure.  相似文献

It has been reported that the human cell line NHIK 3025 has a specific cytoplasmic glucocorticoid receptor. When these cells were exposed to glucocorticoids, the cell cycle time was prolonged. Cells, synchronized by mitotic selection, were subjected to the synthetic glucocorticoid dexamethasone throughout the cell cycle. Only cells exposed in the first half of G1 phase had a lengthened cell cycle time. Most of the prolongation was also located within the G1 phase. The dexamethasone growth inhibition was reversible and could be detected only in the cell cycle where the cells were exposed to the steroid. DNA-histograms of asynchronous cells were recorded by flowcytometry at various times after steroid exposure. These histograms also showed G1 phase sensitivity and G1 phase prolongation after exposure to dexamethasone. Our results thus indicate that these cells have a dexamethasone-sensitive restriction point in mid-G1 phase of the cell cycle.  相似文献

The site of substitution of the peripheral branches on the core mannose-residues in the complex-type carbohydrate units of glycoproteins has been studied by Smith degradation. By using glycopeptides of known structure, it is shown that the site of attachment of the (1→4)-linked, third branch to the mannose core in tri- and tetra-antennary structures may be conveniently demonstrated by this technique. A critical factor was found to be the concentration of acid used for cleavage of the periodate-oxidized product. This finding could explain discrepancies concerning the previously published structures of fetuin glycopeptides. The technique was applied for study of the branching pattern in human transferrin, porcine thyroglobulin, and rat plasma and brain glycoproteins. The results support the concept that, in glycoproteins from most sources, the third (1→4)-linked branch is attached to the (1→3)-linked mannose residue of the core portion in the complex-type structures. An indication for a novel type of branching pattern in human transferrin was, however, also obtained.  相似文献

Functioning quantum dot (QD) sensitized solar cells have been fabricated using the vacuum deposition technique atomic layer deposition (ALD). Utilizing the incubation period of CdS growth by ALD on TiO2, we are able to grow QDs of adjustable size which act as sensitizers for solid‐state QD‐sensitized solar cells (ssQDSSC). The size of QDs, studied with transmission electron microscopy (TEM), varied with the number of ALD cycles from 1‐10 nm. Photovoltaic devices with the QDs were fabricated and characterized using a ssQDSSC device architecture with 2,2',7,7'‐tetrakis‐(N,N‐di‐p methoxyphenylamine) 9,9'‐spirobifluorene (spiro‐OMeTAD) as the solid‐state hole conductor. The ALD approach described here can be applied to fabrication of quantum‐confined structures for a variety of applications, including solar electricity and solar fuels. Because ALD provides the ability to deposit many materials in very high aspect ratio substrates, this work introduces a strategy by which material and optical properties of QD sensitizers may be adjusted not only by the size of the particles but also in the future by the composition.  相似文献

Base excision repair (BER) is initiated by a DNA glycosylase and is completed by alternative routes, one of which requires proliferating cell nuclear antigen (PCNA) and other proteins also involved in DNA replication. We report that the major nuclear uracil-DNA glycosylase (UNG2) increases in S phase, during which it co-localizes with incorporated BrdUrd in replication foci. Uracil is rapidly removed from replicatively incorporated dUMP residues in isolated nuclei. Neutralizing antibodies to UNG2 inhibit this removal, indicating that UNG2 is the major uracil-DNA glycosylase responsible. PCNA and replication protein A (RPA) co-localize with UNG2 in replication foci, and a direct molecular interaction of UNG2 with PCNA (one binding site) and RPA (two binding sites) was demonstrated using two-hybrid assays, a peptide SPOT assay and enzyme-linked immunosorbent assays. These results demonstrate rapid post-replicative removal of incorporated uracil by UNG2 and indicate the formation of a BER complex that contains UNG2, RPA and PCNA close to the replication fork.  相似文献
